Bisdemethoxycurcumin (BDMC) is a non-toxic natural yellow pigment extracted from the rhizome of Curcuma longa L., which is a common food stuff. The aim of this study was to investigate the antibacterial activity of BDMC and the antimicrobial synergism of BDMC combined with antibiotics against methicillin-resistant Staphylococcus aureus (MRSA). Also, BDMC tested anti-toxic and anti-inflammatory efficacy the pathogenic toxin factor at sub-inhibitory concentration as well as bacterial activity by the toxicity of S. aureus. A broth microdilution assay was used to determine the minimal inhibitory concentration (MIC) of BDMC. Against all strains, the MIC of BDMC was 7.8 μg/mL. The data indicated that BDMC had high antimicrobial activity against MRSA. The synergistic effects of BDMC combined with antibiotics were determined using a checkerboard dilution test and time-kill curve assay and the results showed that the antimicrobial effect of combined therapy had obvious superiority, particularly BDMC combined with gentamicin (GT). The BDMC was analyzed in combination with membrane-permeabilizing agents Triton X-100, and ATPase inhibitors N, N'-dicyclohexylcarbodiimide. Addition of membrane-permeabilizing agents or ATPase inhibitors together with a low dose of BDMC potentiated BDMC anti-MRSA activity. The producing penicillin-binding protein 2a (PBP2a) protein level was analyzed using western blotting. The expression of PBP2a level was observed reduced while sample were treated with BDMC compared with untreated. In addition, we analyzed BDMC inhibite the transcriptional level of blaR1, blaZ, mecA and mecR1 gene in S. aureus by RT-qPCR. All the results revealed that BDMC is a potential natural antimicrobial agent. S. aureus produces multiple virulence factors, resulting in food infection and poisoning. Among the extracellular proteins produced by S. aureus that contribute to pathogenicity, the exotoxins (SEA, SEB and HLA) are thought to be of major significance. Sub-inhibitory concentration of BDMC decreased the production of SEA, SEB and HLA in MRSA in a dose-dependent manner. Here, the transcriptional level of sea, seb, hla and agr (accessory gene regulator) in S. aureus was inhibitied by BDMC, suggesting that the reduced production may be affect the secretion of exotoxins. Also, BDMC inhibits production of IL-6, TNF-α in inflammatory reactions induced by S. aureus exotoxin and NO production induced by LPS or PGN inhibited in a concentration dependent manners in RAW 264.7 cells. These findings suggest that BDMC may be useful for the treatment of MRSA infections when used in combination with β-lactam antibiotic. Also, the expression of virulence factors in S. aureus is sensitive to action of BDMC, which may be advantageous candidate in the inflammation treatment of toxin-producing S. aureus infection.
