최근 김치의 공장 생산이 크게 증가하는 등 김치의 산업화가 진행됨에 따라 과학적인 생산, 가공, 보존을 위한 연구가 활발히 이루어 지고 있으나, 아직까지 김치 제조 중 절임 과정에서 발생하는 절임 폐수의 처리에 관한 연구는 전무한 실정이다. 그러므로, 본 연구에서는 김치 공장에서 다량 발생되는 절임 폐수를 이용하여 단세포단백질로 사용할 수 있는 효모균체를 생산하고, 동시에 절임 폐수의 유기물을 제거하여 환경오염을 줄이는 방법을 연구하였다.김치공장에서 발생되는 절임 폐수를 미생물 배지로 사용하기 위하여 그 성분을 분석한결과
최근 김치의 공장 생산이 크게 증가하는 등 김치의 산업화가 진행됨에 따라 과학적인 생산, 가공, 보존을 위한 연구가 활발히 이루어 지고 있으나, 아직까지 김치 제조 중 절임 과정에서 발생하는 절임 폐수의 처리에 관한 연구는 전무한 실정이다. 그러므로, 본 연구에서는 김치 공장에서 다량 발생되는 절임 폐수를 이용하여 단세포단백질로 사용할 수 있는 효모균체를 생산하고, 동시에 절임 폐수의 유기물을 제거하여 환경오염을 줄이는 방법을 연구하였다.김치공장에서 발생되는 절임 폐수를 미생물 배지로 사용하기 위하여 그 성분을 분석한결과, pH가 5-6 범위였으며, 염농도는 7∼12%로 매우 높았고, 환원당은 약 0.5∼1.0g/ℓ 였으며, BOD는 약 1,100mg/ℓ, COD는 1,300∼1,800mg/ℓ인 것으로 조사되었다. 또한, 절임폐수의 성분은 공장에 따라 차이가 큰 것으로 나타났다.절임 폐수에서 생육이 우수한 균주를 선발하기 위하여 절임 폐수, 토양, 장류 등에서분리한 효모와 내염성 균주로 알려진 효모 등을 배양하여 가장 높은 생육을 나타낸 Pichiaguilliermondii A9을 선발하였다. 이 효모는 절임 폐수에서 배양시 염농도가 10%까지는생육에 영향을 받지 않았으며, 초기 pH 3∼6의 범위에서 영향을 받지 않고 생육하였고, 최적 생육온도는 30℃인 것으로 나타났다. 절임 폐수에 Ammonium sulfate, potassiumphosphate 및 미량 원소의 첨가는 생육 증진 효과를 나타내지 않았다. 발효조(조업부피 1ℓ)에서 P. guilliermondii A9를 배양한 결과, 300 rpm의 교반 속도에서 0.5vvm의 통기량으로도 최고 균체량을 얻을 수 있었다. 이 효모 균체의 성분 분석 결과 단백질이 40%, 지질은 약 12%를 함유하였다. 한편, P.guilliermondii A9는 절임 폐수에서 배양시 24시간 이내에 BOD를 약 90% 이상 제거하였다.균체 생산을 증가시키기 위하여 김치 공장에서 배추를 다듬는 과정에서 발생되는 배추쓰레기의 착즙액을 절임 폐수에 첨가한 결과 균체 생산량이 크게 증가하여, 30% 첨가시에3.4g/ℓ의 건조 균체를 얻을 수 있었으며, 이는 절임 폐수만을 사용했을 때 보다 약 5배가증가한 것이었다.따라서, 김치 공장에서 다량 발생되어 문제가 되는 절임 폐수는 내염성 효모인 P.guilliermondii A9을 배양하여 단세포 단백질로 사용할 수 있는 균체를 생산하므로써 효과적으로 자원화 할 수 있으며, 동시에 환경오염도 줄일 수 있었다. 또한, 김치공장에서 발생되어 문제가 되는 배추쓰레기를 착즙하여 절임 폐수에 첨가함으로써 균체 생산을 크게 증가시킬 수 있었으므로 이는 김치공장에서 생산되는 서로 다른 두종류의 폐기물을 동시에 처리할 수 있는 효과적인 유기성 폐기물의 자원화 방안으로서 실용화 할 수 있을 것으로 기대된다.

In recent years, there has been considerable interest inresearching several aspects of kimchi with respect to the industrial production.However, no studies have been carried out on the treatment of the large quantities ofwaste brine that are produced in a kimchi factory and released into the environmentwithout any treatment. Therefore, we have selected yeast strain that showed thehighest growth in waste brine, and studied the growth conditions. This could bothmake possible the used of kimchi waste brine as a substrate for the production of singlecell protein, and remove potential pollutants from the waste water.The analysis of the waste brine samples obtained from the different kimchifactories showed that the pH ranged from 5 to 6; and the NaCl concentration wasbetween 7 and 12%(w/v). The reducing sugar content was 0.5g/ℓ to 1.0g/ℓ. BODof the samples was about 1,100 mg/ℓ and COD ranged from 1300mg/ℓ to 1800 mg/ℓ.Pichia guilliermondii A9 was selected from 70 isolates of yeast demonstratingsubstantial growth in waste brine. During the culture of Pichia guilliermondii A9 inwaste brine, BOD reduced rapidly to one tenth of the original load.The growth of Pichia guilliermondii A9 was not inhibited by NaCl concentration ofup to 10%. However, it was reduced drastically at concentrations greater than 12%.The optimum temperature for growth was 30℃. The pH of the waste brine onlyslightly affected growth in the range between pH 3 and 6. The addition of ammonium,potassium and various trace elements did not enhance the cell growth. When Pichiaguilliermondii A9 was cultured in waste brine using the fermenter with a workingvolume of 1ℓ, the cell mass reached a constant maximum values at the air flow ratesof above 0.5vvm and at the impeller speed of 300 rpm. The yeast cells cultured inwaste brine contain 40% protein and 12% lipids.When the waste brine was enriched with cabbage juice from waste cabbage, thefinal cell mass increased proportionally with the amount of added organic material,reaching 3.4 dry cell g/ℓ with 30% of cabbage juice.In conclusion, the large amount of waste brine generated from kimchi productioncould be used directly for the culture of the osmotolerant yeast. The use ofsupplemental nutrients, contained in the juice from the vegetable waste, effectivelyincreases the production of cell mass. Therefore, these results suggest an interestingpractical application for the treatment of two different types of waste produced duringkimchi manufacturing, and useful for the process of yeast culture.
