연구의 목적 및 내용
■ 연구 목적: 항산균 및 병원성 인자에 의한 면역세포의 분화/활성/억제를 조절하는 기전 연구를 통해 숙주면역을 조절하는 병원성인자를 발굴하고 항산균의 종 특이적 병인기전을 원론적인 측면에서 재조명하여 이를 기반으로 숙주의 방어기전과 새로운 치료
target 발굴, 난치성 면역질환으로의 응용기법을 제시하고자 함.
■ 1단계 연구: 1) M. tuberculosis, M. abscessus, M. avium complex를 중심으로 다양한 세포, 동물 감염모델 확립, 2) 균종, 균주에 따른 병원
연구의 목적 및 내용
■ 연구 목적: 항산균 및 병원성 인자에 의한 면역세포의 분화/활성/억제를 조절하는 기전 연구를 통해 숙주면역을 조절하는 병원성인자를 발굴하고 항산균의 종 특이적 병인기전을 원론적인 측면에서 재조명하여 이를 기반으로 숙주의 방어기전과 새로운 치료

■ 1단계 연구: 1) M. tuberculosis, M. abscessus, M. avium complex를 중심으로 다양한 세포, 동물 감염모델 확립, 2) 균종, 균주에 따른 병원성 비교/분석과 병원성에 따른 면역학적 특성 규명, 3) comparative genomics, proteomics, transcriptomics 기법
을 활용한 항산균의 총체적 병원성인자 발굴, 재조합 단백질 생산, 항원 library 구축, 4) 이를 기반으로 in vitro screening을 통하여 면역세포의 분화/활성/억제를 조절하는 병원성 인자의 선별과 대량 생산
■ 2단계 연구: 1) 항산균 또는 선별된 병원성 인자에 의한 면역세포의 분화/활성/억제 기전과 특성 분석을 통하여 2) 각 항산균의 균종에 따른 특이적인 면역병인을 규명하고 3) 면역세포의 polarization을 조절하는 병원성 인자를 선별, 특성에 맞게 class화,
4) 각 병원성 인자의 세포 수용체 및 세포내 신호전달 규명
■ 3단계 연구: 1) 면역세포의 polarization을 이용한 항산균 감염의 면역학적 치료기법 연구, 2) 항산균 항원과 수지상 세포를 이용한 면역세포기반 항산균 감염의 예방/치료 백신개발, 3) 항산균 항원과 면역세포를 이용한 난치질환 치료기법에 대한 응용 연구
연구결과
■ 면역세포의 분화/분석 기술 확립: 수지상세포 (BMDC, IKDC, pDC)와 큰포식세포(M1/M2)의 분화기술과 활성/억제를 평가할 수 있는 분석기술 확립
■ 세포/동물 모델을 이용한 항산균의 병원성 평가와 항원 library 구축: 표준균주와 임상균주의 병원성과 면역반응 분석을 바탕으로 균종 특이적 병인기전 규명, 다양한 기법을 이용한 항원발굴과 500개의 재조합 단백질 clone으로 구성된 항원 library 구축
■ 면역세포의 특성을 조절하는 항산균 항원의 class화: in vitro screening을 통하여 수지 상세포의 활성 (20종), 억제 (5종), 면역관용 (2종)을 유도하는 항산균 항원 발굴
■ 각 병원성 인자의 세포 수용체 및 세포내 신호전달 기전 규명: TLR4 agonist (8종), TLR2 agonist (4종), LPS 결합 단백질 (1종)을 발굴하고 각 관련 신호전달 기전규명
■ 결핵균, M. kansasii 감염 특이적 골수유래 IKDC와 항원 특이적 NK cell의 분화규명
■ 큰포식세포의 세포사멸을 유도하는 항원 발굴: 결핵균 2종, M. abscessus 1종 발굴
■ 발굴항원을 이용한 결핵의 subunit vaccine (Rv0351, Rv2769c, Rv2299c) 평가, 수지상세포 기반 M. abscessus 감염의 치료기법 검증, 결핵의 booster 백신 효능 검증
■ 항산균 항원과 면역세포를 이용한 난치질환 응용연구: 수지상세포의 활성화를 통하여 Th1-type의 T-cell 면역을 유도하는 결핵균의 항원 (Cancer 모델, HBHA와 Rv0652) (Asthma 모델, HspX와 Hsp90항원)을 이용한 면역치료백신 개발 연구
연구결과의 활용계획
■ 항산균의 종 특이적 병인기전 이해와 면역병리 기전의 기초연구 자료 제공
■ 항산균 또는 핵심병원성 인자에 의한 면역세포의 분화/억제/활성 연구에 대한 기반기술력 확보와 emerging pathogen의 근본적인 해결을 위한 균측 치료 타깃으로의 활용
■ 구축된 항원라이브러리를 공유, 활용함으로써 유사 감염질환에 대한 질병 발달기전 및 방어면역 기작의 이해와 치료법 및 백신개발에 대한 국내 연구의 활성화 유도

■ Research purpose: Investigation of host defense mechanism, identification of novel therapeutic targets, and application for incurable immune diseases via study on mycobacterium species-specific pathogenesis or their virulence factors regulating the differentiation, activation, and inhibition of immune cells
■ Step 1: 1) Establishment of in vitro and in vivo infection models for M. tuberculosis, M. abscessus, M. avium complex, 2) Comparative analysis of virulence according to mycobacterium species and strains, 3) Identification of species-specific virulence factors, production of recombinant proteins, construct of antigen library via comparative genomics, proteomics, transcriptomics, 4) Bulk production of selected factors related to the differentiation, activation, and inhibition of immune cells via in vitro screening. ■ Step 2: 1) Characterization of mechanism of virulence factors regulating the differentiation, activation, and inhibition of immune cells, 2) Investigation of mycobacterium species-specific immunopathogenesis, 3) Classification of virulence factors regulating polarization of immune cells, 4) Investigation of virulence factors in signal transduction and interactign receptors. ■ Step 3: 1) Study on immunological interventions for mycobacterial infection based on immune cell polarization by antigens, 2) Development of DC-based preventive/therapeutic vaccine against mycobacterial infection, 3) Practical application using mycobacterial antigens and immune cells for incurable immune diseases
Result
■ Establishment of research tools for differentiation, activation, and inhibition of immune cell including dendritic cells (BMDC, IKDC, pDC) and macrophage (M1/M2). ■ In vitro and in vivo evaluation of mycobacterial pathogenesis by comparing immune responses between reference and clinical strains and investigation of specie-specific pathogenesis.
Construction of antigen library consisting of 500 recombinant clones discovered by various tools. ■ Classification of mycobacterial antigens regulating immune cells: Identification of activator (20), inhibitor (5), inducer of tolerogenic response for DCs. ■ Investigation on receptors and the related signal transduction of selected virulence factors: Identification of
TLR4 agonist (8), TLR2 agonist (4), and LPS-binding protein (1). ■ Investigation on differentiation of bone marrow immune cells to IKDCs and NK cells by the M. tuberculosis- or M. kansasii-infection or their antigen-specific manner. ■ Identification
of macrophage cell death-inducers from M. tuberculosis (2) and M. abscessus (1). ■ Evaluation of subunit vaccine efficacy for Rv0351, Rv2769c, and Rv2299c in tuberculosis, validation of DC-based therapy for M. abscessus infection, and evaluation of DC-based BCG booster for M. tuberculosis infection. ■ Application of mycobacterial antigen and immune cells for incurable immune diseases: DC-based immunotherapy using M. tuberculosis antigens inducing Th1-type T-cell immune response for cancer (HBHA and Rv0652) and asthma (HspX and Hsp90).
