김(Porphyra spp.)은 해조류 중 홍조류의 일종으로 한국, 중국, 일본에서 널리 생산·소비되고 있다. 그러나 김의 일반성분 및 효능에 대한 보고가 일부 행해졌을 뿐 그 유용성분 및 생리활성에 대한 연구는 미흡한 실정이다. 그래서 본 연구에서는 잇바디돌김의 지방세포 분화억제 활성을 평가하고, 김에 함유된 유용한 화합물들을 분자수준에서 밝히고자 하였다. 지방세포인 3T3-L1 세포에 잇바디돌김 50% MeOH 추출물을 처리한 결과, 농도 의존적으로 peroxisome proliferator-activated receptor-γ2 (PPAR-γ2), adiponectin, 그리고 leptin의 발현을 억제하여 지방세포 분화를 저해하는 효과가 있음이 확인되었다. 이어 김에 함유된 유용한 화합물들을 분자수준에서 밝히고자, 잇바디돌김 50% MeOH 추출물(42.92 g)을 75% MeOH로 용해시켜 원심 분리하여 얻은 상등액 (30.2 g)을 대상으로 silica gel 및 ODScolumn chromatography, MPLC 및 ODS-HPLC을 실시하여 총 6종의 화합물을 분리하였다. 단리된 6종의 화합물 들을 대상으로 NMR 분석을 행한 결과, glutamic acid (1), arachidonic acid (2), shinorine (3), porphyra-334 (4), α–D-galactopyranosyl(1→6)-β–D-galactopyranosyl 1-O-glycerol (5), palythinol (6)로 구조해석 되었다. 이 중 동정된 화합물 3, 4, 그리고 6은 해조류 및 해양 동물계에 존재하는 mycosporine-like amino acid류로써 항산화 작용을 비롯하여 항염증, UV 차단, 항피부노화, 항피부암 등의 다양한 생리활성을 갖는 유용성분으로 보고된 바 있다. 이에 잇바디돌김에 함유된 MAA류 화합물인 shinorine 및 porphyra-334는 김의 지표성분으로 활용이 기대되어 본 연구에서 ODS-HPLC 분석에 의해 잇바디돌김에서 두 화합물의 정성 및 정량 방법을 확립하였다. 또한 shinorine 및 porphyra-334의 지표성분 및 다양한 생리활성 평가에 활용하기 위한 일환으로, silica gel- 및 ODS-MPLC를 이용하여 잇바디돌김 50% MeOH 추출물로부터 두 화합물을 신속하게 분리할 수 있는 방법을 확립하였다. 본 연구 결과는 김 품종의 성분 분석, 다양한 생리활성 구명, 품질관리, 저장성 평가 등에 있어서 기초 자료로 활용 가능성이 제시되었다.

The bioactive compounds such as polysaccharides, carotenoids, minerals, and mycosporine-like amino acids are presented in laver. However, studies on biological effect and chemical constituents of laver have not yet been performed. This study was to examined the effect of P. dentata 50% methanol extract in lipid accumulation in 3T3-L1 adipocytes. Treatment of P. dentata 50% methanol extract resulted in a significant decrease in lipid accumulation in 3T3-L1 adipocytes, which showed correlation with a reduction of mRNA expression of adipogenic genes, including peroxisome proliferator-activated receptor-γ2 (PPAR-γ2), adiponectin, and leptin. Additionally, purification and isolation of bioactive compounds from 50% methanol extract of laver was performed. Six compounds were purified and isolated by flash column chromatographies of silica gel and ODS. These compounds were identified as glutamic acid (1), arachidonic acid (2), shinorine (3), porphyra-334 (4), α–D-galactopyranosyl(1→6)-β–D-galactopyranosyl 1-O-glycerol (5), and palythinol (6), based on NMR experiments. Compound 3, 4 and 6 are mycosporine-like amino acids that are contained in the seaweed and have biological effects, such as antioxidant, anti-cancer, anti-inflammatory, and UV protective activities. Mycosporine-like amino acids identified in this study, porphyra-334 and shinorine were considered as useful compounds, which contribute to biological effect of P. dentata. Quantitative analyses of shinorine and porphyra-334 in P. dentata was established. Additionally, a simple method for purification of high purity shinorine and porphyra-334 from P. dentata was established. These results warrant further studies for evaluating the quality and biological effects of P. dentata.
