Effect of three treatments, i.e., drying, leaching, and roasting, on flavonoid contents or flower was tested using multiple processing methods per treatment. Total flavonoid content decreased under the various drying methods, confirming that flavonoid content is heat-dependent. This finding was more pronounced for freezing and oven-drying (15.3 and 13.8 mg/g DW, respectively), with a 10% difference in the total flavonoid content between the two methods. Under leaching conditions, the flavonoid content decreased with increasing treatment time. Notably, roasting methods did not result in loss of flavonoid content. The total flavonoid content in the rapeseed varieties decreased in the following order: Youngsan, Tammi, Tamra, Naehan, Hanra, Mokpo No. 68, and Mokpo No. 111. Acqutiy Ultra Performance Liquid Chromatographic System (Waters Co., Milford, MA, USA), 을 사용하였고 Xevo G2-S Q-TOF Mass Spectrometer (Waters Co.)를 사용하여 ositiveion mode ([M+H]+)로 분석하였다.
HPLC 및 UPLC-Q-TOF/MS 분석을 통하여 유채꽃 내에서 19종류의 플라보노이드(Table 1 and Fig. 3)를 분리 및 동정하였다. 플라보노이드 성분은 isorhamnetin 계열 7종류, quercetin 계열 6종류, kaempferol 계열 6종류로 나타났다.

2014년 4월 18일 바이오에너지작물센터(농촌진흥청 국립 식량과학원, 무안)에서 ‘내한’, ‘목포 68호’, ‘목포 111호’, ‘영산’, ‘탐라’, ‘탐미’, ‘한라’ 7품종의 유채꽃을 수확하였다. 수확한 7품종의 유채꽃은 자연 풍건한 후 플라보노이드 함량을 분석하였다. 이후 실험은 플라보노이드 함량이 가장 높은 ‘영산’ 품종으로 진행하였다.
유채 꽃의 이용가치를 높이기 위한 기능성 차 개발을 하기 위하여 ‘5가지 건조조건’, ‘9가지 침출 조건’, ‘덖음 유무’에 따른 유채꽃 내 flavonoid 성분을 조사하였다.
유채꽃 1 g을 3가지 침출 온도(70, 80, 90℃) 및 시간(1, 3, 5분)으로 침출하여 유채꽃차 100 mL를 얻었다(1 g/100 mL). 유채꽃차 시료는 여과한 후 밀폐된 용기에 넣어 초저온 냉동고(SFDSF 12, Samwon frezing engineering Co.
자연 풍건한 유채꽃을 끓는 1% 소금물(w/v)에 약 1∼2 초 정도 담가 불순물 제거 및 소독을 하였다. 이후 소독한 유채꽃을 자연건조(실험실 내, 온실 내 자연건조), 열풍 건조 (40℃, 60℃ oven dry), 동결 건조 총 5가지 건조방법으로 건조를 진행하였다.
플라보노이드 추출은 유채꽃, 유채꽃 침출물에 따라 각각 다른 추출방법을 사용하였다(modified from Perez-Gregorio et al., 2010).

4 mg/g DW로 나타났다. 5가지 건조조건에 따른 flavonoid 함량은 동결건조(15.3)에서 가장 높았으며, 40℃ 열풍건조(14.9), 실내건조(14.4), 온실건조(13.9), 60℃ 열풍건조(13.8 mg/g DW) 순으로 나타났다. 9가지 침출조건에 따른 flavonoid 함량은 90℃ /5분 (13.
7가지 품종의 유채꽃 내 flavonoid 함량은 ‘영산’(19.4 mg/g DW)이 가장 높았고, 함량 범위는 11.5∼19.4 mg/g DW로 나타났다.
7품종 유채꽃 내 주요 플라보노이드 성분의 함량범위는 각각 isorhamnetin-3-O-sophoroside-7-O-D-glucoside이 4.4∼ 8.0, isorhamnetin-3-O-D-glucoside이 1.7∼3.6, kaempferol-3-O-D-glucoside-7-O-D-glucoside이 0.7∼2.0 mg/g DW으로 나타났다.
9가지 침출조건에 따른 flavonoid 함량은 90℃ /5분 (13.2)>80℃ /5분(11.5)>90℃ /3분(11.0)>80℃ /3분 (8.8)>70℃ /5분(7.4)>90℃ /1분(7.0)>70℃ /3분(6.3)>80℃ /1분(5.5)>70℃ /1분(3.8 mg/g DW) 순으로 나타났으며, 침출온도가 높을수록, 침출시간이 길수록 함량이 높았다.
Isorhamnetin-3-O-D-glucoside함량이 가장 높은 품종은 ‘탐미’였으며, 특이 하게 ‘목포 68호’는 나머지 6품종들과는 달리 isorhamnetin-3-O-D-glucoside (1.7)보다 kaempferol-3-O-D-glucoside-7-O-D-glucoside (1.8 mg/g DW)의 함량이 높게 나타났다.
각각 ‘목포 111호’에서 quercetin-3-O-sinapoylsophoroside-7-O-D-glucoside이, ‘내한’, ‘탐라’, ‘목포 111호’,에서 isorhamnetin-3-O-gentiobioside-7-O-D-glucoside 이 검출되지 않았다.
따라서 분석 결과 ‘영산’ 품종, 동결건조, 90℃ /5분 침출, 덖음 처리하였을 때 유채꽃차의 플라보노이드 고 함유 기능성 개발에 유용할 것이라 사료된다.
모든 품종에서 총 플라보노이드 함량 대비 각 성분별 비율은 isorhamnetin-3-O-sophoroside-7-O-D-glucoside (42), isorhamnetin-3-O-D-glucoside (19), kaempferol-3-O-D-glucoside-7-O-D-glucoside (9%) 순으로 높았고, 각 성분 함량을 계열별로 합쳐 정리한 결과(Table 3.), isorhamnetin 계열(10.5), kaempferol 계열(3.1), quercetin 계열(1.3 mg/g DW) 순으로 isorhamnetin 계열이 가장 높았다.
본 실험에서 덖음 처리에 따른 flavonoid 함량은 덖음 차에서 12.3 mg/g DW으로 나타나 무처리 (5.9 mg/g DW)에 비해 약 2.1배 이상 높았다.
본 실험에서 자연건조와 열풍건조에 따른 플라보노이드 함량을 비교하였을 때 자연건조 그룹(실내, 온실)의 플라보노이드 함량 평균값은 14.2 mg/g DW이었으며, 열풍건조 그룹(40, 60℃)은 14.3 mg/g DW 으로 두 그룹 간 함량 차가 거의 없었다. 유채꽃을 자연건조(246.
, 2013). 본 실험에서는 40℃ 열풍건조(14.9)에 비해 동결건조 시료(15.3 mg/g DW)에서 플라보노이드 함량이 높게 나타나 이전 실험들과 차이를 보였다.
소독한 후 5가지 건조방법에 따라 건조한 ‘영산’ 유채꽃내 총 flavonoid 함량은 동결건조시료에서 15.3 mg/g DW으로 가장 높았으며 이어 40℃ 열풍건조(14.9)>실내건조 (14.4)>온실건조(13.9)>60℃ 열풍건조(13.8 mg/g DW)순으로 나타났다(Table 4).
유채꽃 내 flavonoids는 3가지 계열(isorhamnetin계열 7종류; quercetin계열 6종류; kaempferol계열 6종류)이었으며, 총 플라보노이드 함량 대비 isorhamnetin계열 비율이 가장 높았다.
