따라서 본 연구에서는 수산식품의 대표적 식중독 세균인 Vibrio parahaemolyticue 대한 균의 성장변화를 정량적으로 예 측하는 수학적 모델의 개발을 보고한 전보(13)에 이어 수산 가 공식품인 게맛살에서 또 하나의 미생물학적 위해 요소로 정해 져 있는 L. monocytogenee 대하여 제품에 균의 오염 시 균의 성장변화를 정량적으로 예측할 수 있는 수학적 모델의 개발을 위한 기초 자료를 제공하고자 온도, 초기균수의 변화에 따른 L. mgocytogeg의 성장 예측모델을 개발하였다.

제안 방법
MIC broth에서 온도와 초기 균수에 따른 L. monocytogenese] 최대 증식속도 상수(k)를 예측할 수 있는 정량 평가 모델을 polynomial model과 square root model을 이용하여 개발하였으며, 개발된 정량 평가 모델에 의하여 예측된 최대증식속도상수(k)를 이용하여 온도와 초기균수의 변화에 따른 L monocytogenes 의 성장을 Gompertz model로 예측하여 증식 곡선으로 나타내었다.
공시 균주의 배양 공시균주의 배양은 향후, 균의 성장 모델을 포장연제품인 게 맛살에 적용하기 위해 게맛살 성분조성(조단백 7.8%, 조지방 0.2%, 탄수화물 16%, 회분 2.5%, 수분 73.5%)을 고려한 MIC broth를 제조하여 사용하였다(Table 1).

균의 성장에 미치는 온도, 초기 균수의 영향을 알아보기 위하여 동결 보존한 균주를 45℃ water bath에서 급속 해동한 후, 미리 준비한 멸균 MIC broth 50mL에 접종하고 37℃ incubator (Sanyo, Japan)에서 24시간 전 배양 하였다. 전 배양한 균주를 200 mL의 MIC broth에 초기 균수가 1.0X102, 1.0X103, 1.0X 1(/ CFU/mL가 되도록 접종하고 온도 15, 20, 25, 37, 40℃에서각각 배양하였다
대상 데이터
본 실험에 사용한 균주는 Listeria monocytogenes ATCC 19113으로 한국생명공학연구원 유전자은행(KCTC)으로부터 분 양 받았으며 brain heart infusion broth(BHI, Difco Laboratories, Detroit, MI, USA)에 전 배양하여 1 nL의 vial tube에 15%의 glycerol# 첨가하여 -801에 동결보관하면서 실험 전 Modified Imitation Crab(MIC) broth에서 37℃, 24시간 전 배양 하여 사용하였다.
데이터처리
Parameter들의 결정과 정량 평가 모델의 개발 최대증식속도상수(k)와 온도 및 초기균수와의 상관관계를 규 명하기 위하여 Gompertz model로부터 구한 최대증식속도상수 (*)를 수학적 정량평가모델인 polynomial model 식 (2)과 square root model 식 (3)에 각각 적용하고(13), SPSS(SPSS Inc., 2000) program(Ver. 10.0)으로 각 식의 parameters- 결정한 후, Sigma Plot(SPSS Inc., version 7.0)으로 도식화 하였으며 각 배양 조건을 대입하여 실제 실험으로부터 구한 최대증식속도상수(k)와 모 델식으로부터 구한 최대 증식속도상수(k)와의 상관관계를 비교 하였다

각 실험구별 시간의 경과에 따른 균의 성장은 일정시간 간 격으로 Plate count agar(Difco Laboratories, Detroit, MI, USA)를 사용하여 표준평판배양법 (14)으로 생균수를 측정하여 나타내었다.

후속연구
gnocWogezzes의 온도에 따른 성장의 손쉬운 모.니터링과 정 량 적인 위해도 평가가 가능할 것으로 기대되며, 수분활성이나 좀 더 넓은 pH 범위, 여러 첨가물의 농도, 포장시 충진하는 N2 또는 CO2 농도나 이러한 조건들 간의 상호 작용 등, 물리 화학 적인 요인에 관한 추가적인 연구를 계속한다면, 보다 다양한 환경 조건하에서 보다 정확하게 L, monocytogenes의 성장을 정량적으로 예측할 수 있을 것으로 기대된다.
