In this study, we compared black ginseng to red ginseng (RG) that have been steamed 5 times (BG). The ginsenoside contents of each extract were confirmed using high-performance liquid chromatography (HPLC) and it has shown that the ginsenoside components has been altered through increased processing. The anti-inflammatory and anti-nociceptive effects of BG are not widely studied. In this study, lipopolysaccharide (LPS)-stimulated RAW 264.7 murine macrophage cells were used and treated with ethanol extracts of RG and BG for in vitro studies. We found that, the highest concentration of these extracts has inhibited NO production to near-basal levels without exhibiting cytotoxicity. Pro-inflammatory cytokine expressions at the mRNA level were investigated using qRT-PCR and Real-Time PCR. Both extracts down-regulated the expression of pro-inflammatory mediators i.e. iNOS and COX-2 and pro-inflammatory cytokines i.e. IL-1β, IL-6 and TNF-α. Comparatively, BG exhibited better inhibition of pro-inflammatory mediators and cytokines. Protein expression was determined using western blot analysis. The ginseng samples inhibited LPS induced inflammation through the NF- κB and MAPK pathway. BG exhibited stronger inhibition for comparatively. To further confirm the anti-inflammatory and possible anti-nociceptive activities of RG and BG, xylene-induced ear edema model in mice and carrageenan-induced paw edema in rats were carried out and tested with the samples along with dexamethasone and indomethacin, which is widely, used non-steroidal anti-inflammatory drugs (NSAIDs). The results from both animal edema models further confirms that BG is more potent ginseng extract as an anti-inflammatory agent, possesses anti-nociceptive properties and has a stronger potency compared to the NSAIDs. Hence, we compared the anti-inflammatory abilities of RG and BG for the first time and confirmed that BG is the extract with the best potency. Future experimentation will unravel more biologically beneficial abilities for BG extract
