○ 연구결과
본 연구에서는 비열살균 기술의 식품산업에의 활용도 증진을 위해 기계적 장치로 생성할 수 있는 살균소독수의 표면처리 살균기술을 절임식품에 적용할 수 있는 기술적 기반을 마련하고 실용화하기 위하여 이산화염소수와 전기분해수의 활용 기술 개발을 목표로 하였다. 이를 위하여 국내 식품산업에서 도입이 검토되고 있는 이산화염소수의 고순도와 고수율 생성기술과 확보하여 원료의 세정, 이산화염소수의 살균소독에 따른 생성 부산물의 화학적 안전성 분석, 살균 유효성 평가와 더불어 전기분해수의 살균 유효성을 평가하여 절임식품의 전처리에
○ 연구결과
본 연구에서는 비열살균 기술의 식품산업에의 활용도 증진을 위해 기계적 장치로 생성할 수 있는 살균소독수의 표면처리 살균기술을 절임식품에 적용할 수 있는 기술적 기반을 마련하고 실용화하기 위하여 이산화염소수와 전기분해수의 활용 기술 개발을 목표로 하였다. 이를 위하여 국내 식품산업에서 도입이 검토되고 있는 이산화염소수의 고순도와 고수율 생성기술과 확보하여 원료의 세정, 이산화염소수의 살균소독에 따른 생성 부산물의 화학적 안전성 분석, 살균 유효성 평가와 더불어 전기분해수의 살균 유효성을 평가하여 절임식품의 전처리에 활용할 수 있는 기초적인 자료를 도출하였다.
이를 토대로 참여업체인 싱그람종합식품의 단무지 생산공정에 적용하여 원료 전처리, 절임, 탈염 및 충진수로 활용할 수 있는 적용 가능성을 검토하였다.
1. 살균소독수 생성시스템 개발 및 생성조건 확립
신선/신선편이 농산물 생산공정의 미생물학적 위생성 향상을 위해 광범위하게 사용되고 있는 염소계 살균소독제 대체 물질로 주목받고 있는 이산화염소수를 아염-염소가스(NaClO2-Cl2(g)) 방식의 기기장치로 생성할 수 있는 생성장치를 제작하였다. 이산화염소수 발생 최적조건을 검토하기 위하여 생성원료인 NaClO2 농도(0.01~0.1%)와 Cl2 gas 투입량(100~1,000 g/hr)에 따른 chlorite, chlorate, FAC 및 chlorine dioxide의 생성량을 조사하였다. Chlorate, FAC 및 chloride dioxide 생성량은 NaClO2 및 Cl2 투입량이 증가할 수록 생성량이 증가하는 경향을 나타내었다. NaClO2 농도 및 Cl2gas 투입량에 따른 수율은 chlorite 및 chlorate와 생성량이 직적접인 영향을 미치므로 chlorite와 chlorate의 생성량이 낮았던 NaClO2 및 Cl2의 투입농도범위에서 높은 값을 나타내었다. 즉, 0.01%
NaClO2에서는 200~600 g/hr Cl2 gas에서 95% 수준, 0.05% NaClO2에서는 400 g/hr의 Cl2 gas에서 97%를, 그리고 0.1% NaClO2에서는 500-1,000 g/hr에서 96% 이상의 수율을 나타내었다. 0.01% NaClO2에서는 200, 300, 400, 600 및 1,000 g/hr Cl2 gas에서 93% 수준을, 0.05% NaClO2에서는 900g/hr Cl2 gas에서 94.0%, 그리고 0.1% NaClO2에서는 800과 900 g/hr Cl2 gas에서 95.0%의 최대 순도를 나타내었다. 30 ppm 이산화염소수의 5℃ 및 10℃에서의 경시적인 변화를 검토한 결과 ClO2 및 Cl2는 140분 경과시까지 그다지 크지 않았던 반면 25℃에서는 초기 30.6 ppm에서 30분 경과시 7.6ppm 수준으로 급격히 감소하는 것으로 나타났다.
NaCl 농도를 달리하면서 생성한 중성전기분해수의 물성을 측정한 결과 HClO 함량은 전해액인
NaCl 농도가 높아질수록, 그리고 분해시간이 증가할수록 대체적으로 함량이 증가하는 뚜렷한 경향을 나타내었다. 0.01% NaCl 전해액에서는 분해 30분후 11.4 ppm으로 최대값을, 0.02% 전해액에서는 분해 30분후 49.6 ppm으로 최대값을 나타내었다. 생성수의 pH는 NaCl의 양이 증가하고 분해시간이 길어질수록 대체적으로 상승하여 pH 7~9 범위를 나타내었다.
