본 연구과제는 중추신경계의 catecholamine 신경기능과 그 영향을 받는 시상하부-뇌하수체의 내분비축에 있어서 호르몬 유전자 구조의 분석, 신호전달기작 나아가서 조직 특이성 유전자 발현의 조절 기작을 밝히고자 수행되었다.

본 연구과제는 중추신경계의 catecholamine 신경기능과 그 영향을 받는 시상하부-뇌하수체의 내분비축에 있어서 호르몬 유전자 구조의 분석, 신호전달기작 나아가서 조직 특이성 유전자 발현의 조절 기작을 밝히고자 수행되었다.
1. Epinephrine 합성 효소인 phenylethanolamine N-methyltransferase (PNMT)의 흰쥐 전체 유전자를 성공적으로 클로닝하고 그 전체 염기서열을 완전히 분석하였다. 흰쥐 PNMT 유전자의 Promoter 부위에는 전형적인 glucocorticoid response element(GRE), SP1, TATA box 등이 존재하며, GRE는 glucocorticoid에 의해서 예민하게 반응하였다. 대표적인 향정신약물인 reserpine을 사용하여 catecholamie의 첫번째 합성효소이며 속도제한 효소인 tyrosine hydroxylase 와 PNMT 효소의 발현차이와 중추신경계와 말초신경계의 발현차이를 연구하였다. Tyrosine hydroxylase는 reserpine에 의해 중추와 말초조직 모두에서 활성과 유전자 발현이 증가하였다. PNMT는 말초에서 활성과 유전자 발현이 증가하였고 뇌저에서는 활성은 증가한 반면, 유전자 발현은 감소하였다. 이로 미루어 볼때 PNMT는 중추와 말초에서 조직 특이한 유전자 발현조절이 있는 것으로 사료된다.
2. Catecholamine과 같은 중추신경적인 영향과 뇌로 feedback한 호르몬의 영향에 의해서 조절되는 시상하부 GnRH 뉴우론의 유전자 발현 조절에 관하여 연구하여 다음과 같은 결과를 얻었다.
1) Estrogen과 testosterone은 농도과 시간 의존적으로 LH의 분비와 합성에 억제적 작용을 하면 반면에 이들 steroid는 GnRH 함량과 유전자 발현에는 커다란 영향을 주지 않았다. 그리고 난소를 절제하고 estrogen이 사전처리된 상황에서 progesterone은 GnRH 유전자 발현을 촉진하였다. GnRH 뉴우론에는 steroid 호르몬의 수용체가 결합하는 부위가 없는 것으로 보아 progesterone의 촉진적 효과는 GnRH 뉴우론을 둘러싼 여러 신경전달의 network으로 조절된다고 사료된다.
2)Catecholamine을 신경말단에서 고갈시키는 6-hydroxydopamine(6-OHDA)을 in vitro 배양중인 시상하부 조직에 처리하면 대조군에 비해서 GnRH mRNA는 약 60%정도 감소되었다. 또한 catecholamine을 보다 선택적으로 차단하는 DSP로 비슷한 영향을 보였다. 난소절제 후 estrogen과 progensterone을 처리하여 증가된 시상하부의 GnRH 유전자 발현은 dopamine-β-hydroxylase(DBH)의 활성차단제인 DDC에 의해서도 유의적으로 감소하였다. 따라서 in vitro와 in vivo 실험조건에서 catecholamine 특히 norepinephrine이 GnRH 유전자 발현에 중요한 영향을 미치고 있음을 시사한다.
3) 흥분숭 아미노산 신경전달에 의한 GnRH 유전자 발현에 관한 영향을 조사하기 위해서 NMDA 수용체의 길항제인 MK-801과 non-NMDA 수용체의 길항제인 CNQX를 난소절제후 estrogen과 progesterone을 처리한 실험모델에 주사한 결과, MK-801(0.2 ㎎/㎏)은 대조군에 비해 유의적으로 GnRH mRNA를 감소시킨 반면에 CNQX는 거의 영향이 없었다. 또한 MNDA 처리에 의한 GnRH 유전자 발현 양상을 NMDA 처리한 후 30분에선 60분 사이에 높은 증가를 보이다가 90분 이후에는 감소하였다. 이와같은 NMDA의 증가효과는 α-1 아드레너직 수용체의 길항체인 prazosin 처리에 의해서 억제됨으로 보아 GnRH 유전자 발현에 미치는 NMDA와 같은 흥분성 아미노산 신경입력은 catecholamine을 매개하여 일어나는 것으로 사료된다. 억제성 아미노산인 GABA 신경입력도 GnRH 유전자 발현조절에 영향을 미치며, GABA-A 수용체 활성제인 muscimol과 GABA-B 1 수용체 활성제인 baclofen은 모두 농도의존적으로 혈청내 LH수준을 감소시켰으나 예상과는 달리 muscimol은 낮은 농도에서 GnRH mRNA를 대조군에 비해 약 60%정도 증가시켰고, baclofen의 효과는 2-3배 정도 증가시켰다. 이 결과는 예상치 못한 결과이며 억제성 신경입력으로 알려진 GABA는 GnRH 뉴론과 LH 분비에 아미도 차등적인 역할을 할 것으로 추정된다.
