제안 방법

또한, LPS 1 μg/ml으로 활성화된 Raw 264.7 대식세포에 독성이 나타나지 않는 범위의 홍국 추출물 100, 200, 400, 800 μg/ml 농도로 각각 처리하여, 세포 독성을 확인하였다.

Raw 264.7 cell은 American Type Culture Collection (ATCC, Manassas, VA, USA)에서 분양받았으며, 10% fetal bovine serum (FBS) (Welgene Inc., Gyeongsan. Gyeongbuk, Korea)과 1 X antibiotic-antimycotic solution (Welgene Inc., Gyeongsan. Gyeongbuk, Korea)을 함유한 Dulbecco’s modified Eagle medium (DMEM) (Welgene Inc., Gyeongsan. Gyeongbuk, Korea)을 사용하였으며, 37℃와 5% CO2를 유지하는 배양기에서 배양하였다.
데이터처리
평균은 SPSS (SPSS Inc, Armonk, NY, USA)를 이용한 비모수적 Kruskal-Wallis 및 Mann-Whitney 분석을 사용하여 비교하였고 유의성은 p<0.01 및 p<0.05에서 검사하였다.
모든 실험은 최소 3회 반복하였고 데이터는 평균±표준편차 (S.D.)로 표현하였다.

성능/효과
홍국 추출물 내의 총 폴리페놀 함량은 800 μg/ml 농도에서 258.4 μg/g으로 확인되었고, 발효하지 않은 쌀은 100.1 μg/g으로 확인되어 발효하 지 않은 쌀 대비해 1.5배 이상 높은 결과를 확인했다(Fig. 1, Table 1).

NO 생성량은 홍국 에탄올 추출물의 농도에 따라 감소되었고, 400 μg/ml 농도에서는 13.28 μM의 NO 생성량이 확인되어, LPS 처리에 의한 NO 생성을 LPS를 처리하지 않은 음성대조군에서 생성되는 NO (9.38 μM) 수준까지 감소시키는 저해효과를 나타내었다(p<0.05).
홍국의 NO 생성 저해 활성을 확인하기 위해 Raw 264.7 대식 세포에 홍구 추출물 100, 200, 400, 800 μg/ml로 처리 후 LPS로 염증을 유발한 결과 모든 농도에서 NO 생성 억제효과가 나타났다(Fig. 4).
