실험방법: 좌측 상악 절치 경부 결찰 24시간 후부터 CM-SD 200, 100 및 50 mg/kg을 멸균 증류수에 현탁시켜, 각각 5 ml/kg의 용량으로 매일 1회씩 10일간 강제 경구투여하고, 체중, 치주골 손실율 (alveolar bone lossindex), 상악 치은 조직내 prostaglandin E2 (PGE2), matrix metalloproteinase (MMP)-8, interleukin (IL)-1β 및 tumor necrosis factor (TNF)-α 함량, 지질 과산화 (malondialdehyde; MDA 함량), inducible nitric oxide synthase (iNOS) 및 myeloperoxidase (MPO) 활성, receptor activator of nuclear factor kappa-B ligand (RANKL) 및 osteoprotegerin (OPG) mRNA 발현의 변화를 조직병리학적 변화와 함께 관찰하였다. 본 실험에서 CM-SD의 약효는 NSAIDs 계열의 항염제인 IND 5 mg/kg 경구 투여군과 비교 평가하였다.
결과: 본 실험의 결과, 치아 경부 결찰 후 현저한 체중 및 증체량의 감소, 치주골 손실율, 치은 조직내 MPO 및 iNOS 활성, PGE2, MMP-8, IL-1β, TNF-α 및 MDA 함량, RANKL/OPG mRNA발현 비율의 현저한 증가가 인정되었으며, 조직병리학적 검사에서 치은 조직내 염증세포의 침윤, 콜라겐 섬유가 차지하는 비율의 감소, 조골세포 수 및 활성 감소, 이와 관련된 치주골량의 감소; 즉, 전형적인 치주염 및 이와 관련된 치주골 소실이 유발되었다. 한편 이러한 치아 경부 결찰로 유발된 치은염 및 관련 치주골 소실이 모든 3 용량의 CM-SD 200, 100 및 50 mg/kg의 10일에 걸친 연속 투여에 의해 투여 용량 의존적으로 현저히 억제되었고, 특히 적어도 본 실험의 조건하에서 CM-SD 200 mg/kg은 IND 5 mg/kg과 비교할만한 항염 효과를, CM-SD 50 mg/kg은 IND 5 mg/kg과 비교할만한 치주골 소실 억제 활성을 각각 나타내었다.
결론: 이상에서, CM-SD는 항염, 항산화, MMP-8 억제, RANKL/OPG mRNA 발현 조절을 매개하여, 200 mg/kg 경구 투여 용량에서 IND 5 mg/kg 경구 투여 용량과 비교할만한 항염 효과를 나타내는 것으로 관찰되었으며, 50 mg/kg 경구 투여 용량에서 ND 5 mg/kg과 비교할만한 치주골 소실 억제 활성을 나타내는 것으로 관찰되었다. 따라서 금후 보다 자세한 기전 연구 및 다양한 실험동물 모델에서의 약효 평가를 통해 새로운 보다 효과적인 치주 질환 개선 건강 식품 소제 등으로의 개발 가능성이 매우 높을 것으로 기대된다.

PURPOSE: The objective of present study is to observe the dose-dependent inhibitory potentials of Curcumin 34.9% Spray Dry Powders (CM-SD), a new curcumin formulation, improved the solubility and absorption by Aribio (Seungnam, Korea), were evaluated on EPD rats induced by ligation placement on the cervix of upper left incisior teeth, as a process to develop potent natural medicinal foods for preventing or treating periodontal diseases. In this experiment, indomethacin (IND), a non-selective cyclooxygenase inhibitor was selected as potent reference drug at 5 mg/kg oral administration.
MATERIALS AND METHODS: One day after ligation placements, three different dosages of CM-SD – 200, 100 and 50 mg/kg were orally administered, once a day for 10 days, in the current experiment. The changes on the body weight and gain during experimental periods and alveolar bone loss index were analyzed, and for detecting anti-inflammatory effects, myeloperoxidase (MPO) activities, prostaglandin E2 (PGE 2) contents, proinflammatory cytokine – interleukin (IL)-1β and tumor necrosis factor (TNF)-α contents were evaluated in gingival tissues with matrix metalloproteinase (MMP)-8 contents and histopathological changes on the gingival tissues around ligature placed incisior teeth. In addition, maxillary gingival tissue iNOS activities, lipid peroxidation, receptor activator of nuclear factor kappa-B ligand (RANKL) and osteoprotegerin (OPG) mRNA expressions were also monitored, in the current experiment. The results of mixtures were compared with those of IND, one of well documented non-steroid anti-inflammatory drugs (a non-selective cyclooxygenase inhibitor) 5 mg/kg treated rats.
RESULTS: As results of ligature placements on the cervix of upper left incisior teeth in rats, severe decreases of body weight and gains, increases of alveolar bone loss scores, gingival MPO and iNOS activities, PGE2, MMP-8, IL-1β, TNF-α and MDA levels, gingival tissue RANKL/OPG mRNA expressions were observed with increases of inflammatory cell infiltrations, and decreases of collagen fiber occupied regions in gingival tissues, osteoblast cell numbers and OB/BS (osteoblast cell activities), and alveolar bone volumes at histopathological inspections; these are means periodontitis and alveolar bone losses were relatively well induced by ligature placements, in the present study. However, anti-inflammatory and anti-oxidative effects - decreases of gingival MPO and iNOS activities, PGE2, IL-1β, TNF-α and MDA levels, numbers of inflammatory cells infiltrated in gingival tissues, and decreases of MMP-8 contents and increases of collagen fiber occupied regions in gingival tissues, favorable inhibitory effects on EPD related alveolar bone losses - decreases of alveolar bone loss scores, RANKL/OPG mRNA expressions, increases of osteoblast cell numbers and activities, alveolar bone volumes were demonstrated by oral administration of three different dosages of CM-SD 200, 100 and 50 mg/kg, dose-dependently. Especially, CM-SD 200 mg/kg treated rats showed favorable anti-inflammatory activities as comparable to IND 5 mg/kg, and CM-SD 50 mg/kg treated rats showed favorable alveolar bone preserve activities as comparable to IND 5 mg/kg, at least in a condition of this study.
CONCLUSION: The results obtained in this study suggest that oral administration of CM-SD has favorable inhibitory effects on ligation-induced periodontitis and related alveolar bone losses, through anti-inflammatory, anti-oxidative, MMP-8 inhibitory and RANKL/OPG mRNA expression modulatory activities, dose-dependently, as comparable anti-inflammatory activities to IND 5 mg/kg in CM-SD 200 mg/kg, and as comparable alveolar bone preserve effects to IND 5 mg/kg in CM-SD 50 mg/kg, at least in a condition of the current experiment. It, therefore, is expected that appropriate oral dosages of CM-SD will be promising as a new potent protective or therapeutic agent for relieving periodontitis and related alveolar bone loss, through more detail and plentiful animal studies, in future.
