Human에서 본DDS 제제의 용량을 2배로 감량하였음에도 불구하고 시판제제와 비슷한 생물학적동등성을 나타내었다. 첨가제 자료 검토 및 배합성 확립 - 본 연구에 SMEDDS의 제조에 사용되는 오일, 지방산 및 계면활성제류와 다양한 가용화 및 제어방출 첨가제 조성의 배합성을 물성[점도, 함량, DSC 특성, 방출율] 자료를 바탕으로 점검하였으며 또한 허가 및 산업화에 응용시 배합성을 검토하였다. 최적제제의 공정, Batch 변화 및 scale-up확립 - 반고형상의 SMEDDS는 연질캅셀로 제조공정, 최적처방조성, 제조공정인자(온도, 혼합, 포장등)에 대한 조건을 확립하였다. 실험실적 조건에서 함량, 방출 및 점도등 batch 간의 차이가 없었으며 대량생산을 위하여 RP Scherer에서 scale up에 따른 문제점을 고찰하고 있다. 제품의 포장연구 - 본 DDS 제제는 연질캅셀의 충진이 바람직하며 수분에 민감하므로 방습 포장이 요구된다. 포장 재료로 PTP 혹은 PVC-PE-PVDC/Alu blister포장등이었으며 추후 대량생산공정과 연계하여 포장 방법 및 재료의 선정과 제제의 안정성을 동시에 고려하였다. 조건부 제품허가 및 생동성 허가서 작성 - 기준 및 시험법의 조건을 확립하기 위한 시판제제와 본 DDS 제제의 비교 용출 결과 본 DDS 제제가 위액과 장액 모두에서 용출율이 높아 두제제간의 비교용출율은 동등하지 않았고 식약청 생동 규정에 의하여 조건부허가용 생동 계획서를 이미 작성 완료하였으며 추후 식약청으로부터 조건부 품목허가를 얻을 예정이다. 최종연구결론 - 본 연구를 통하여 가용화 및 제어방출 기술에 의한 경구용 생체이용율 극대화한 제제에 대한 최적 조성의 처방화 및 평가를 완료하였다. 최적조성의 본DDS 연질캅셀 제제에 대한 기준 및 시험법, 정량법, 안정성, PK 동물자료와 비교용출 자료를 확보하였으며 최종적으로 제제에 대한 scale-up 공정중에 있다. 본 제제는 반고형, 반투명의 무정형 점액제제로 기존 제제에 비하여 높은 용출율, 안정성과 생체이용율을 나타내었다. 본 기술개발로 고가의 기존의 1회 100mg을 투약하는 기존제제를, 1회 약50mg으로 낮추어 환자의 복용 순응도 및 약물의경제성을 높인, 본DDS제제의 산업화가 1-2년안에 가능하리라 생각한다.

Research goal: Development and evaluation of itraconazole (lTR) preparation with controlled bioavailability using solubilization and controlled release techniques. Study of solubilizing compositions: The solubility of ITR in various oils, fatty acids and surfactants, and in their microemulsion systems was investigated. Tween and oleic acid appeared to be better excipient. Controlled release and coating techniques: The novel DDS is a semisolid, opaque and amorphous SMEDDS. This preparation turned into o/w microemulsion when contacted with water in a controlled fashion. Preparation and physiycochemical properties: SMEDDS mainly consisting of oil. fatty acid and surfactant was prepared using heat melting method. The SMEDDS had 100-800nm size distribution and amorphous structure as studied by DSC. Release characteristics: The release properties. preparation conditions. rheology. stability were highly dependent on formulation compositions of SMEDDS. The SMEDDS turned into microemulsion when orally given and showed extremely increased dissolution compared to commercial Sporanox capsule. Stability study: The drug contents and physical properties in optimized SMEDDS were not changed during storage at room temperture (RT) or 40℃/75% RH. The dissolution rate was stable at RT but slightly decreased at 40°C/75% RH. Pharmacokinetic comparisons: The current DDS showed 2-3 times greater AUC and Cmax when orally given in rats. This novel DDS was observed to be bioeqivalent with commercial Sporanox capsule even at twice lower dose. Selection of excipients and compatability: various types of oil. fatty acid and surfactant were chosen and their compatabilities were also evaluated based on rheology. ITR contents, DSC properties and release characteristics for further application on FDA approval and industrialization. Processing. batch variation and scale-up: The formulation compositions. filling conditions and processing parameters such as temperature, mixing and package of semisolid SMEDDS in soft capsules were established. The batch variation of the lab-scale SMEDDS was not observed. The scale-up processing conditions were also on investigation by RP Scherer. Packaging study: SMEDDS is filled in soft capsule. The packaging materials such as PTP or PVC-PE-PVDC/Alu blister was used to protect against humidity. The detailed method and type of packaging materials must be carefully inspected for future scale-up process. Documentation for IND and BE test: The current SMEDDS was not bioequivalent in gastric and intestinal fluid with Sporanox according to comparative dissolution guideline between the two. In addition. documentation for bioeqivalency test was also established for IND works. Final remarks: Novel DDS with enhanced dissolution, stability and bioavailability was optimally formulated using advanced solubilization and controlled release techniques. The standard analytical methods were also established. This unique semisolid, opaque and amophous SMEDDS preparation with twice lower dose provided an alternative to current commercial capsule within 1-2 years. The current SMEDDS have improved patient compliance and economical cost.
