Background: β-ocimene is a monoterpene compound which is a component of essential oils extracted from plants. It has been reported that essential oils, which contains high concentration of β-ocimene play important roles on skin wound healing, anti-inflammation, antioxidant and antibacterial activity. However, the effect of βocimene on hair growth and its molecular mechanism has not been identified.
Objective: We studied the hair growth promoting effect of β-ocimene and investigated the underlying molecular mechanism.
Methods:We investigated whether β-ocimene facilitates telogen-to-anagen induction in vivo and in organ culture using vibrissa hair follicle. We also examined the prolongation effect of β-ocimene on anagen stage in vivo. In addition, we studied the effect of β-ocimene on the proliferation and migration in dermal papilla cells (DP cells). We also investigated molecular mechanism of β-ocimene via ERK, AKT, Bcl2, and Bax pathway by western blot.
Results: Topical application of β-ocimene increased hair weight, matrix cell proliferation around hair follicles and the number of mature hair follicle. These results suggest β-ocimene accelerated telogen-to-anagen induction in vivo. In addition, β-ocimene prolonged anagen stage longer than control. Moreover, β-ocimene induced a significant hair shaft elongation and anagen stage prolongation of individual hair follicles in vibrissa organ culture. β-ocimene significantly increased the proliferation and migration of DP cells. The phosphorylation levels of ERK and AKT were significantly increased by β-ocimene. β-ocimene increased expression of Bcl-2 and decreased expression of Bax.
Conclusion: These results suggest that β-ocimene stimulates proliferative and migratory activity of DP cells by increasing phosphorylation of ERK and AKT. Moreover, β-ocimene inhibited apoptosis by regulation of Bcl-2 and Bax expression. Therefore, β-ocimene has a potential as a novel therapeutic reagent for hair loss.
