본 연구에서는 functional genomics(cDNA microarray)를 이용하여, 2003년 시행한 선행 연구(유전재학 기법을 이용한 급성 간 독성 유형별 유전자 발현, TRC 사업, 식품의약청)를 통해 1) 대표적인 유형별 독성 물질들을 대상으로 만성 간 손상에서 유전자 발현 변화를 대규모로 분석, 2) Bioinformatics를 활용한 유전자 발현 자료의 분석과 생물학적 검증 실험을 통하여 간 독성 유형 특이적 분자 생체 지표 후보 물질을 개발한다. 특히 이들 간 독성 물질의 미량의 지속적인 노출에 의한 간 독성 증상 유무기 및 경제기에 있어 특이적인 유전자를 구별할 수 있는 생체 지표 후보 물질을 개발 3) 도출된 생체지표 후보물질과 조직 병리학적 및 생화학적 지표와 상관성을 분석을 통하여 간 독성 유형과 증상 유무기, 독성 물질의 노출, 간 독성 병변의 진행을 예측 가능한 특이적인 생체지표 시스템을 확립 4) 개발된 생체지표 유전자를 조합하여 특이성, 민감성이 우수하며, 비용이 저렴한 간 독성 유전자 chips의 개발을 위한 유용한 자료를 제공 5) 본 실험에서 얻어진 모든 재료는 데이터베이스로 구축하여 공공기관의 web을 통해 관련 연구자들에게 공개하여 우리나라 독성학 연구의 발전에 기여함이 그 목적이었다.
이러한 연구 목적을 위해, 수컷 Sprague Dawley 랫에, 각각 간 독성 유형의 대표적 2가지 독성물질을 미량으로 만성 노출 시켰으며 유전자 발현을 cDNA array로 조사하였다. 2가지 간세포 독성물질로부터 나타나는 각각 유전자 발현을 패턴별로 hierarchial 및 K-mean clustering하여, 각 손상에 다른 유전자의 데이터베이스를 확보하였다.($CCl_4$: 0.5㎖/㎏: 380개 유전자, 4패턴 cluster. 0.25㎖/㎏: 384개 유전자, 4개로 cluster, Acetaminophen; 500㎎/㎏; 100개 유전자, 5개 패턴 cluster, 250㎎/㎏: 189개 유전자, 5개 cluster) $CCl_4$를 이용한 지방산 변화에 있어 무증상 기(30d-high, low) 및 경계기 (60dr-high, low), 증상 기 변화(90d-high, low)를 가지는 특이적인 유전자는 모두 704개, Acetaminophen을 이용한 간세포 괴사에 있어 특이적인 발현을 보이는 유전자는 378개였다. 또한 GeneMAPP을 이용하여 대사 및 신호전달 경로를 분석하여, 각각의 손상에 있어, 10여종의 새로운 생체대사 및 신호 경로를 발굴하였다. 본 연구의 데이터는 간 독성 유형에 따른 특이적 유전자 발현 및 특이적 패턴을 발굴하였으며, 더 나아가 각각의 간 독성 유형에 나타나는 병리조직학적 지표에 대해서 특이적이고도 광범위한 유전자 발현 패턴을 보여줌으로 간독성 toxicogenomic 연구에 새로운 연구 방법으로 유용성을 제시하고 있으며, 더 나아가 본 연구 데이터가 간독성 ″toxchip″을 구축에 유용한 자료로 생각된다.

Toxicogenomics, the study of changes of molecular biomarkers (DNA, RNA, protein and metabolites) following toxicant exposure, could deliver insight into potential human toxicity earlier in the drug development process. The DNA microarray is a prototype high throughput technology applied in the toxicogenomics and used to measure the mRNA expression of thousands of genes simultaneously, hold promise for future application in efficient screening of therapeutic drugs. And it has provided a revolutionary opportunity to monitor genome-wide gene expression.
Chemicals exposure of animals can result in an array of adverse pathological responses including hepatotoxicity, This study investigates gene expression and histopathological alterations in response to various hepatotoxins treatment in order to 1) construct computational data bases of gene expression derived from livers of two chemicals treated rats based on chronic exposure 2) to histopathologically anchor gene expressions global and specific patterns according to the each hepatotoxin, hepaototoxicants processing and exposure amount of hepatoxicant, and 3) to gam insight into mechanism(s) of hepatotoxicity, cDNA array was performed to get the large-scale differential gene expression from the liver tissues with chronic exposure(0. 30days, 60days and 90days) of two chemicals,($CCl_4$ for fatty change and early cirrhosis; acetaminophen for hepatic necrosis) to male Sprague-Dawley rats with high and low dosage of each chemicals, Hierarchical clustering and principal component analysis were taken in elassifying each sample of two chemicals in agreement with microscopic observations. Data from cDNA microarray analysis corroborated observed histopathological alterations such as hepatocellular necrosis, fatty change and cholestasis. These data has provided insight into the patterns of specific and global gene expression in the histological processes of each subtype of hepatotoxin and has indicated that the histopathologic approach in toxicogenomics may be useful for the development of ″tailor-made toxchip″.
