Extraction and preliminary purification의 변형한 방법을 적으시오. | 추출 및 일차정제(Extraction and preliminary purification): Flavonoid compound의 추출 및 일차 정제는 Park(1987) 및 Mun and Park(1995)의 방법을 변형하여 사용하였다. 즉, 상온에서 건조시킨 약 5~10 g의 잎을 분쇄하여 이로부터 85% aqueous methanol(MeOH) 200 ml로 2회, 50% MeOH 200 ml로 1회씩 상온에서 각각 12시간 flavonoid compound를 추출한 뒤, 이를 여과하여 rotary vacuum evaporator로 농축시켰다. 농축된 추출액은 chloroform과 ethyl acetate (EtOAc)를 사용하여 flavonoid compound만을 추출하였으며, 그 결과 얻어진 flavonoid compound를 포함하는 EtOAc fraction은 다시 rotary vacuum evaporator 를 사용하여 소량으로 농축하여, cellulose thin layer chromatography plate(Merck, thickness 100 µm, 20 cm ×20 cm)에 소량 spotting한 뒤 이를 TBA(tert.-butanolacetic acid-water, 3:1:1, v/v/v)와 15% HOAc(15% acetic acid in water)를 용매로 하여 2차원 전개시켜 UV light 하에서 각 분류군에 분포하는 flavonoid compound의 profile 을 확인하였다. 이후, 상기한 2-dimensional TLC 상에서 확인된 flavonoid compound를 분리하기 위하여 동일 EtOAc fraction을 Whatman 3MM chromatography paper 위에 가는 band 모양으로 streaking한 후 15% HOAc를 solvent로 하여 1차원으로 전개시켰다. 그 결과 paper chromatogram 상에서 분리된 각 flavonoid band를 가늘게 자른 후 100% MeOH로 elution하여 농축, 여과한 후 이를 high performance liquid chromatography(HPLC)를 사용하여 포함되어 있는 flavonoid compound들을 완전히 분리, 정제하였다. |
