바실러스 서브틸리스 및 웨이 셀라 김치 박테리아 고스트는 염산, 황산, 질산, 아세트산, 시트르산, 말레산, 수산화 나트륨, 수산화 칼륨, 탄산나트륨, n- 부탄올, 2- 프로판올, 에탄올, 메탄올의 최소억제농도에 의해 제조되었다. 그 중 염산, 황산, 질산 수산화나트륨을 60 분간 처리했을 때 바실러스 서브틸리스에서 생존 가능한 콜로니가 발견되지 않았고, n-부탄올, 2-프로판올, 염산, 아세트산, 수산화칼륨, 탄산나트륨, 황산, 질산, 수산화나트륨을 60 분 동안 처리했을 때 웨이 셀라 김치균의 생존가능한 콜로니는 검출되지 않았다. 염산을 처리한 바실러스 서브 틸리스 와 수산화 나트륨을 처리한 웨이셀라 김치로 만들어진 박테리아 고스트만이 완전하게 DNA가없고 효율적이었다 (실시간 PCR에 의해 확인 됨). 주사 전자 현미경으로 산과 알칼리에 의해 유도된 박테리아고스트 외벽에 막 관통 터널이 형성되었음을 확인하였다. 또한, 산 과 알칼리에 유도된 박테리아 고스트의 단백질 프로파일에 상당한 차이가 있었다. 독성 실험을 통해 박테리아 고스트를 설치류 대식세포에서의 처리한 후, 생존율을 확인 하였다. 박테리아 고스트를 처리한 대식세포에서 염증성 사이토카인 3종 (IL-1β, IL-6, TNF-α) 발현을 조사분석함으로써, 박테리아 고스트에 의한 대식세포의 면역반응을 관찰하였다. IL-1β, IL-6, TNF-α 모두 박테리아 고스트를 처리하였을 때 활성이 증가하였으며 특히 염산에 의해 제조된 바실러스 서브틸리스 박테리아 고스트에서 큰 활성을 보였다. 신경줄기세포에서 BG를 처리 한 후 분화를 유도하여 신경 분화에 대한 효과를 확인 하였다. 신경줄기세포의 분화를 유도하였을 때 (-bFGF) 신경분화마커인 Tuj1과 신경영양인자(NT-3, NT4/5)의 발현이 증가하였고, LPS를 처리하면 신경독소로 인하여 신경세포의 분화가 억제되었다. 일반적인 조건 (-bFGF) 에서 신경 분화를 시켰을 때 보다 염산에 의해 제조된 바실러스 서브틸리스 박테리아 고스트와 수산화 나트륨에 의해 제조된 웨이셀라 김치 박테리아 고스트를 처리하여 분화 시켰을 때 신경분화가 현저히 증가함을 확인할 수 있었고 시냅스가 더 많이 생성됨을 알 수 있었다. 기존의 생균과 BG의 차이를 확인하기 위해 2DE 영상 분석과 MALDI-TOF 분석을 수행 하였다. 기존의 생균에서 없던 단백질이 박테리아 고스트상에서 발견되었다. 위의 모든 결과 DNA가 존재하지 않는 박테리아 고스트는 생균과 유사한 면역능이 유지되면서도 또한 새로운 단백질들을 갖게 되어 보다 특이적 신규성 면역 강화제 개발에 효율적임을 제시한다.

Chemicals used to produce BG consist of groups of acids, alkalis and alcohols including hydrochloric acid (HCl), sulfuric acid (H2SO4), nitric acid (HNO3), acetic acid (CH3COOH), citric acid (C6H8O7), maleic acid (C4H4O4), sodium hydroxide (NaOH) , potassium hydroxide (KOH), sodium carbonate (Na2CO3), n–butanol, 2–propanol, ethanol and methanol. Bacillus subtilis cells that were treated for 60 minutes with some kinds of chemicals (HCl, H2SO4, HNO3 and NaOH) accoding to MICs did not show any colony after plating on the LB agar plates for the detection of the viability. In case of Weissella kimchii, MICs of several chemicals (n–Butanol, 2–Propanol, HCl, CH3COOH, KOH, Na2CO3, H2SO4, HNO3 and NaOH) made no colony after plating on the MRS agar medium in order to analyze the viability. Of BGs produced by chemical treatments, only BGs of HCl-induced Bacillus subtilis and NaOH-induced Weissella kimchii were completely nonliving and DNA-free (confirmed by real-time qPCR). A scanning electron microscope (SEM) showed that both acid and alkali induced BGs formed a membrane-membrane lytic tunnel structure in the cell envelope. However, Bacillus subtilis induced by NaOH showed severe damaged. In addition, there were significant differences in protein profiles on SDS-PAGE gels between acid and alkali induced BGs. For the analysis of cytotoxicity in marcrophages cells exposed to BGs, bacterial ghosts (BGs) were treated to murine macrophages and their viabilities were determined by using Cell Counting Kit–8 (CCK-8 , Sigma-Aldrich, St. Louis, Mo. USA). Compared the cytokine expressions of the macrophage cell (control) with those of macrophage cells exposed to WKimgGs (Weissella kimchii ghosts) and to BSGs (Bacillus subtilis ghosts), most of macrophages exposed to BGs (WkimGs, BSGs) induced IL-1β,IL-6 and TNF-α higher than control macrophages non-exposed to BGs. In the neural stem cells exposed to BGs, differentiation was strongly induced by means of the high expression of neurotrophic factors (NT3, NT4/5) and neuronal marker gene (Tuj1). 2DE image analysis and PMF (Peptide Mass Fingerprinting)/MALDI-TOF analysis were performed to confirm the differences of the protein profiles between intact live bacteria and BGs.
