The column for separation of four compounds was used Luna C18 column and maintained at 40℃. The mobile phase for gradient elution consisted of two solvent systems. The analysis was carried out at a flow rate of 1.0 mL/min with PDA detection at 254 and 280 nm. To evaluate production and expression of Th2 chemokines, ELISA and RT-PCR were conducted in tumor necrosis factor (TNF)-α and interferon (IFN)-γ-stimulated HaCaT cells with or without BPTSS or silymarin, a positive control for skin inflammation. To measure Th2 cytokines, primary mouse splenocytes were treated with BPTSS and performed ELISA for interleukin (IL)-4, 5, 13. Calibration curves were acquired with r2>0.9999. The contents of geniposide, liquiritin, baicalin, and glycyrrhizin in BPTSS were 5.06 ㎎/g, 7.33 ㎎/g, 27.56 ㎎/g, and 7.81 ㎎/g, respectively. BPTSS reduced TARC and RANTES production and mRNA expression in TNF-α and IFN-γ-treated HaCaT cells. BPTSS inhibited IL-4, 5, and 13 production in mouse splenocytes. Our data will be a helpful information to upgrade quality control and anti-allergic effects of BPTSS.

본 연구에서는 인간 피부 상피세포와 마우스 비장세포를 사용하여 방풍통성산의 항알러지 효능을 검색하였으며, 이에 앞서 방풍통성산의 품질 관리를 위한 성분 동시 분석을 실시하였다. 이들 구성 성분들에 대한 HPLC 분석 결과가 이전의 여러 논문16-18)에서 발표된 바 있으며, 이들을 참고하여 분석하였다.
세포독성 평가를 위하여 96 well microplate에 1×103 cells/well의 농도로 세포를 분주하여, 방풍통성산을 농도별(0, 10, 20, 50, 100, 200, 500 ㎍/㎖)로 처리하여 24 시간 배양하였다. Cell counting kit(CCK)-8 solution을 10 ㎕씩 첨가하여 4 시간동안 반응시킨 다음, microplate reader(Benchimark Plus, Bio-Rad, MN, USA)를 사용하여 450 nm에서 흡광도를 측정하였다.
대상 데이터
8-10 주령의 수컷 BALB/c 생쥐 (Orient Bio, Seongnam, Korea)를 경추탈골하여 희생한 후 비장을 적출하였다. 비장으로부터 단일 세포를 분리한 후 RBC lysis buffer(Sigma-Aldrich, St.
HaCaT 세포는 American Type Culture Collection(Rockville, MD, USA)로부터 분양 받아 사용하였다. 세포 배양은 10% fetal bovine serum이 포함된 DMEM 배지를 사용하였으며, 37℃, 5% CO2 조건에서 실시하였다.
본 실험에 사용된 방풍통성산의 구성 한약재는 ㈜옴니허브(Youngcheon, Korea)와 ㈜HMAX(Jecheon, Korea)에서 구입하여 동국대학교 이제현 교수(Gyeongju, Korea)로부터 감별을 받은 후 사용하였으며, 각각의 구성 한약재들의 표본(2008-KE-23-1~KE-23-18)은 한국한의학연구원 한약방제연구그룹에 보관하였다.
이론/모형
Baker(Philipsburg, NJ, USA)에서 구입하였으며, glacial acetic acid는 특급시약으로 Junsei(Tokyo, Japan)에서 구입하여 사용하였다. 방풍통성산 내 주요 구성 성분의 함량을 분석하기 위하여 LC-20A 시스템(Shimadzu Co., Kyoto, Japan)을 사용하였으며, 데이터 분석은 LCsolution software(Version 1.24)를 이용하여 처리하였다. 4종 성분의 분리를 위해 Phenomenex사의 Luna C18(5㎛, 4.

성능/효과
HPLC 분석 결과 방풍통성산의 구성 한약인 치자의 geniposide가 5.06 ㎎/g, 황금의 baicalin이 27.56 ㎎/g, 그리고 감초의 liquiritin과 glycyrrhizin이 각각 7.33 ㎎/g, 7.81 ㎎/g으로 검출되었다.
또한 방풍통성산은 TNF-α와 IFN-γ에 의하여 유도된 TARC 및 RANTES mRNA 발현을 감소시켰다. Positive control로 사용된 silymarin11)의 효능과 비교하였을 때, 케모카인의 분비 억제 효능은 방풍통성산이 우수하였으나, mRNA 발현 감소에 대해서는 silymarin이 더 우수하였다.
이들 구성 성분들에 대한 HPLC 분석 결과가 이전의 여러 논문16-18)에서 발표된 바 있으며, 이들을 참고하여 분석하였다. 그 결과 구성 한약인 치자의 geniposide가 5.06 ㎎/g, 황금의 baicalin이 27.56 ㎎/g, 그리고 감초의 liquiritin과 glycyrrhizin이 각각 7.33 ㎎/g, 7.81㎎/g으로 검출되었다.

피부 상피 세포에 TNF-α, IFN-γ 등의 자극으로 생성된 TARC, RANTES와 같은 케모카인들은 c-c chemokine receptor type 4(CCR4)-positive Th2 세포에 결합하여 이들을 염증 조직으로 유도한다20). 본 연구 결과 방풍통성산은 TNF-α와 IFN-γ에 의한 TARC와 RANTES의 분비를 억제시키는 경향이 있음을 확인하였다. 또한 방풍통성산은 TNF-α와 IFN-γ에 의하여 유도된 TARC 및 RANTES mRNA 발현을 감소시켰다.
호산구는 염증 매개인자 분비를 유도하는 IL-5에 의하여 활성화 된다22). 본 연구의 결과 방풍통성산은 마우스 비장 세포에서 Th2 사이토카인인 IL-4와 IL-5의 분비량의 감소를 확인하였다.
후속연구
이상의 결과들은 방풍통성산의 품질관리를 위한 기초자료로 사용할 수 있을 것으로 사료된다. 또한 방풍통성산은 Th2 케모카인과 사이토카인의 생성 억제를 통하여 항알러지 효능을 나타내는 것이며, 이와 같은 결과는 향후 한약을 이용한 알러지 치료제 개발 연구의 기초 자료로 활용될 수 있을 것으로 사료된다.
이상의 결과들은 방풍통성산의 품질관리를 위한 기초자료로 사용할 수 있을 것으로 사료된다. 또한 방풍통성산은 Th2 케모카인과 사이토카인의 생성 억제를 통하여 항알러지 효능을 나타내는 것이며, 이와 같은 결과는 향후 한약을 이용한 알러지 치료제 개발 연구의 기초 자료로 활용될 수 있을 것으로 사료된다.
이상의 결과들은 방풍통성산이 Th2 케모카인 및 사이토카인 분비 억제시키는 경향이 있으며, 향후 아토피 피부염과 천식 특이적 실험 모델을 활용한 추가 실험을 통하여 보다 확실한 효능 검증이 필요할 것으로 사료된다.
이와 같은 결과는 방퐁통성산은 한약 복합 처방인 반면 silymarin은 단일 성분인 차이점으로 인한 것으로 사료되며, 향후 처방의 효능을 동등한 기준에서 비교하기 위한 대조군의 확보가 필요할 것으로 생각된다. Th2 사이토카인은 천식의 병리생리학적 측면에서 필수 인자이다.
현재까지 성분 동시분석에서 확인한 4가지 성분의 항알러지 또는 항알러지성 피부염 효능에 대해 발표된 논문은 없으며, 이들 성분들 중 항알러지 효능에 대한 활성 성분(active compound)을 확인하기 위한 추가 실험이 요구된다.
