목적 : 홍화씨 폴리페놀의 어떤 성분이 뼈 보호효과가 우수한지 알아내고, 그 작용기전을 밝히기 위하여 동물실험과 골아세포실험을 병행하였다.
1) 골다공 쥐실험: 홍화씨 폴리페놀인 세로토닌, 리그난, 플라본의 뼈보호작용
연구방법 : 난소를 제거하거나 가수술한 쥐에게 여러 홍화성분을 4주동안 먹인 다음, 다리 뼈인 경골을 분리하여 조직학적인 차이를 전자현미경과 광학현미경으로 관찰하였다.
식이성분: 홍화박 (15, 30%), 폴리페놀 추출물(SE1, SE2), 리그난, 플라본 및 세포토닌
연구결과 : 경골의 골밀도

연구결과 : 경골의 골밀도 분석결과, 난소제거 4주 후에 심한 골다공증을 보였고, 골수에는 지방함량이 다량 증가하였다. 한편, 난소제거 후 홍화박과 홍화의 폴리페놀 추출물을 섭취한 쥐에서는 골밀도가 대조군의 75- 87% 정도로 나타났다. 이는 홍화씨 성분을 섭취하지 않은 쥐의 골밀도보다 두배 정도 높은 값이며, 골수의 지방함량도 대조군과 차이가 없었다. 뼈가 형성되고 있는 부분인 ost eoid를 염색한 결과, 난소제거군은 ost eoid가 정상치의 30%정도 밖에 되지 않았지만, 홍화를 먹인 4군에서는 정상치와 거의 같은 수준으로 나타났다. 홍화박과 홍화씨 폴리페놀 추출물의 뼈 보호작용은 거의 같게 나타났다.
세로토닌, 리그난, 플라본을 4주동안 먹인 쥐 경골밀도는 각각 대조군의 82, 68, 65%로 높게 유지되었다. 한편 뼈를 분해하는 파골세포의 수는 난소제거군은 대조군보다 66%나 높았지만, 세로토닌, 리그난, 플라본 섭취군에서는 대조군보다 20- 30% 정도 높게 나타났다.
골아세포실험 2: 홍화씨 폴리페놀의 골아세포 증식 및 분화 촉진작용 기전
연구방법 : 폴리페놀들의 작용이 estrogen r eceptor를 경유하여 작용하는지 알아보기 위하여 수용체에 특이적인 ant ag onist를 처리하였다. 또한 중요한 신호전달체계인 MAP kin ase를 경유하는지 알아보기 위하여 ERK1/ 2의 인산화를 West ern blot으로 측정하였다.
연구결과 : estrog en receptor ant ag onist를 전처리하면, 폴리페놀인 리그난, 플라본, 및 세라토닌에 의한 골아세포의 증식이 감소되고, 분화인자들인 collagen , alkaline ph osphat as 유전자 발현도 감소하였다. 또한 폴리페놀들은 ERK1/ 2의 인산화를 10분 이내로 빠르게 증가시켰으며, 이증가는 estrogen receptor antagonist에 의해 억제되었다.

We previously report ed that feeding safflower seeds attenuated ovariectomy (Ovx)-in duced bone loss in rats. In the present study, we investigated whether the bone-protecting effect of safflower seeds is due to polyphenolic compounds. Female Sprague-Dawley rats (12 week old) were subjected to bilateral Ovx or sham surgery. On day 0, Ovx rats were randomly assigned to groups and either fed a diet containing powder of defatted safflower seeds (SP) or polyphenolic compounds extracted from safflower seeds (SE), or injected with 17-estradiol (E2) for 4 weeks. Ovx resulted in a dramatic reduction in trabecular bone mass of the proximal tibia (approximately 40% of the sham value), increase in fat deposition in the bone marrow. E2 treatment completely prevented bone loss as well as the marrow adiposity, as examined by scanning electron microscopy and histomorphometry. Feeding both diets containing SP and SE exerted almost the same degree of bone-protecting effects (approximately 80% of the bone mass of the sham group) and prevented fat deposition in the bone marrow, although their effects were lower than that of E2. The amount of theosteoid bone, reflecting newly forming bone, decreased to 35% of the sham group in the Ovx rats, but was maintained at almost near the sham level both in the Ovx+SP and Ovx+SE groups.
In an attempt to find out which polyphenolic compound is effective in bone formation, effects of lignans, flavones and serotonin derivatives extracted from safflower seeds on the proliferation of ROS 17/2.8 osteoblast-like cells were also assessed in vitro. All of lignans, flavones and serotonin derivatives stimulated proliferation of the osteoblast-like cells in a dose-depen dent manner ($10^-^1^5-10^-^6$ M), as potently as E2 and genistein, a soy isoflavone. The present data demonstrated that polyphenolic compounds in safflower seeds stimulated bone formation and prevented bone loss caused by estrogen deficiency. The beneficial effects of safflower seeds seem to be mediated, at least partly, by the stimulating effect of the polyphenolic compounds on proliferation of osteoblasts.