2. Biocide로서의 살균 유효성 및 화학적 안전성 평가
총 42종의 박테리아, 곰팡이 및 효모를 대상으로 5, 10 및 15 ppm의 이산화염소수로 살균속독의 효과를 평가한 결과 대부분 5 ppm 농도에서 6 log 수준의 감균효과를 나타내었으나 이산화염소에 대한 감수성은 균주별로 다소의 차이를 나타내었다. 중성전기분해수의 살균유효성을 평가한 결과, E.coli에 대해서는 HClO 농도 35.5 ppm 이상일 때 99.999%의 감소 효과가 있었고 이때 전기분해수의 물성은 pH 8, ORP 705 mV였다. S. aureus에 대해서는 85.1 ppm의 HClO 함량일 때 유의한 결과를 나타내었으며 이때 전기분해수의 물성은 pH 8.60, ORP 750 mV였다. 이산화염소수의 살균소독 부산물인 chlorite 등의 chlorine species를 분석한 결과, 케일과 상추를 시료로 하여 호울(whole)과 절단(cut) 형태의 시료를 10, 30, 50 및 100 ppm의 이산화염소수에 침지세정하면서 이들의 경시적인 생성 변화량을 검토하였다. 상추에서는 침지시간의 경과에 따라 chlorite는 0.2~2 ppm, chlorate는 약 2~5 ppm 증가하였으며, 절단 상추는 chlorite는 0.3~6.3 ppm, chlorate는 0.6~9.1 ppm 증가하였다.
케일은 경우에는 이산화염소의 농도 증가에 따른 chlorite의 증가는 관찰되지 않았으나 chlorate는 약 0.4~1.5 ppm 증가하였다. 이러한 경향은 치콘, 깻잎 및 케일에서도 유사하였다. ClO2, free Cl, Oxi-Cl의 농도는 경시적으로 감소하는 경향을 나타내었으나 NH2Cl, NHCl2 및 chloride는 이산화염소의 농도 및 침지시간에 따른 경시적인 변화를 나타내지 않았다. 또한 상추, 치콘, 깻잎 및 케일의 침지 세정중 THMs의 생성량을 분석한 결과, 4종의 채소류에서 469~1,139 ppb의 chloroform과 84~173 ppb의 dichlorobromomethane이 검출되었으며, 상추에서는 60~113 ppb의 dibromochloromethane이 검출되었다. 그리고 감귤, 방울토마토, 딸기 및 사과에서도 채소류와 유사한 수준의 chloroform과 dichlorobromomethane이 검출되었으나 감귤에서는 1,074-1,670 ppb의 bromoform이, 방울토마토에서는 57~89 ppb의 dibromochloromethane과 16~158 ppb의 bromoform이 100 ppm의 이산화염소수에서 검출되었다. Chloroform과 dichlorobromomethane은 원수로부터 유래하는 것으로 추정되었으나 dibromochloromethane과 bromoform은 살균 과정에서 생성된 것으로 판단되었다.
상추 등 4종의 채소류를 시료로 chlorite 및 chlorate의 생성을 분석한 결과, 호울(whole)보다는 절단(cut) 형태에서 증가량이 높았으며 100 ppm의 이산화염소수로 살균할 경우 chlorite는 약 3 ppm수준, chlorate는 10 ppm 수준으로 증가하였다. 또한 상추, 치콘, 깻잎 및 케일의 침지 세정중 THMs의 생성량을 분석한 결과, 원수에서 유래하는 THMs외에 dibromochloromethane과 bromoform도 과채류의 종류나 각종 토양, 또는 자연계 유래의 유기물질(NOM ; natural organic matter)에 의해 생성 가능성이 있는 것으로 판단되었다. 상추의 경우 이산화염소의 농도에 따라 Haloacetic acids(HAAs)인 CAA는 다소 증가하는 경향을 나타내었으나 침지시간에 따른 유의적 차이는 나타나지 않았다. CAA, BAA, DCAA, TCAA, BCAA, DBAA 모두 수십 ppb 수준으로 검출되었으며 이러한 경향을 케일에서도 유사하였다.
