The aim of this study was to isolate a lytic bacteriophage that specific for Salmonella Enteritidis and examine its efficacy for controlling a cocktail of S. Enteritidis in raw chicken breast meat. Four lytic phages were isolated from a river in the vicinity of a duck farm (Anseong, Gyeonggi-do, Korea). All four phages exhibited broad lytic activities against 11 S. Enteritidis, 11 S. Typhimurium, S. paratyphi A, S. Sandiego, and S. Typhi, and which are belong to the Myoviridae and Siphoviridae families. They were determined to be stable, exhibited similar degrees of resistance to heat and pH, latent periods ranging from 5 to 30 min and burst sizes from 30 to 294 PFU/ml. In addition, the phage particles were 100% adsorbed within 18 to 40 minutes. We determined the activity of the four bacteriophage strains by assessment of lysis at MOI 104 of S. Enteritidis cultured in vitro. There were significant differences among the four phages, with CAU-SEP-4 resulting in the most significant decrease. In light of these findings, bacteriophage CAU-SEP-4 was selected to control cocktails of S. Enteritidis in vitro. We determined the activity of the individual bacteriophage strains and as a cocktail by assessment of lysis at MOI 104 of a cocktail S. Enteritidis cultured in vitro. As expected, the phage cocktail treatment was significantly more active compared to treatment with bacteriophage CAU-SEP-4 alone. In light of these findings, the bacteriophage cocktail was selected to control cocktails of S. Enteritidis on chicken breast meat. Viable cell counts were significantly reduced (P < 0.05) in raw chicken breast samples when bacteriophage cocktail was treated at 4 °C for 7 days (MOI from 104 to 106). These results indicate the potential efficacy of this bacteriophage cocktail as a biological agent against S. Enteritidis in raw chicken breast meat.
