A Study on Antioxidant and Antimicrobial Activities of Cosmos bipinnatus Flower Extracts for Cosmeceutical Formulations This study analyzed the contents and compositions of free amino acids, free sugars, phenolic acids, total polyphenol and flavonoid contents of hot water and 80% methanol extracts of C. bipinnatus flowers. Their antioxidant and antimicrobial activities were also investigated and results are as follows. A total of 17 kinds of free amino acids were isolated in each of the extracts. The total amino acid content of the hot water extract was 21.90 mg / g with lysine present at the highest concentration of 6.75 mg / g. In the case of the 80% methanol extract, the total amino acid content was 15.09 mg / g with glutamic acid present at highest concentration 4.20 mg / g. There were 5 kinds of protein-free organic compounds with a total content of 2.58 and 1.86 mg / g in the hot water and 80% methanol extract, respectively. 3 kinds of free sugars were isolated only in the hot water extract. The total polyphenol contents in the hot water and 80% methanol extracts were 58.44 ± 2.74 62.21 ± 1.92 mg / g respectively. On the other hand, the total flavonoid contents in the hot water and 80% methanol extracts were 54.68 ± 1.56 and 77.98 ± 1.90 mg / g respectively. These results show that both total polyphenol and total flavonoid contents are slightly higher in the 80% methanol extract. 4 different kinds of phenolic acids and 1 flavonoid were isolated only from the hot water extract. Rosmaricinic acid was the most abundant phenolic acid with 3,384.3 ug / g, and the most common flavonoid was quercetin (15.968 ug / g). No phenolic acids or flavonoid compound was identified in the 80% methanol extract. DPPH radical scavenging activity ranged from 66.36 to 71.79% with the concentrations of the hot water and 80% methanol extracts used (50, 100, 150, 300 ug / ml). Both extracts showed a similar trend where their radical scavenging activity increasing with increasing concentrations. ABTS radical scavenging activity ranged from 80.44 –82.46% at sample concentrations(50, 100, 150, 300 ug / ml). Similarly, both extracts showed a similar trend where their radical scavenging activity increasing with increasing concentrations. The FRAP ability also tended to increase with increasing sample concentration in both extracts. In the hot water extract, the antibacterial activity against B. subtilis, E. coli, K. pneumonia, P. aeruginosa, and S. typhimurium showed no difference even with increasing concentrations. However, it was able to inhibit S. aureus growth with a 17% decrease in growth observed at 1000ppm. Similarly, the antibacterial activity of 80% methanol against B. subtilis, K. pneumonia, P. aeruginosa, S. typhimurium showed no significant effect with increasing treatment concentrations. However, E. coli showed a decrease of about 4.6% growth at 1000ppm compared to that of 700ppm. Moreover, S. aureus showed a decrease of 10.5% growth at 1000 ppm.