3. 이산화염소수를 이용한 전처리 활용기술 개발
Acetic acid, malic acid 및 citric acid의 첨가량을 각각 0, 0.5 및 1.0%로 첨가한 다음 시간의 경과에 따른 산성화 이산화염소수의 염소계 화합물의 변화량을 분석한 결과, 60분 경과시까지 1.0% acetic acid 첨가가 ClO2 소실량이 약간 큰 것으로 나타났으며 Cl2와 total Cl량에는 첨가농도에 따른 차이가 크지 않은 것으로 생각되었다. 30 ppm의 이산화염소수에서는 소금의 첨가 농도가 증가할 수록 ClO2의 감소량이 대체적으로 증가하고 특히 20%와 30%의 첨가에서는 현격한 감소를 나타내어 20% 소금 첨가에서는 30분후 초기 농도의 60%가 감소하고 30% 소금첨가에서는 70%가 감소하였다.
상추, 치콘, 깻잎, 케일의 채소류에 대한 세정 침지 살균에서는 이산화염소 농도 의존성 감균 효과를 나타내어 3~4 log의 총균수 감소 효과가 있었다. 과일의 경우에도 농도 의존성 감균효과는 동일하였으며 대략 3~5 log의 총균수 감균효과가 있음이 확인되었다.
NaClO2-Cl2 방식으로 발생시킨 이산화염소수(순도 96.0%, 수율 97.0%)를 10-30 ppm의 농도로 상추, 치콘, 깻잎 및 케일을 침지하여 살균소독하면서 이산화염소수의 ClO2, Free Cl, NH2Cl, NHCl2, Oxi-Cl, Cl-, ClO2- 및 ClO3- 함량의 경시적인 변화량을 분석하였다. 이산화염소수중의 ClO2, Free Cl, NH2Cl, NHCl2, 및 Oxi-Cl는 모든 처리구에서 침지시간의 경과에 따라 유의적으로 감소하였다. 상추, 치콘, 깻잎 및 케일의 침지 30분후 이산화염소수중의 chlorite 증가량은 10 ppm 이산화염소수에서는 0~0.24 ppm, 30 ppm의 이산화염소수에서는 0.22~0.65 ppm이었으며, chlorate 증가량은 10 ppm의 이산화염소수에서는 0.27~0.39 ppm, 30 ppm의 이산화염소수에서는 0.38~1.24 ppm이었다. 한편, 절단상추, 치콘, 깻잎 및 케일의 침지 30분후 이산화염소수중의 chlorite 증가량은 10 ppm 이산화염소수에서는 0.39~0.57 ppm, 30 ppm의 이산화염소수에서는 0.38~2.38 ppm이었고, chlorate의 증가량은 10ppm의 이산화염소수에서는 0.50~1.06 ppm이었고, 30 ppm의 이산화염소수에서는 0.51~5.95 ppm 이었다. 사용한 이산화염소수의 농도가 증가할 수록, 침지 시간이 경과할수록 chlorite와 chlorate의 농도는 증가하였으며, 절단한 경우가 절단하지 않는 경우에 비해 chlorite와 chlorate의 증가량이 높았다.
케일을 acetic acid, citric acid 및 malic acid를 0.5% 및 1.0% 농도로 첨가한 이산화염소수에 세정하면서 경시적인 미생물, 염소 화합물 함량 변화를 살펴본 결과는 이산화염소수에 malic acid를 첨가하여 세정한 케일의 미생물학적 세정 효과가 가장 큰 것으로 나타나 0.5%와 1.0% malic acid 첨가 이산화염소수 침지 세정은 30분에 무첨가에 비해 2 log cycle의 상승효과를 나타내어 101 CFU/g 이하로 감소하여 약 5 log cycle의 감균효과를 나타내었다.
4. 위생적 고품질의 절임식품(단무지) 제조를 위한 적용기술 개발
원료무의 살균을 위해 전기분해수인 강산성 전기분해수, 약알칼리성 전기분해수 및 미산성 전기분해수, 그리고 이산화염소수의 농도를 달리하여 침지 세정한 결과, 초기 균수 1.7×105 CFU/g에서 30분후 강산성 전기분해수는 1.6×103 CFU/g, 약알칼리성 전기분해수는 30분후 1.0×103 CFU/g, 미산성 전기분해수는 1.2×103 CFU/g으로 감소하였으며, 이산화염소수의 경우에는 초기균수 1.7×105 CFU/g에서 10 ppm의 이산화염소수 처리로 30분후 9.7×102 CFU/g으로, 20 ppm의 이산화염소수 처리로 30분후 2.7×102 CFU/g으로, 그리고 30 ppm의 이산화염소수 처리로 1.6×102 CFU/g으로 균수가 감소하였다. 각 살균소독수의 유효농도를 감안할 때 이산화염소수>미산성 전기분해수>강산성 전기분해수․약알칼리성 전기분해수 순으로 살균소독력이 높은 것으로 판단되었다.
