녹색꽃양배추의 화뢰 성숙단계 및 부위에 따른 Sulforaphane과 항산화 물질 함량 비교 Comparison of Sulforaphane and Antioxidant Contents according to Different Parts and Maturity of Broccoli
본 연구는 브로콜리 화뢰의 성숙단계 및 부위별 sulforaphane과 항상화 물질 등을 분석하여 비교하고자 하였다. 브로콜리의 기능성 성분인 sulforaphane은 GC/MS, ascorbic acid는 HPLC, chlorophyll과 β-carotene의 함량은 UV-spectrophotometer에 의해 분석되었다. 브로콜리 화뢰의 성숙단계에 따른 sulforaphane 함량을 조사한 결과 ‘녹제’와 ‘그레이스’ 모두 적숙기에서 sulforaphane의 함량이 가장 높게 나타났으며, ‘녹제’가 ‘그레이스’에 비해 다소 높은 함량을 나타내었다. 한편 ascorbic acid 함량을 조사한 결과 ‘녹제’와 ‘그레이스’ 모두 과숙기 화뢰가 적숙기보다 높은 함량을 나타냈고 미숙기에서는 두 품종 모두 ascorbic acid가 검출되지 않았다. β-carotene의 함량은 두 품종 모두 적숙기에서 함량이 가장 높았고, 미숙기가 가장 낮았다. 품종별 차이는 ‘녹제’가 ‘그레이스’보다 더 높은 함량을 나타내었다. The contents of sulforaphane and other antioxidants were estimated by GC/MS, HPLC and UV-spectrophotometer. Among the cultivars tested ‘Nokjae’ contained higher sulforaphane level than ‘Grace’ and its content varied depending on different maturity stages of broccoli; immature, mature and overmature stages. In the case of ‘Nokjae’, the highest sulforaphane level was detected at mature stage as compared to the other two stages. The ascorbic acid contents in both cultivars were found to be higher in the overmature stage than in either immature or mature stage. However, β-carotene contents were not in accordance with the maturity stages, its highest level was detected at the mature stage in both cultivars, whereas the lowest at the immature stage. Chlorophyll contents were also varied depending on the maturity stages. The highest level was detected at the overmature stage in ‘Nokjae’ and at the mature stage in ‘Grace’, while its lowest level was observed at the immature stage in both cultivars. Among the three parts of broccoli tested, florets of ‘Nokjae’ contained the highest level of sulforaphane, whereas leaf exhibited a very low level. The stalk of broccoli had the highest level of ascorbic acid; in contrast, the leaf contained high level of β-carotene and chlorophyll. In summary, our data showed that sulforaphane contents of broccoli were the highest in the florets at the mature stage, whereas ascorbic acid, β-carotene and chlorophyll levels were higher in leaves and stalks of broccoli.
