In this point, the purpose of this study was to evaluate the stability of encapsulated materials and to find applicability to food industry. Branched-chain amino acid (BCAA) as a functional substance was encapsulated with liposome prepared at pH 4 or pH 7 on the basis of isoelectric point of BCAA. All the samples were stored respectively at 4°C, 25°C and 40°C for 5 days and measured encapsulation efficiency, changes in particle size, zeta potential value and cumulative release rate. The particle size of liposomes in pH 7 was larger than pH 4, due to repulsive force between BCAA and hydrophilic part of liposomes. Zeta potential value of liposomes was higher in pH 4 than pH 7, so it is predicted that stability of storage would be higher in pH 4. Encapsulation efficiency was over 96% in both pH 4 and 7. Cumulative release rate was increased in 40°C storage condition, therefore it is concluded that these liposomes were sensitive to thermal changes. From these research about liposomes containing BCAA, it was tried to encapsulate probiotic bacteria with liposome. However, probiotic bacteria have mobility with flagella so they were not encapsulated stably in liposomes. Therefore, it is concluded that liposome is not suitable method for encapsulating probiotic bacteria and performed the other way, extrusion. So, this study was conducted to evaluate the protective effect of encapsulated probiotics Lactobacillus acidophilus in simulated gastrointestinal tract. L. acidophilus were homogeneously encapsulated by extrusion device in alginate or alginate/chitosan microspheres, which size was larger than alginate microspheres and all the size of microspheres was smaller than 100 μm. Encapsulation efficiency was more than 99% and the viability of L. acidophilus after encapsulation was more than 97%. The protective effect of microspheres was higher than control in simulated gastrointestinal tract. Also, mucoadhesive ability was improved in alginate and alginate/chitosan-microspheres than control cells. To prolong the cell’s storage period, control and encapsulated cells were freeze-dried with saccharides as lyoprotectants. As a result, sucrose showed the highest survival rate among all the samples. Also, scanning electronic microscopic image showed L. acidophilus was randomly embedded in the matrix of lyoprotectant. Therefore, these encapsulation process could protect L. acidophilus from gastrointestinal tract and improve mucoadhesive ability in intestine. Moreover, opitmal lyoprotectant could help survive L. acidophilus during freeze drying process.
