본 연구는 니파팜(Nipa palm), 몰로키아(Molokhia) 및 핑거루트(Finger root) 추출물들이 화장품의 기능성 천연 원료로써 적용 가능성을 살펴보기 위하여 실시하였다. 추출물은 에탄올을 용매로 2시간 동안 가열 환류시키고 이를 여과한 후, 농축 및 동결건조 등의 공정을 통하여 얻어졌다. UV-vis 분광광도계를 사용하여 추출물의 자외선 흡수 스펙트럼을 살펴본 결과, 니파팜과 핑거루트에서 자외선 UVB 영역의 빛을 흡수하는 피크가 나타났다. UV-vis 분광광도계에 적분구를 장착하여 각 추출물의 투과율을 비교한 결과, 핑거루트는 UVB 영역의 빛을 차단하는 효능이 우수하였다. DPPH 라디칼 소거능 분석 실험에서 니파팜의 항산화 활성은 SC₅₀ 값이 48 ppm으로 다른 추출물에 비하여 매우 우수하게 나타났다. 니파팜 추출물의 총 폴리페놀 함량은 Folin- Ciocalteu 시약을 사용하여 정량화했을 때, 대략 275 GAE ㎎/g의 값으로 나타났다. 니파팜 추출물의 항산화 활성은 UV나 고온의 조건에서도 4주 동안 비교적 안정하게 유지되었다. 추출물들의 항균활성을 살펴보기 위하여 황색포도상구균(그람 양성)과 대장균(그람 음성)에 대한 디스크 확산 시험을 실시한 결과, 핑거루트 추출물에서만 두 세균 모두에게서 Clear Zone이 생성되어 항균효과가 있음을 알 수 있었다. 콜라게나아제 활성저해 분석시험을 통하여 니파팜 추출물이 주름방지에 우수한 효능이 있음이 나타났다. 본 연구의 결과들을 종합해보면, 니파팜 추출물은 항산화제 및 주름 방지제로써, 핑거루트 추출물은 자외선 차단제 및 항균제로써 화장품에 활용 가능성을 보여 주었다. 이와 같은 결과들을 바탕으로 니파팜과 핑거루트 추출물을 적용한 화장품 제조를 실시하였다. 화장품 제조에 앞서, 추출과정에서 용매와 추출방법이 추출물에 미치는 영향을 먼저 살펴보았다. 니파팜 추출에서 용매는 에탄올 또는 물 단독으로 사용했을 때 보다 물과 에탄올 혼합물을 사용하였을 때가 항산화 효능이 더 우수하게 나타났고 특히 50:50 비율로 혼합하였을 때 항산화 효능이 가장 우수하였다. 50:50 비율로 용매를 고정하고 가열 환류, 초음파, 교반 등과 같은 3가지 추출 방법을 비교한 결과, 추출 방법은 니파팜 추출물의 효능에 큰 영향을 미치지 않았다. 핑거루트 추출에서도 에탄올 단독으로 추출할 때보다는 물과 에탄올을 혼합물로 추출했을 때가 항산화 효능이 매우 우수하게 나타났다. 그러나 핑거루트의 자외선 차단효능 비교에서는 에탄올을 단독으로 추출하였을 때가 효능이 가장 우수하였고, 용매 혼합물에서는 물의 양이 많아질수록 추출물의 자외선 차단효능이 감소하는 결과가 나타났다. 물과 에탄올을 혼합용매로 니파팜과 핑거루트 추출물을 다량으로 확보한 후, 이들 추출물을 다양한 비율로 처방한 O/W 선스크린 화장품 제형을 제조하고, 이들 화장품 제형의 여러 특성을 비교하였다. 모든 화장품 제형은 처방 비율에 상관없이 pH 6 내외의 약산성으로 나타났다. 외관은 니파팜 추출물의 적갈색과 핑거루트 추출물의 노란색이 혼합되어 황갈색의 선명한 색상을 나타났지만 이들 추출물에 이산화티탄을 혼합한 경우에 제형의 색상은 옅어지고 무채색에 가까워지게 된다. 니파팜과 핑거루트 추출물은 화장품 제형의 점도, 경도, 크림성 등을 감소시키는 반면, 이산화티탄은 이들 물성을 높여주었다. 핑거루트 추출물은 합성 자외선 흡수제인 옥토크릴렌(octocrylene)과 거의 동등한 수준의 자외선 차단 효과를 보여주었고, 이산화티탄과 혼합된 제형에서는 옥토크릴렌보다도 더 우수한 자외선 차단효능을 보여주었다.

This study attempted to investigate a usability of nipa palm, molokhia and finger root extracts as a natural cosmeceutical ingredient. For this, each extract was thermally refluxed for 2 hours, using ethanol as a solvent. Then, it was filtered, concentrated and freeze-dried. According to analysis of ultraviolet rays absorption spectrum, using UV-VIS spectrophotometer, a peak absorbing UVB light was found in nipa palm and finger root extracts. When the total transmittance was measured after connecting an integrating sphere to the UV-VIS spectrophotometer, finger root revealed great efficiency in blocking UVB light. From DPPH radical scavenging assay, nipa palm extract was far superior to other extracts in terms of antioxidant activity with 48ppm (SC50). Total polyphenol contents in nipa palm extract were 275 GAE mg/g when quantified, using the Folin–Ciocalteu reagent. In terms of antioxidant activity, furthermore, nipa palm extract remained relatively stable for 4 week even at UV or high-temperature conditions. To examine the extracts’ antimicrobial activities, a disk diffusion test was performed on Staphylococcus aureus (gram-positive) and E. coli (gram-negative). A clear zone was found in both bacteria in finger root extract only, confirming antimicrobial effects. The anti-wrinkle effects of nipa palm extract were confirmed through the collagenase inhibition assay. The above results show that nipa palm extract would be available as an antioxidant and anti-wrinkle ingredient while finger root extract could be used in developing UV-blocking and antimicrobial cosmetics. Based on the above results, cosmetics containing nipa palm and finger root extracts were developed. Prior to such development, the effects of a solvent and extraction method on extracts were investigated. In nipa palm extraction, mixture of water and ethanol as a solvent was greater than water only in terms of antioxidant activity. In particular, the antioxidant activity was the highest in a 50:50 mixture. When three extraction methods, such as thermal reflux, ultrasonic, and stirring, were compared, using the 50:50 mixture as a solvent, they had no significant influence on the antioxidant activity of nipa palm extract. In finger root extraction as well, mixture of water and ethanol was far greater than ethanol only in terms of antioxidant effects. However, the UV-blocking effect of finger root was the highest when ethanol was solely extracted. In solvent mixture, as the amount of water increased, the extract’s UV-blocking effect decreased. O/W sunscreen formulations were manufactured using the extracts in different ratios, and their properties were compared. All cosmetic formulations were mildly acidic with about pH 6 regardless of the mixing ratio. In terms of appearance, vivid tawny color, a combination of reddish brown (nipa palm) and yellow (finger root), appeared. However, when TiO₂was added to the extracts, the color became lighter, almost achromatic. While nipa palm and finger root extracts reduced the viscosity, hardness and creaminess of cosmetic formulations, TiO₂enhanced their physical properties. In terms of UV-blocking effects, finger root extract was almost equivalent to synthetic octocrylene, but the finger root extract was superior to the octocrylene when TiO₂was mixed together in the formulation.
