Background Sarcopenia is a progressive loss of muscle mass and function with aging, that associated with physical frailty and increased risk of morbidity. Ginseng and amino acid supplement have been proven to enhance muscle protein synthesis, improve physical training, protect against muscle damage. In this study, we aim to evaluate the effects of Korean red ginseng and amino acid supplements on dexamethasone induced myopathy for the prevention and treatment of sarcopenia.
Methods Sprague dawley rats were randomly divided into control(CON), dexamethasone(DEX), 3 intervention groups following dexamethasone induced myopathy; Korean red ginseng (DEX-KRG), amino acid supplements (DEX-AAS), Korean red ginseng + amino acid supplements (DEX-KRG+AAS). After 7days of Dexamethasone suppression (2mg/kg·day, intraperitoneally), Korean red ginseng (166.6mg/day, orally), amino acid supplements (750mg/day, orally) were administered for 2weeks in rats. The following parameters were evaluated: body weight, grip strength, muscle fiber cross sectional area, expression of myosin heavy chain isoforms in skeletal muscle (Tibialis anterior).
Results Body weight and grip strength were inhibited by the administration of dexamethasone, and although weight showed a significant increase independent of intervention after the recovery period, grip strength only showed a tendency to recover and had no statistical significance. The comparison of the fraction by the muscle fiber cross sectional area in DEX and intervention group(DEX-KRG. DEX-AAS, DEX-KRG+AAS) showed an increased area of Type IIb muscle fiber in DEX-KRG+AAS group. However, the relative size measurement of muscle fiber showed some differences between each group depending on the type of muscle fiber, but it was difficult to verify the effect of certain interventions. In the MyHC transition observed through western blot, the DEX-KRG group appears to increase Type Ⅱb and Type IIx muscle fibers after the recovery period, but had no statistical significance.
Conclusion These data suggest that the Korean red ginseng is likely to promote skeletal muscles recovery following dexamethasone-induced myopathy through the induction of slow-to-fast fiber type transition, further studies are required to evaluate Korean red ginseng as a supplementary agent for sarcopenia.
