Treatments for H. pylori infection have mainly depended on the standard triple therapy which is consist of antibiotics and proton pump inhibitors. However, conventional antibiotic therapy has limitations in H. pylori eradication because of an increasing failure rate caused by the development of resistant strains and the low compliance of patients. It is therefore necessary to develop other eradication methods. To solve these problems, we have demonstrated that anti-bacterial activity and anti-inflammatory activity of Angelica dahurica against H. pylori. Anti-bacterial activity, inhibition of cagA gene expression, urease inhibition and anti-adhesion activity against H. pylori to AGS cells were investigated. Inhibition of IL-8 mRNA expression and IL-8 production of Angelica dahuricae were also investigated in order to examine the inhibition of inflammation induced by H. pylori infection in AGS cells. The MICs and MBCs of MeOH extracts of Angelica dahurica (MeOHEx), byakangelicol (BYAK) and scopoletin (SCOP) were 250 mg/mL, 100 mg/mL and 100 mg/mL, respectively. cagA gene expression and urease activity were significantly reduced in the presence of Angelica dahurica. Cell cytotoxicity were not observed at a concentration of 250 mg/mL of MeOHEx, 400 mg/mL of BYAK and 400 mg/mL of SCOP and these concentrations were higher than that of MBC of each component. Proinflammatory cytokine IL-8 mRNA expression was reduced in the presence of Angelica dahurica and production of IL-8 was also reduced in the presence of 12.5 mg/mL of BYAK (40%) and 50 mg/mL of SCOP (28%). Conclusively, Angelica dahurica showed potent anti-helicobacter and anti-inflammatory activity antibacterial activity and Angelica dahurica may be candidate for alternative medicinal plants for the treatment of H. pylori infection.
