L. plantarum 200655는 API ZYM kit를 이용하여 발암효소인 β-glucuronidase의 생산여부를 확인한 결과, 발암효소를 생산하지 않는 것으로 확인되었다. Lactic acid bacteria have been used to ferment a variety of foods, including dairy products, meat, coffee, and many plants. Probiotic properties including antioxidant and immune-enhancing effects of Lactobacillus plantarum 200655 isolated from kimchi were evaluated. The tolerance of three strains (L. plantarum 200655, L. plantarum KCTC 3108, and L. rhamnosus GG) to bile salts (0.3% oxgall, 24 h) was similar, and L. plantarum 200655 showed the highest tolerance to gastric juice (0.3% pepsin, 3 h). L. plantarum 200655 showed higher cell surface hydrophobicity and adhesion ability on HT-29 cells. L. plantarum 200655 did not produce β-glucuronidase and was sensitive to ampicillin, tetracycline, chloramphenicol, and doxycycline. Additionally, L. plantarum 200655 showed the highest antioxidant effects in DPPH and ABTS radical scavenging, and β-carotene bleaching assays. RAW 264.7 cells treated with L. plantarum 200655 produced more nitric oxide, induced nitric oxide synthase, and cytokine related to immune-enhancing effects such as interleukin-1β and interleukin-6. Therefore, L. plantarum 200655 could be useful as a probiotic strain for people. Probiotic properties and antioxidant activity of Pediococcus pentosaceus SC28 and Lactobacillus brevis KU15151 were evaluated. Gastric acid and bile salts tolerance of P. pentosaceus SC28 and L. brevis KU15151 were more than 90%, respectively. These LAB strains did not produce β-glucuronidase and were resistant to gentamicin, kanamycin, streptomycin, ciprofloxacin. L. brevis KU15151 showed higher adhesion to HT-29 cells and antioxidant activity in DPPH and ABTS radical scavenging, and β-carotene bleaching assay than P. pentosaceus SC28. Black gamju, a Korean traditional rice-based beverage, was fermented using P. pentosaceus SC28 and L. brevis KU15151, respectively. After fermentation, viable LAB cell number of fermented black gamju was increased (8.25-8.95 Log CFU/mL) and pH value of fermented black gamju was decreased (pH 3.33-3.41) compared non-fermented black gamju (pH 4.37). After extraction of non-fermented and fermented black gamju, ABTS radical scavenging and β-carotene bleaching assay were conducted. Fermented black gamju showed higher antioxidant activity than non-fermented black gamju. Specially, Fermented black gamju using L. brevis KU15151 had superior antioxidant activity. Therefore, P. pentosaceus SC28 and L. brevis KU15151 could be useful as probiotic strains and black gamju could be used as a probiotic drink having antioxidant effect.
