Sfs 유전자의 전체 염기배열을 결정하여 해석해 본 겨로가 MluI-PstI의 2126bp 중에는 2개의 PRF가 존재 하였다. ORF1은 sfs 유전자를 coding 하고 있으며, ORF2는 truncated protein 을 coding 하고 있었다. 조절영역 DNA 의 Promoter 로 예상되는 상류의 영역에 CRP binding site 가 존재 하였으며, cAMP-CRP complex 에 의하여 sfs 유전자의 발현이 촉진 되어짐을 sfs-lacZ 융합 유전자를 작성하여 Western blotting 으로서 확인 하였다.

In Escherichia coli, CRP formms a complex with cAMP and acts as a transcriptional regulator of many genes, including sugar metabolism operons. The E. coli MK2001, Which is introduced the altered crp*1, is functional in the expression of lac, ara, and man, in lthe absence of cAMP. However, the expression of mal gene is fully activated by the addition of cAMP or cGMP. The object of this study i sloning of the sfs genes, which is invovled in regulation of mal gene expression with the altered crp*1 gene and structural analysis and characterization of the gene at the molecular level. The results are as follows: The author has cloned 5 different E. coli genes which stimulate the maltose metabolism in a crp*1, cya: : km (MK 2001) background. Mewly identified genes were designated as sfs. One of the sfs genes (pPD 26), located at the 53.2 min. map position on the E. coli chromosome, was further analyzed. Expression of the genes, which is involved in maltose metabolism, malQ and malP, increased to 5.8-fold in the presence of a plasmid, pCMA 27, containing the subcloned sfs gene. The nucleotide sequence of a common 2126 bp segment of the pCM1 was determined and two open reading frames (ORF1 and ORF2) were detected. The ORF1 encoded the sfs gene and ORF2 encoded a truncated protein. Potential CRP binding site is located in the upstream of the putative promoter in the regulatory region. Expression of the cloned sfs gene was positively regulated by the cAMP-CRP complex.