이산화염소는 식염의 농도가 0~30%로 증가할 수록 ClO2의 감소량이 증가하였으며 Cl2, Oxi-Cl 및 chloroamine인 monochloroamine 및 dichloroamine은 경시적으로 감소하였다. 절임중 pH의 변화는 절임수와 절임방법에 따른 차이가 다소 보였으며 절임 후 15일째부터는 수도수를 절임수로 사용한 절임무를 제외한 타 처리구의 경우 pH 5.73~6.03의 수준을 나타내었다.
염장무의 산도는 전반적으로 60일 이후 모든 처리구에서 급격한 감소를 보였으며 절임 방법에 있어서는 대체적으로 건식법에 의한 절임 무가 습식법에 의한 무보다 다소 높은 수준을 나타냈다. 절임 중 염도의 변화는 건식 및 습식의 절임방법에 따른 차이가 뚜렷하게 나타났으며, 건식 절임의 경우 모든 처리구에서 초기 9.00-9.07%이었던 것이 저장 15일째에 급격히 증가한 이후 저장 90일에 17%수준을 나타내었다. 습식 절임법의 경우에는 13% 수준을 나타내었다. 염장무의 hardness는 전반적으로 저장 초기에 다소 증가한 후 저장 30일에 급격히 감소하여 416.7~557.0 g/cm2의 수준을 보인 초기치의 50% 수준인 205.1~292.2 g/cm2의 범위였다. 염장무의 색도는 전반적으로 저장기간이 경과함에 따라 L(lightness)값은 감소하였고 색차값(△E)은 증가하는 경향이었으며 절임방법에 따른 차이가 뚜렷하게 나타났는데 건식 절임법의 경우 a(redness) 및 b(yellowness)값은 증가하는 경향으로 나타났다. 원료무의 경우 총균수, 효모 및 곰팡이류, 대장균군에서 각각 5.1×106, 4.8×106, 1.2×102이었으며 절임방법에 따라 다소 다른 균수 변화를 나타내었다. 대조구에 비해서 전기분해수로 세척한 후 건식 절임한 처리구의 경우 2 log cycle 정도가 감소하였던 반면 습식 절임 처리구는 3 log cycle 정도 감소 효과를 나타내었다. 대장균군의 경우 저장기간 동안 모든 처리구에서 검출되지 않았다. 탈염중 미생물의 변화를 분석한 결과 강산성 전기분해수, 약알칼리성 전기분해수 및 30 ppm의 이산화염소수로 탈염한 탈염무의 미생물 감균 효과가 우수한 것으로 나타났다. pH는 대조구에 비해 약알칼리성 전기분해수와 이산화염소수를 탈염수로 사용한 경우 다소 높은 값을 나타내었나, ORP는 상대적으로 큰 차이를 나타내지 않았다. 또한 탈염속도는 30 ppm의 이산화염소수와 30 ppm의 약알칼리성 전기분해수 및 30 ppm의 이산화염소수를 mixed oxidant로 사용한 경우 가장 빠른 것으로 나타났다. Hardness는 대조구와 유사한 수준이었다. 탈염과정중 색도의 변화는 대조구에 비해 이산화염소수탈염처리가 황색도를 보다 빨리 저하시켜 관능적으로는 밝은 색상을 나타내는 특징을 가지고 있었다. 산도의 경우에도 30 ppm의 약알칼리성 전기분해수를 제외하고는 이산화염소수를 사용한 경우에 48시간 이후 산도의 감소속도가 상대적으로 빨랐다. 탈염정에서 30 ppm의 이산화염소수를 활용한 경우 총균수는 약 2 log cycle, 효모와 곰팡이는 약 1 log cycle의 감균 효과가 있었다. 미생물군수의 변화는 절임방법 및 조미액 제조수에 따라 대조구에 비해 1~2 log cycle 정도 낮은 균수를 나타내었다. pH와 산도는 유의적 차이가 없었다.
