Transgenic mice에서의 목적 유전자는 mouse EC-SOD를 이용하였고, 발현 효율을 증진시키기 위해 rabbit $\beta$-globlin intron 2 염기서열과 chicken ovalbumin polyA를 transcription termination signal 염기서열을 삽입하여 이용하였다. 피부의 keratinocyte에서 특이적으로 발현하는 KL-14 promoter는 마우스 skin library로 부터 cloning하여 구축하였다. 2) EC-SOD 형질전환 마우스 생산 BABL/c 웅성마우스는 구입하여 사용하였으며, 형질전환동물의 생산은 21U의 PMSG와 hCG을 이용하여 과배란된 마우스로부터 1 세포기 수정란을 회수하고, effendorf사의 manuplation을 이용하여 웅성전핵에 약 $2p{\ell}$정도의 유전자를 도입한 후, 정관 결찰된 숫컷과 교배한 가임 신마우스의 난관에 이들 수정란을 이식하였다. 이식된 수정란을 기준으로 약 6% 즉 10마리의 형질전환 마우스가 생산되었다. 3) 외부자극에 의한 EC-SOD 과발현 마우스의 질병 저항성 연구 (1) 발암제처리에 따른 ROS 생성능력 규명 EC-SOD 과발현 마우스에 대한 발암제 처리는 TPA($4{\mu}g/14$ 주)를 이용하였다. TPA를 1 또는 2.5 nmol 농도로 처리하고, 24시간 후 마우스 피부조직을 채취하였다. 이를 10%의 formalin에 하룻밤 고정한 후, 8OH-dG staining 방법을 이용하여 ROS 생성정도를 탐색한 결과 EC-SOD 과발현 마우스에서 그 생성량이 감소함을 확인할 수 있었다. (2) 면역조직학적 연구 8주령 EC-SOD 과발현마우스와 정상마우스에 대한 TPA 유도 세포증식 정도를 관찰하였다. TPA를 처리하고 24시간 후 1ml BrdU를 주사하였다. BrdU 염색정도를 확인하기 위하여 Streptavidin Biotin System (Zymed laboratories, USA)을 이용하였다. TPA를 처리한 마우스를 비교하여 본 결과 0.2 nmol 처리군의 경우 대조군 마우스와 EC-SOD 과발현마우스간에 염색정도에 큰 차이가 확인되지 않았다. 하지만 0.4 nmol TPA 처리군에서는 EC-SOD 과발현마우스에 비해 대조군마우스에서 많은 세포증식이 확인되었다. 이러한 결과는 EC-SOD의 과발현이 TPA 유도 세포증식을 억제할 수 있는 것으로 판단할 수 있었다.

They function by efficiently catalyzing dismutation of two superoxide anions to hydrogen peroxide and oxygen. Mammalian systems contain three isoforms of SOD: Cu/Zn-SOD (SOD1) a cytoplasmic enzyme, Mn-SOD (SOD2) in mitochondria, and EC-SOD (SOD3) in the extracellular space, EC-SOD is one of the major tissue defense enzymes that protects against toxic effects of ROS. In the skin specifically, EC-SOD primarily distributed in the epidermis and dermis, but the role of EC-SOD in the skin is poorly known. In the present study, we developed a line of transgenic mouse expressing mouse EC-SOD in the skin to investigate whether EC-SOD suppresses tumor formation in the BMBA/TPA two stage skin carcinogenesis model. We demonstrated that EC-SOD transgenic mice have significantly diminished tumor formation compared with non-transgenic littermates. We further analyzed epidermis cell proliferation and DNA damage extent under TPA treatment. It provided a view that EC-SOD might play a protective role in skin carcinogenesis. 2. Methods 1) Generation of EC-SOD transgenic mice To induce skin-specific expression of mouse EC-SOD (756bp), human keratin 14 promoter (1,952bp) described as a keratinocyte-specific promoter was cloned from the pBS $KS^+$ vector with a human keratin 14 promoter region. The expression vectors were constructed by recombination of EC-SOD gene into pCR II TOPO vectors (Invitrogen, USA). In addition chicken ovalbumin poly A (192bp) was inserted. The plasmid DNA containing the sense-strand transgene vector was purified using Qiagen Midi kit, and digested with HindIII and Xhol to produce a linear construct consisting of the human K14 skin promoter, the mouse EC-SOD CDNA and a polyadenylation sequence. The DNA construct was microinjected into fertilized C57BL/6 X C3H embryos. Ten founder mice expressing the mouse EC-SOD were established by breeding into C57BL/6 X C3H hybrid mice. Transgenic mice were confirmed by PCR using primer hk14-2F: TTGTCTCTAATAGAGGGTC; SOD-2R: TCAAGCCTGTCTATCTTCT;(95$^{\circ}C$ for 1 min; 51$^{\circ}C$ for 1min; 72$^{\circ}C$ for 1 min, 30 cycles). The line with the highest transgene expression was selected for further analysis. 