4) 세포내 신호전달기작과 GnRH 유전자 발현의 연계성을 조사하기 위하여 protein kinasea(PKa) 활성제인 forskolin과 PKc 활성제인 PMA를 배양중인 시상하부 조직에 처리하였다. Forskolin과 PMA는 농도의존적으로 GnRH mRNA를 증가시켰으며 forskolin과 PMA 동시처리하였을 때 누적효과는 관찰되지 않았다. 또한 forskolin과 PMA는 다양한 시상하부 단백질의 인산화 양상에도 영향을 미쳤다. 이 결과는 PKa와 PKc의 신호전달체계의 활성화가 특정 단백질이 인산화 혹은 탈인산화를 통하여 궁극적으로 핵내에 영향을 미쳐 GnRH 유전자 발현에 영향을 미침을 강력히 시사한다.
3. 시상하부의 영향을 직접적으로 받는 뇌하수에서 합성, 분비되는 황체형성호르몬(LH) β subunit의 생합성조절기작을 유전자 발현 수준에서 규명하기 위한 연구의 일환으로 뇌하수체 전엽 배양세포에서 GnRH가 LHβ subunit의 유전자 발현에 미치는 영향, 난소호르몬이 LHβ subunit의 유전자 발현에 미치는 영향, 뇌하수체전엽 배양세포에서 GnRH와 난소호르몬의 작용 기작을 조사하여 다음과 같은 결과를 얻었다.
1) 흰쥐의 뇌하수체 전엽 배양세포에서 LH의 분비가 GnRH 투여 종도에 비례하여 증가하였으나, α subunit mRNA의 농도는 증가하지 않았고, LHβ subunit mRNA의 농도는 GnRH 농도에 비례하여 증가하였다. 이는 LH의 분비와 합성이 GnRH에 의해서 조절받고 있음을 시사한다. 또한 LHβ subunit mRNA의 농도는 estradiol과 progesterone을 단독으로 또한 GnRH와 동시에 투여하였을 때 이들 난소호르몬에 의존적으로 증가하였으며, estradiol이 progesterone보다 LHβ subunit mRNA 의 농도를 더욱 증가시켰다. 이는 난소 호르몬이 뇌하수체에 직접 작용하여 LHβ유전자 발현을 조절함을 시사한다.
2) 뇌하수체전엽 배양세포에서 GnRH가 LHβ subunit의 유전자 발현을 조절할 때, 어떠한 세포내 신호전달 경로를 경유하는 지를 조사한 결과, PKc와 cAMP 의존성 PKa의 신호전달경로가 공히 관여되어 있음이 관찰되었다. 뇌하수체 전엽 배양세포에 PKc 활성제인 PMA와 adenylate cyclase 활성제인 forskolin에 의해 증가한 LHβ subunit mRNA 수준은 전사억제제인 actinomycin D 처리로 억제되었다. 또한 calcium chealator인 EGTA와 channel blocker인 verapamil은 GnRH, PMA, forskolin에 의한 LH 분비를 선택적으로 억제한 반면, LHβ subunit mRNA 증가를 모두 억제하였다. Estradiol과 progesterone은 LHβ subunit mRNA decay를 억제하여 stability를 증가 시켰으나 PMA나 forskolin은 이러한 decay 정도에 영향을 미치지 못하였다.
