Human salivary gland-derived stem cells possess a great potential to differentiate into multiple lineages and self-renewal capacity, allowing them to be utilized as patient-specific cell-based therapeutics. Therefore, we propose the alternative patient-derived stem cell therapy to repair damaged salivary glands and generate new salivary gland tissues. First, we focus on development of radiation induced dry mouth animal model. After receiving X-ray irradiation(15Gy) on C57BL/6 mice, histological evaluation and qPCR analysis of cells within submandibular gland tissue were performed. ECM(Extra cellulcar matrix) remodeling and dysfunctions of salivary gland were observed on various histological evaluation. And qPCR results demonstrated that inflammation, fibrosis, apoptosis and dysfunctions of salivary gland were observed. Second, we performed the isolation of patient's salivary gland tissue-derived mesenchymal stem cell successfully. And we observed mesenchymal stem cell phenotypes from patient's salivary gland tissue-derived cells on Flow cytometry. Also, our initial findings demonstrated that cells cultured on 3D bioprintable matrix were able to survive, proliferate, and form acinar cell-like 3D organoids. In summary, we successfully created a radiation-induced dry mouth animal model and evaluated dysfunctions of salivary gland tissue and its residing cells by histological and gene expression analyses. In addition, we also demonstarated that patient specific stem cells could be isolated from salivary gland with mesenchymal stem cell potintial and these cells could undergo self-assembled organoid formation.
