Listeria monocytogenes is one of the foodborne pathogens that usually causes listeriosis. The biggest problem is a great viability since it can survive in a wide range of environments that are hard to control, especially ready-to-eat products, such as milk, cheese, sausage, and smoked fish. To improve the safety of the product and extend shelf life, it is essential to control L. monocytogenes. The purpose of this study is to isolate lactic acid bacteria with antimicrobial activity, evaluate its applicability as a cheese starter, and evaluate the inhibitory effects on Listeria monocytogenes. Furthermore, the study aims to determine the synergistic effect of combining nisin with bacteriophage in controlling L. monocytogenes. From raw milk, Lactococcus lactis strain with antibacterial activity was isolated and identified. The strain CAU2013 was a low acidifier making it a suitable candidate as an adjunct starter culture. The anti-listerial activity of strain CAU2013 was determined in milk broths at 30℃ to confirm its potential application in the food matrix. L. monocytogenes was not detected after 6 h incubation in skim milk broth whereas, 1 log unit decrease was observed after 9 h incubation in whole milk. The laboratory-scale fresh cheese was produced using the commercial starter culture TCC-3 as primary starter culture and L. lactis CAU2013 as an adjunct starter culture. L. monocytogenes (5 log CFU/g) was inoculated during cheese production and monitored during the storage at 4℃ and 10℃ for 5 days. The starter cultures reached to 9 log CFU/g during production, while L. monocytogenes decreased by 1 log unit during a storage. The synergistic effect of bacteriocin and Listeria phages was determined in cheese during the storage at 4℃ and 10℃ for 5 days. L. monocytogenes (6 log CFU/g) was inoculated on the sliced cheese and treated with 2 IU/㎕ nisin and varying concentration of bacteriophage (MOI 1, 10, 100). The treatment of nisin and bacteriophage was most efficient, reducing 3 to 6 log units of L. monocytogenes. The use of the bacteriocin-producing LAB as a starter culture resulted in an efficient inhibition of L. monocytogenes. Therefore, Lactococcus lactis CAU2013 can be used as a protective adjunct starter culture for a cheese production. Additionally, a synergistic effect was observed when combined bacteriocin and bacteriophage were used. This suggests that the combined use of bacteriocin and bacteriophage is more efficient in controlling L. monocytogenes during production and storage conditions and can improve the safety of the product leading to an increase of its shelf-life.
