Human epidermal growth factor (hEGF) is a polypeptide that has several biological functions, such as wound healing and anti-aging and is used in the pharmaceutical and cosmetic fields. Generally, the production of hEGF by Escherichia coli requires cell disruption and refolding of the protein because hEGF is produced inside the cells, which interfere with the efficiency and cost of its production. Therefore, the aim of this study was to engineer Pichia pastoris, which offers several advantages over E. coli, to produce and extracellularly secrete hEGF protein, and to analyze different variables that directly affect the protein production in order to identify the optimal conditions for the enhanced production. To this end, a high expression vector, pBJY_hEGF vector, was constructed. P. pastoris cells were transformed with the vector and proliferated in buffered glycerol-complex medium (BMGY) medium. Later, production of hEGF was induced with methanol in buffered methanol-complex medium (BMMY). In order to increase the production of hEGF, the effect of various parameters (carbon source of BMGY medium, cell density, methanol concentration and interval of addition) were investigated and hEGF production increased to 46.00 mg/L by one-factor-at-a-time (OFAT). Furthermore, the optimal parameters were obtained using the response surface methodology (RSM), which is statistical optimization. At the optimal conditions of 26.04 °C, pH 6.67, and 0.81% (v/v) methanol concentration, the predicted and actual hEGF production were 50.86 mg/L and 53.55 mg/L, respectively.