2) DMBA/TPA induced skin carcinogenesis The treatment groups consisted of 7$\sim$8 week-old transgenic mouse (10 male and 10 female) and theirlittermates. DMBA/TPA two-stage induced carcinogenesis was described somewhere, briefly, the dorsal skin was shaved 2 days prior to application of DMBA and TPA. The mice groups were treated with 20 nmol of DMBA dissolved in 200 ${\mu}l$ acetone on each mouse, after one week, 4g TPA also dissolved in 200 ${\mu}l$ acetone was applied weekly to the same area for 15 weeks. Application 200 ${\mu}l$ acetone alone to mice were used as a control, no tumors were observed. Mice were examined for skin tumors every week. 3. Results 1) Characterization of mouse EC-SOD transgenic mice To verify the transgenic mice whether properly overexpressed the mouse EC-SOD gene in the skin, EC-SOD mRNA and protein in the skin were assessed. mRNA levels of EC-SOD gene and tissue-specific expression were analyzed by RT-PCR. The representative band showing that 3 lines of transgenic mice overexpressed EC-SOD gene, and line 27 was used in this study. Immunohistochemical staining showed that the level of EC-SOD protein was also increased in the skin of transgenic mice. These results demonstrate the characteristics of EC-SOD skin-specific overexpressed transgenic mice. 2) 8OH-dG detection assay We focused the effect of TPA-induced oxidative stress on DNA damage. 8OH-dG detection assay was carried out. Our data suggest that TPA treatment induced DNA damage both in transgenic mice and non-ransgenic littermates. 80H-dcadducts appeared to be a more detection in non-transgenic mice but is lower in the transgenic mice, companying with a TPA dose-dependent manner. High TPA concentration is more influential. These results indicate that increased EC-SOD reduced the 8OH-dGexpression, in other words, overexpression of EC-SOD protected against oxidative damage to the DNA. 3) TPA induced cell proliferative response in EC-SOD transgenic mice and wild-type mice The effect of TPA on cellular proliferation in mouse ear between transgenic and littermates were investigated by BrdU-labeling. We analyzed the BrdU-uptake after the application of TPA to the skin of inner aspect of the ear. We found a mild effect in both genotypes using 0.2 wnol and a more induction of cell proliferation using 0.4 nmol TPA in transgenic than in non-transgenic mice. These results showed that the epidermis cell showed accelerated proliferative response after the application of TPA treatment, overexpressed EC-SOD could inhibit the proliferation rate induced by TPA treatment, otherwise the effect of TPA appears to depend on the dose used. 4) Reduced tumors formation in EC-SOD transgenic mice on DMBA-TPA induced skin carcinogenesis model For the effect of increased EC-SOD in tumor formation, tumor incidence and tumor numbers were examined. Our data showed that there were no tumors detected in transgenic and non-transgenic mice treated with acetone alone. Tumor appeared at 6 week after DMBA-TPA treatment in some individuals of both genotypes. 100% of the non-transgenic mice developed tumors, whereas 61% of transgenic mice developed tumors at the 18 week. The tumors yields were about 2-fold decrease in transgenic mice than in normal mice on average. These results suggest that increased EC-SOD in mouse skin reduced tumorigenicity in the two-stage carcinogenesis model.