3) 뇌하수체전엽 배양세포에 estradiol을 투여하였을 때 GnRH 수용체의 수가 유의하게 증가하였으나, progesterone 투여는 증가시키지 못했다. 또한 estradiol과 progesterone을 투여 하였을 때 농도의존적으로 뇌하수체 전엽세포에서 세포질 분획의 PKc 활성도가 유의하게 증가 하였다. 이상의 결과로 보아 GnRH는 LH의 분비와 마찬기지로 LHβ subunit의 생합성을 전이전 단계에서 조절하는 가장 중요한 인자로서 PKc를 활성화하고 cAMP를 매개하여 작용을 나타내는 난소호르몬은 뇌하수체에 직접 작용하여 LH의 분비와 LHβ subunit의 유전자 발현을 조절하며, 특히 estradiol은 부분적으로 GnRH 수용체의 수를 증가시키고 세포질분획의 PKc를 활성화하여 성선자극호르몬 분비세포에서 GnRH의 작용을 조절하는 것으로 생각된다. 또한 estradiol과 progesterone은 LHβ subunit mRNA stability를 조절하는 중요한 요인으로 생각된다.
4. LH를 비롯한 FSH, TSH 등 뇌하수체 당단백질 호르몬 유전자의 발현조절에 관한 기작을 이해하기 위하여 여러 각도로 연구를 수행하였다. Transfection 실험을 간편히 행할 수 있도록 각종 벡터를 개발하였으며 이를 이용하여 당단백질 호르몬 subunit 유전자들이 조직특이적으로 또한 여러 조절인자에 의해 발현되기 위해 필요한 DNA 서열을 각 유전자의 5'-flanking 지역에서 확인하였다. 이와 함께 당단백질 호르몬을 각 유전자의 5'-flanking 지역에서 확인하였다. 이와 함께 당단백질 호르몬 유전자를 발현하는 세포주를 개발하기 위해 여러가지 방법으로 시도하였으나 이는 여의치 않았다. 한편, 유전자의 5'-flanking 지역에 결합하여 발현조절에 관여하는 trans-acting factor를 찾기 위하여 핵추출물내의 단백질이 당단백질 호르몬 유전자와 결합하는 부위를 확인하였으나 이같은 DNA-단백질 결합반응의 역항에 대해서는 결론을 내릴 수 없었다. 다만 FSHβ subunit 유전자의 경우, estrogen 호르몬 수용체가 이 유전자의 5'-flanking 지역에 결합함을 확인하였으며, 또 이에 의해 유전자의 발현이 촉진될 수 있음을 transfection 실험을 통해 증명하였다. 결론적으로 본 연구는 신경 및 내분비계의 유전자 발현의 상호작용을 체계적이고 종합적으로 조사하였으며, 세포 및 분자수준에서 조절기작을 해결하는데 기여하였다고 사료된다.

The present study aims to determine the regulatory elements of neuronal and hormone gene and to elucidate the possible action mechanism underlying underlying tissue-specific gene expression in a variety of experimental setting. 1. To obtain information on the genomic structure and regulation of the gene encoding epinephrine synthesizing exzyme. phenylethanolamine-N-methyltransferase(PNMT), which palys a pivotal role in expression of adrenergic phenotypes, bovine PNMT cDNA was used as a prove to screen a rat genomic library constructed in EMBL3 replacement vector at BamHI sites. About 10? phage plaques were examined and one clone showing strong hybridization signals was isolated by three repetitive screening procedures. The recombinant DNA purified from this rat genomic clone hybridized to bot 5' and 3' end specific bovine PNMT cDNA probes. Complete nucleotide sequence analysis of a genomic clone showed that this gene is splitted into 3 exons with about 2.0 kb in length. The structure of exons of the rat PNMT gene was similar to those of the human and bovine PNMT genes, but the structure of promoter and introns were quite different. There were TATA box, GRE and SP1 sequences in the promoter of the rat PNMT gene. The GRE sequences are highly inducible by the treatment of dexamethasone in the primary chromaffin cell culture. Tyrosine hydroxylase(TH) and PNMT are differentially regulated in the contral nervous and peripheral adrenal tissues by reserpine. 2. The hypothalamic GnRH neurons are known to be influenced by central input like streoid hormone. The molecular regulation of GnRH gene expression and the signal transduction mechanism of GnRH neurons are however poorly elucidated. 1) In vivo administration of estrogen and testosterone induced an inhibitory effects on LH secttetion and a and LHβ gene expression in a dose- and time- related manner without significant altering GnRH mRNA levels, However, a single injection of progesterone to ovariectomized estrogen-primed rats significantly increased GnRH gene expression. Since there is no steroid receptors in GnRH neurons and more importantly there is no steroid-response element(SRE) in the GnRH promoter region, it is quite resonable to presume that steroid, in particular progesterone may indirectly affect GnRH gene expression presumably through some other neuronal network in a vincinity of the GnRH neurons. 2) To elucidate whether catecholaminergic neurotransmission may affect GnRH gene expression, 6-hydroxydopamine (6-OHDA) was tested with superfused hyphthalamic tissues in vitro. Treatment with 6-OHDA significantly reduced GnRH mRNA levels. And treatment with DSP-4 or DDC, wich are known neurotoxins blocking catecholaminergic neurotransmission, was also effective in decreasing GnRH mRNA levels. These data indicate that caecholamine, in particular, norepinephrine may play an important role in transsynaptic regulation of GnRH gene expression. 