본 연구는 벚나무 추출물의 Raw 264.7 세포에서 세포증식억제효과와 LPS로 유도된 세포독성에 대한 억제효과, NO 생성능 억제효과를 보였다. 또한, RAW 264.7 세포에서 TNF-α와 IL-6, IL-1β의 사이토카인 함량은 LPS 처리 군에서 가장 높았고, LPS에 벚나무 추출물을 처리한 군에서 추출물 농도가 높을수록 유의하게 낮았다(P<0.05). 벚나무 추출물은 LPS로 유도된 Raw 264.7 대식세포에서 전 염증성 사이토카인 발현을 감소시켰다. 천식효능을 검증하기 위하여 5주령 수컷 BALB/c mice로 ovalbumin 유도 천식질환모델을 이용하여 시험물질인 벚나무(Prunus serrulata var. spontanea) 천연물(이하 PS)의 투여로 인한 싸이토카인과 ovalbumin specific IgE 개선 효능, 폐조직 병리검사를 분석하여 항염증 효과 및 천식 효능을 확인하고자 하였다. 벚나무 추출물의 투여로 ovalbumin 유도 천식질환모델에서의 기관지폐포세척액(BALF) 내 염증세포 변화에 대한 효과를 본 결과 총 Total cell(WBC)를 포함한 Eosinophilc, neutrophil, lymphocyte, monocytes의 세포 수가 벚나무 추출물로 인해 유의하게 감소하였다. 혈청 내 싸이토카인 계열 TNF-alpha, IL-4, 면역글로불린 (IgE) 농도는 천식유발 음성 대조군에서 정상군에 비하여 유의적으로 증가하였는데, PS 투여에 의하여 감소시키는 효능을 보였다. Ovalbumin 유도 천식질환모델에서 폐포와 세기관지 주위에 침윤된 염증세포들이 증가된 것을 확인할 수 있었고, 벚나무 추출액을 투여한 결과 세기관지 형태학적 변형이나 점막 비후 현상이 다소 개선되는 것으로 관찰되었다. 이는 벚나무 추출물의 투여로 인해서 폐포 조직의 염증발현 억제와 조직손상을 현저히 감소시키는 것으로 확인할 수 있었다. 이상과 같은 결과로 벚나물 추출액의 천식 염증 개선 효능이 우수한 것을 확인하였으며, 본 연구의 벚나무(Prunus serrulata var. spontanea)추출물 이외의 부위별 천식효과에 유용한 물질 탐색 연구가 필요하다고 사료된다. 이상의 결과로부터 벚나무 추출물이 폐포 조직의 염증 발현 및 조직 손상을 유의하게 감소시키는 것을 확인하였으며, 이는 벚나무 추출물이 천식 염증 개선에 탁월한 효능이 있음을 확인하였다.

This paper aims to establish Prunus serrulata var. spontanea’s potentials and applications, by defining its terms and functional effects. In this study, using an ovalbumin-induced asthma disease model using 5-week-old male BALB/c mice, the efficacy of cytokine and ovalbumin-specific IgE improvement due to the administration of a natural product (PS) of cherry tree (Prunus serrulata var. spontanea) as a test substance, lung tissue pathology was analyzed to confirm anti-inflammatory and asthmatic effects. Also, a metabolite analysis for separation and identification of the functional materials was analyzed. The purpose of Study 1 was to analyze the anti-inflammatory effect of Prunus serrulata var. spontanea. As a result of an in vitro cytotoxicity test using RAW 264.7 cell, NO production was significantly reduced when the Chung-Dae extracts were treated at concentrations of 2, 8, and 16 μg/mL (p<0.05). The pro-inflammatory cytokines TNF-α, IL-6 and IL-1β significantly reduced when treated with Prunus serrulata var. spontanea extract at concentrations of 2, 8, and 16 μg/mL compared to the LPS group. From the above results, it can be seen that the plant-derived compound, Prunus serrulata var. spontanea extract, inhibits the inflammatory response induced by LPS in RAW 264.7 cells. In particular, it regulates the inflammatory response by inhibiting the expression of pro-inflammatory cytokines (TNF-α, IL-6, IL-1β). The purpose of Study 2 was to analyze the anti-inflammatory effect of Prunus serrulata var. spontanea using an ovalbumin-induced asthma disease model. As a result of examining the effect on changes in inflammatory cells in bronchoalveolar lavage fluid in an ovalbumin-induced asthma disease model by administering this PS sample, the total cells (WBC), Eosinophic, neutrophil, lymphocyte, and monocytes was significantly decreased. Shekanin, isoflavon, catechin and procyanidins were the major active materials detected in Prunus serrulata var. spontanea.
