This study was conducted to investigate the effects of two fermentation methods using Hericium erinaceum Mycelium (HE) and second fermentation using Leuconostoc mesenteroides by using snail meat of Fruticiola sieboldiana species, The anti-aging activity of the snail extract was evaluated. Amino acid content of the snail extract before fermentation was 19 species. The total amino acid content was 15.8wt%. The total amino acid content of the snail extract analyzed by the second fermentation process was 76.7 wt%. Fatty acids were 3.7 wt%. The polysaccharide content was 16.7wt%. The moisture content was 2.9 wt%. Other ingredients were 4.2 wt%. Through the fermentation, 19 kinds of amino acids were unchanged. The total amino acid content increased by 136% and 285% to 37.4 wt% and 76.7 wt%, respectively, after the 1st and 2nd fermentation.In particular, the contents of four amino acids of serine, glycine, arginine, and proline were significantly increased. The DPPH free Radical scavenging activity (IC50%) of the extract was 7.3 mg/mL, which was twice as good as that of the snail extract before fermentation. The inhibitory effect of elastin inhibitor (IC50%) on the secondary fermented snail extract was 29.2% higher than that of the control group. And it was more than 41.2% more than the ordinary extract. The inhibitory activity of tyrosinase (IC50%) of the 2nd fermented snail extract was 44.0% higher than that of the control group. It was more than 177.6% more than the ordinary extract. The growth rate of fibroblast of the second fermented snail extract was 27% higher than that of the control group. And it was more than 16% higher than the ordinary extract. The collagen biosynthesis rate of the second fermented snail extract was 31% higher than that of the control group. And it was more than 24.8% higher than the general extract. As a clinical study, a sheet mask pack containing 5% of a snail fermented extract was prepared to confirm skin improvement effect of the second fermented snail extract. The application method was applied to the skin three times a week for 4 weeks for woman in her thirties and the moisturizing effect, the elasticity improvement, the roughness, the melanin pigment and the wrinkle improvement were studied. As a result, the melanin-reducing effect of the 2F-SEM was decreased by 15.0% or more than that of PM. Wrinkle improvement was improved by 5.1%. Pigmentation was decreased by 15.0% and melanin reduction effect was shown. Sensory evaluation of mask pack showed excellent difference in softness, moisture feel, and fine wrinkle improvement. Secondary fermented snail extracts obtained through this study are expected to be used as raw materials for cosmetics with antioxidant effect.
