AMP-activated protein kinase (AMPK) functions as a metabolic master through regulating and restoring cellular energy balance. In skeletal muscle, AMPK increases myofibril protein degradation through the expression of muscle-specific ubiquitin ligases. Mori Folium, the leaf of Morus alba, is a traditional medicinal herb with various pharmacological functions; however, the effects associated with muscle atrophy have not been fully identified. In this study, we confirmed the effects of AMPK activation by examining the effects of 5-aminoimidazole-4-carboxamide ribonucleotide (AICAR), an activator of AMPK, on the induction of atrophy and expression of atrophy-related genes in C2C12 myotubes. We also investigated the effects of the ethanol extract of Mori Folium (EEMF) on the recovery of AICAR-induced muscle atrophy in C2C12 myotubes. It was found that exposure to AICAR resulted in the stimulation of Forkhead box O3a (FOXO3a); an up-regulation of muscle-specific ubiquitin ligases such as Muscle Atrophy F-box (MAFbx)/atrogin-1 and muscle RING finger-1 (MuRF1), and a down-regulation of muscle-specific transcription factors, such as MyoD and myogenin; with the activation of AMPK. In addition, AICAR without cytotoxicity indicated a decrease in diameter of C2C12 myotubes. However, treatment with EEMF significantly suppressed AICAR-induced muscle atrophy of C2C12 myotubes in a dose-dependent manner as confirmed by a decrease in myotube diameter, which is associated with a reversed stimulation of FOXO3a by the inhibition of AMPK activation. These results indicate that the activation of AMPK by AICAR induces muscle atrophy, and EEMF has preeminent effects on the inhibition of AICAR-induced muscle atrophy through the AMPK signaling pathway.

따라서 본 연구에서는 상엽 에탄올 추출물(ethanol extracts of Mori Folium, EEMF)이 유발하는 근위축 억제 효능 및 기전 해석을 실시하였다. 이를 위하여 먼저 분화가 유발된 C2C12 myotubes에서 에너지 조절 효소인 AMPK의 약리학적 활성제로서 세포에서 AMP/ATP 비율을 증가시키고 효소 활성화에 필요한 높은 에너지 소비 조건을 모방하는 AMP 유사체인 5-aminoimidazole-4-carboxamide ribonucleotide (AICAR)를이용한 인위적인 근위축 모델의 확립을 실시하였고, EEMF가 인위적으로 유발된 근위축 과정에 어떠한 영향을 미치는지를 조사하였으며, 이때 AMPK가 어떠한 영향을 미치는 지를 확인하였다.
제안 방법
근위축을 유발하기 위하여 사용된 AICAR가 C2C12 myoblasts의 생존율 및 성장에 미치는 영향을 확인하기 위하여 다양한 농도의 AICAR를 24시간 동안 처리한 후 trypan blue 염색 및 MTT assay를 이용하여 조사하였다. 먼저 trypan blue 염색을 실시한 후 hemocytometer를 이용하여 살아있는 세포의 수를 계수하여 비교한 결과 Fig.
AMPK가 활성화되면 FoxO3a를 경유하여 골격근에서 특이적으로 발현 되는 muscle-specific ubiquitin ligases로서 F-box type E3 ligase인 MAFbx/atrogin-1 및 Ring Finger 유형의 E3 ligase인 MuRF1의 증가를 통한 ubiquitin-proteasome 경로의 활성화를 유발함으로서 myogenic differentiation에 관여하는 muscle-specific transcription factors인 MyoD 및 myogenin의 분해를 촉진하여 근위축을 유발시키는 것으로 보고되고 있다[1, 7, 13]. 따라서 C2C12 myoblasts에서 AICAR 처리에 따른 AMPK 및 근위축 관련 유전자의 발현 변화를 확인하였다. 먼저 Fig.

대상 데이터
실험에 사용된 상엽(전북, 임실)은 대한생약식품(주)에서 구입하였으며, 상엽 에탄올 추출물(ethanol extract of Mori Folium, EEMF)을 얻기 위하여 상엽 100 g 당 100% 에탄올 1 l를 첨가하여 60℃, 150 rpm으로 3일간 교반한 후 이를 3,000 rpm에서 20분간 원심분리하여 상층액만 분리하였다. 분리된 상층액을 Whatman 필터(No.
실험에 사용된 세포주인 C2C12 myoblasts는 American Type Culture Collection (Manassa, VA, USA)에서 구입하여 사용하였으며, 90% DMEM, 10% FBS 및 100 unit/ml PS가포함된 성장배지를 사용하여 37℃, 5% CO₂조건 하에서 배양하였다. C2C12 myoblasts를 myotubes로 분화시키기 위하여 100% confluent 상태의 세포를 대상으로 90% DMEM, 2% HS 및 100 unit/ml PS가 포함된 분화배지를 매 2일마다 교체하였으며, 근위축 유도를 위하여 적정농도의 AICAR를 24시간 동안 처리하였다.
데이터처리
모든 실험 결과들의 유의성을 검정하기 위하여 분산분석(ANOVA)을 실시한 후 p<0.05 수준에서 Duncan's multiple range tests를 실시하였으며, 그 결과는 평균(mean) ± 표준편차(standard deviation, SD)로 표시하였다.
이론/모형
05% trypsin-ethylene diamine tetraacetic acid (EDTA)를 처리하여 세포를 부유시킨 다음 2,000 rpm으로 5분간 원심분리하여 상층액을 제거하였다. 이렇게 모인 세포를 대상으로 각각 1 ml의 PBS와 0.5% trypan blue solution (Gibco BRL, Grand Island, NY, USA)을 첨가하여 2분간 염색을 실시하고 hemocytometer에 적용시킨 다음 도립 현미경을 이용하여 살아있는 세포를 계수하였다.
성능/효과
7B에서 나타난 바와 같이 AICAR 처리에 의하여 증가된 AMPK 및 ACC의 인산화가 EEMF에 의하여 억제되는 것으로 나타났으며, FoxO3a의 발현도 억제하는 것으로 조사되었다. 또한 EEMF는 AICAR 처리에 의하여 증가된 MAFbx/atrogin-1 및 MuRF1의 발현을 감소시켰으며, 감소된 MyoD 및 myogenin
의 발현을 회복시키는 것으로 나타났다. 이상의 결과를 살펴볼 때 AICAR 처리에 의하여 유발된 근위축을 EEMF가 억제한다는 것을 알 수 있었으며, 이러한 현상은 AMPK 활성 억제를 통한 전사인자인 FoxO3a를 억제함으로서 유발되는 ubiquitin-proteasome 경로의 차단이 중요한 역할을 하는 것으로 생각된다.
