건조 고등어 추출물에 의한 항산화 및 암세포주 증식 억제 효과 Effect of Extracts from Dried Mackerel on Antioxidant Activity and Inhibition of Growth of Human Cancer Cell Lines원문보기
본 연구에서는 건조 고등어를 용매별로 추출하여 세포 내활성산소종 감소 효과와 인체 결장암 및 섬유육종세포에 대한 증식 억제에 대해 검토하고자 하였다. A+M 추출물 및 MeOH 추출물과 각 분획물들을 5 mg/ml 및 10 mg/ml의 농도로 HT1080 섬유육종 세포에 처리하였을 때 blank군과 control군과 비해 측정 시간 120분 동안 계속적으로 세포 내활성산소종 크게 감소시켰다(p<0.05). 건조 고등어 추출물의 hexane, BuOH, 85% aq. MeOH, water 분획물들도 첨가농도 5 mg/ml에서 측정 시간 120분 동안 계속적으로 세포 내 활성산소종을 감소시키는데 우수한 능력을 나타내었다(p<0.05). 인체 섬유육종세포(HT1080) 증식 억제 실험에서 A+M 추출물과 MeOH 추출물은 둘 다 가장 낮은 농도인 5 mg/ml에서부터 농도 의존적으로 암세포의 증식을 억제시켰다(p<0.05). 건조 고등어 추출물을 hexane, BuOH, 85% aq. MeOH, water로 다시 추출하여 얻어진 각 분획물을 20 mg/ml의 농도로 HT1080 세포에 처리했을 때도 우수한 암세포 증식 억제효과(p<0.05)가 나타나 A+H 추출물 및 MeOH 추출물과 유사한 억제효과를 보였다. 인체 결장암세포(HT-29)에 대한 결과로 A+H 추출물과 MeOH 추출물은 HT1080 섬유육종세포의 결과보다는 다소 암세포 증식 억제 효과가 낮았으나 10 mg/ml 및 20 mg/ml의 첨가농도에서 높은 암세포 증식 억제표과를 보였다(p<0.05). 건조 고등어 추출물의 85% aq. MeOH, hexane, BuOH, water 분획물을 20 mg/ml의 농도로 인체 결장암세포에 처리했을 때 암세포 증식 억제효과를 나타내어 각각 80%, 88%, 96% 96%의 암세포 증식 억제효과(p<0.05)를 보였다. 고등어는 고도 불포화지방산을 많이 함�되構� 있어 지질 산화의 용이성이라는 단점이 있었으나 본 연구의 결과로부터 고등어 유기용매 추출물은 세포 내 활성산소종 크게 감소시켰으므로 고등어에는 천영 항산화성 물질을 있는 것으로 여겨지며 지질과산화물로 인한 세포 및 조직의 손상을 예방할 것으로 기대된다.

This study was carried out to determine the inhibitory effects of solvent extracts from dried mackerel on $H_2O_2-induced$ oxidative stress and growth of cancer cell lines (HT1080 human fibrosarcoma and HT-29 human colon cancer cells). In order to determine the protective effect on $H_2O_2-induced$ oxidative stress, DCFH-DA (2',7'-dichlorodihydrofluorescin diacetate) assay was conducted. All fractions including crude extracts of dried mackerel appeared to significantly reduce the levels of intracellular reactive oxygen species (ROS) (p<0.05). Among the fractions, 85% methanol fraction showed a slightly higher protective effect on production of lipid peroxides. Inhibitory effects of acetone with methylene chloride (A+M) and methanol (MeOH) extracts on the growth of HT1080 and HT-29 cancer cells increased in a dose dependent manner (p<0.05) and the inhibitory effect was more significant on the growth of HT1080 cells. The treatments of hexane, 85% aq. methanol, butanol and water fractions significantly inhibited the growth of both cancer cells (p<0.05). These results indicate that the consumption of mackerel may be recommended as a potent functional food for preventing cellular oxidation and cancer.