3) To determine whether NMDA neurotrnasmission is involved in the regulation of GnRH gene expression, MK-801, a NMDA receptor antagonist, and CNQX, a non-NMDA receptor antagonist, were tested using ovariectomized, estrogen and progesterone treated rats. The inhibition of NMDA with MK-801, but not with CNQX, significantly reduced GnRH mRNA levels. Moreover, treatment with NMDA increased GnRH mRNA within 30-60 min and thereafter returned to the basal level. This time-dependent increase in GnRH mRNA levels by NMDA was blocked by a-1 adrenergic antagonist such as prazosin. It appears then that the NMDA-induced increase in GnRH gene expression occurs through catecholaminergic mediation. A known inhibitory neurotransmitter, GABA is also involved in the regulaton of GnRH gene expression. Intraventricular microinjection of muscimol, a GABA A-type agonist, increased GnRH mRNA levels, whereas that of faclofen, a GABA B-type agonist, significantly enhanced GnRH mRNA levels. This unexpected result remains to be resolved, but GABA may differentially regulate GnRH and Lh synthesis. 4) To examine whether the activation of protein kinase (PK) pathways, such as PKa and/or PKc is involved in GnRH gene expression, forskolin, an activator of PKa pathway and PMA, an activator of PKc pathway, were tested. Both agents stimulated GnRH mRNA levels in a dose-related manner, but these is no additive effitive effects of two agents. Moreover, the activation of signal transduction pathways with forskolin and PMA altered the protein phosphorylation pattern. Then it appears that the activation of signal transduction pathways affects phosphorylation (or dephosphorylation) of cytoplasmic (or nuclear) protein(s), and the extracellular influence eventually ends up to the nucleus which regulates GnRH gene expression. 3. Pituitary LH release is known to be regulated by the hypothalamic GnRH and gonadal steroid hormones. The alteration in LH release might reflect changes in biosynthesis and/or posttranslational procession. Little is, however, known about the regulatory mechanism of their gene expression and posttranslational processing. The present study examined the possible mechanism by which LH subunit gene expression is regulated, the effect of ovarian steroids and GnRH on LH subunit mRNA synthesis as well as LH release, and intracellular signal pathway by which GnRH exerts its effect. The results obtained are as follows: 1) GnRH stimulated LH release from cultured pituitary cells in a dose-dependent manner. The treatment of pituitary cells with GnRH increased LHβ subunit mRNA level in a dose-dependent manner, reaching the maximum(2.7 fold) with 2x10???M GnRH, while no increase was observed in α subunit mRNA level after GnRH treatment. It suggests that GnRH plays a major role in Lh release and LH subunit synthesis by influencing the steady state mRNA level of LHβ subunit. The treatment of pituitary cultured cells with estradiol did not modulate the GnRH-induced LH release, while progesterone augmented the GnRH-induced LH release in a dose-dependent manner. However, the stimulatory effect of progesterone was significantly reduced by its antagonist, RU486. Estradiol and progesterone stimulated LHβ mRNA levels in the presence or absence of GnRH(2x10???M). Moreover, effects of estradiol and progesterone were diminished by the treatment of estrogen antagonist, LY117018 and progesterone antagonist, RU486, respectively. It is therefore suggested that ovarian steroids directly modulate LH synthesis and release at the pituitary level. 2) Intracellular signal pathway by which GnRH exerts its effect on LHβ subunit systnesis and release was investigated. The treatment of pituitary cells with GnRH increased particulated protein kinase (PK) C activity as well as intracellular cAMP consentration in anterior pituitary cell culture. The treatment of pituitary cultured cells with PMA or forskolin increased the steady state level of LHβ subunit mRNA and LH release. Actinomycin D, a transcription inhibitor, suppressed LHβ subunit mRNA level which increased by the treatment with PMA or forskolin. In other hands, the treatment with EDTA and verapamil suppressed LHβ subunit mRNA level which was induced by the treatment of PMA or folskolin, but showed a selective inhibition of LH release by different doses and durations of PMA treatment. Estradiol and progesterone showed suppression of decay of LHβ subunit mRNA, but PMA or forskolin did not. 3) Estradiol significantly increased GnRH binding to its receptor.
