CO₂ 농도는 3% (DCW 3.33 g/L, day 11)일 때, 접종 농도는 5×10⁴ cells/mL (DCW 3.08 g/L, day 8)일 때 E. gracilis의 성장이 가장 좋았다. CM 배지에서 PPC일 때 빛 주기가 길수록 건조 세포 중량 (2.01 g/L, day 14), μ (0.28/ day) 및 생산성 (0.14 g/L/day)이 높았고, HPC일 때 어둠 주기가 길수록 건조 세포 중량 (1.88 g/L, day 9), μ (0.42/day) 및 생산성 (0.14 g/L/day)가 높게 나타났다. CSL에서 PPC일 때 16:8의 광주기에서 건조 세포 중량 (3.69 g/L, day 7), μ (0.69/day) 및 생산성(0.58 g/L/day)이 높았고, HPC일 때 또한 16:8의 광주기에서 건조 세포 중량 (2.84 g/L, day 5), μ (0.56/day) 및 생산성 (0.56 g/L/day)이 높았다. CM 배지에서 HPC일 때의 파라밀론은 8:16의 광주기에서 0.4 g/L (day 11)이었고, CSL에서 HPC일 때 8:16의 광주기에서 0.63 g/L (day 4)으로 가장 높았다.

Euglena gracilis has been used in cosmetics, feed, fuel, health food, and pharmaceuticals in the industry because it contains proteins, minerals, paramylon (β-1,3 glucan), α-tocopherol and lipids. In order to cultivate E. gracilis containing a lot of these high-value products, nutrients are needed. As the culture scale increases, the cost of the medium is increased, it was tested whether the CM medium, which is one of the synthetic medium, could be substituted to the corn steep liquor from the food industrial by-product. In addition, because the pre-cultivation condition and L:D cycle affect the growth of microalgae, growth of E. gracilis and concentration of the product was tested when given three L:D cycles, two pre-cultivation conditions. So, test was conducted according to the concentration of CO₂, inoculum concentration, nitrogen source, and nitrogen concentration under photoautotrophic conditions in order to examine the influence of the culture factors on biomass growth. Growth of E. gracilis was good when the concentration of CO₂ was 3% (DCW 3.33 g/L, day 11), the inoculation concentration was 5×10⁴ cells/mL (DCW 3.08 g/L, day 8), nitrogen source was 250 ppm of (NH₄)₂SO₄ (DCW 3.27 g/L, day 8). In the mixotrophic cultivation, experiment was conducted by applying values obtained from photoautotrophic cultivation. In the CM medium, the longer the light cycle when photoautotrophic pre-cultivation (PPC), the higher the DCW (2.01 g/L, day 14), μ (0.28/day) and productivity (0.14 g/L/day) and the longer the dark cycle when heterotrophic pre-cultivation (HPC), the higher DCW (1.88 g/L, day 9), μ (0.42/day) and productivity (0.14 g/L/day) were high. However, DCW was lower in mixotrophic than photoautotrophic condition, was regarded that organic matter added to CM medium inhibited the growth of E. gracilis. In the CSL, the DCW (3.69 g/L, day 7), μ (0.69/day) and productivity (0.58 g/L/day) were high when PPC and 16:8 of L:D cycle, and DCW (2.84 g/L, day 5), μ (0.56/day) and productivity (0.56 g/L/day) were high when HPC and 16:8 of L:D cycle. CSL showed 4~5 times higher biomass productivity when compared with CM medium, depending on pre-cultivation and L:D cycle. The product of E. gracilis was analyzed for concentration of paramylon and PUFAs. In the CM medium, paramylon was the highest 0.4 g/L (day 11) when 8:16 of L:D cycle and PPC. In the CSL, paramylon was the highest 0.63 g/L (day 4) when 8:16 of L:D cycle and HPC. When cultivated in CSL, paramylon was produced about 1.6 times more at a time about 3 times shorter than CM medium. However, since CSL is not have supplemental nutrient and concentration of paramylon is low when compared to other literatures. it is considered that the concentration of paramylon can be increased if appropriate nutrition is added. The concentration of PUFAs in the CSL was 1.54 times higher than CM medium. Also, the PUFAs content of E. gracilis grown in CSL was higher compared to other literature. Therefore, CSL seems to have an advantage in the production of PUFAs. In conclusion, CSL was possible to replace CM medium for E. gracilis cultivation, and pre-cultivation and L:D cycles affected E. gracilis growth, paramylon and PUFAs concentrations.
