The study of preserving blood stains has been mostly related to the storage temperature after collection, but a new extraction reagent for blood stains at crime scene has been developed as a preceding study of this study. The composition of the developed extract reagent was developed based on the composition of PBS useful for preserving TE and protein useful for preserving and extracting genes. Although the gene extraction performance of the extracted reagent developed in the previous studies has been studied, there has been no research on the protein extraction performance. This study evaluated the protein extraction ability using reagents developed as blood spatter extraction reagents and evaluated the possibility of actual crime field application. Blood stains were collected from 23 healthy men and women, and the protein was extracted five times a month under various temperature and humidity conditions after blood was collected into a vacuum blood vessel without coagulant or anticoagulant. The protein extraction performance of TE and PBS, which are considered as extraction reagents, and the extraction performance of the developed extraction reagents were compared and analyzed in the existing blood stain extraction D.D.W. and the previous studies. The protein extraction ability of the extraction reagents developed using two test materials was cross-validated and the developed extraction reagents were confirmed to have excellent performance in the preservation of proteins as well as genes. Also, the capacity and collection tool of optimized extraction reagents from the process of collecting blood in the field to the analysis stage were presented using swabs before, and the possibility of the developed extraction reagent being used in the crime scene was evaluated by applying the method presented with the extraction reagent in the blood collected in the actual field.