이산화염소수만을 충진수로 활용하여 60일간의 저장중 총균수는 저장 60일까지 102 CFU/g 수준, 효모와 곰팡이수, 대장균군수는 101 CFU/g의 상대적으로 낮은 균수가 유지되었다. 용기포장 통단무지의 경우 전기분해수나 이산화염소수를 사용한 경우, 색상과 씹힘성에서 대조구에 비해 높은 기호도를 나타내었다. 이외 염도 등에서는 유의적 차이를 나타내지 않았으며 약알카리성 전기분해수를 충진수로 사용한 경우 종합적으로 가장 높게 평가되었다. 이러한 경향은 진공포장의 경우에도 유사한 결과를 나타내었다. 충진수로서는 약알칼리성 전기분해수와 이산화염소수가 미생물학적 품질과 이화학적 품질 유지에 보다 효과적인 것으로 판단되었다.

Chlorine disinfectants such as Cl2 and HOCl are the most widely used chemicals for disinfection of drinking water, foods and food contact surfaces. Chlorine dioxide is a strong oxidizing and disinfecting agent which may have a practical application for non-thermal disinfection of foods especially fresh fruit and vegetables, and food contact surfaces such as food processing facilities in food industry. It is used as disinfectant either in a gaseous or aqueous form (ACD; aqueous chlorine dioxide). Chlorine dioxide was introduced as a potential effluent disinfectant and has provided good results and much less formation of harmful organic byproducts such as trihalomethanes (THMs) to substitute for chlorine. Because of these characteristics, chlorine dioxide has been investigated as one of the promising substitute disinfectants for chlorine.
The objective of this work is as follows. The first one is to evaluate disinfection efficacy of ACD, which is generated using NaClO2-Cl2 type generator on some fruits and vegetables including radish root. The second one is to analyze disinfection byproducts such as chlorite, chlorate, THMs and ahloacetic acids (HAAs) formed during immersion cleaning of fresh fruits and vegetables with ACD. The third one is to evaluate disinfection efficacy of electrolyzed water (hypochlorous acid water and hypochlorite water). The last one is to develop practical technology as microbial controls applicable to salted food industry on the bassis of these results.
1. Development of sterilizing water manufacturing system
Optimum concentration of NaClO2 solution and amount of gaseous Cl2 was investigated for production of aqueous ClO2 with high yield and purity using gaseous chlorine-chlorite ClO2 generator which can produce lower concentrations of chlorine dioxide applicable for direct-use in food processing as cleaner/sanitizer. Concentration of NaClO2 solution and amount of gaseous Cl2 was varied from 0.01 0.1% and 100 1,000 g/hr respectively. ~ ~ Produced concentrations of chlorite, chlorate, FAC(free available chlorine) and chlorine dioxide were increased by increasing of supplied concentration of NaClO2 solution and amount of gaseous Cl2. Optimum concentration of NaClO2 solution and amount of gaseous Cl2 was 0.1% and 900 g/hr respectively. (At this concentration, produced) ClO2 and FAC produced at these concentrations were 882.0 ppm and 8.0 ppm, and chlorite and chlorate were not detected. The yield and purity of ClO2 were 97.0% and 96.0% respectively.
Changes in concentration of ClO2 and Cl2 of ACD at 5℃ and 10℃ were not significantly decreased up to 140 min. However concentration of ClO2 at 25℃ was decreased from 30.6 ppm to 7.6 ppm after 30 min.
Concentration of hypochlorous acid of neutral electrolyzed water produced by circulation electrolyzation of NaCl was increased with NaCl concentration and electrolyzation time. Concentration of hypochlorous acid of electrolyzed water produced by 0.01% NaCl electrolyte reached to 11.4 ppm after 30 min of electrolyzation, and those reached to 49.6 ppm in 0.02% NaCl electrolyte. pH of produced electrolzyed water was increased by NaCl concentration and electrolyzation time to pH 7~9.
2. Disinfection efficacy and chemical safety as biocide
To evaluate disinfection efficacy of ACD, which is generated using NaClO2-Cl2 type generator, 42 kinds of food microorganism were tested by quantitative suspension test. Although total bacterial counts decreased largely (6 log level) at 5ppm of ACD, there were only a few changes in sensitivity with different species.
Disinfection efficacy of neutral electrolyzed water showed 99.999% reduction against E. coli only in above 35.5 ppm of HOCl concentration. pH and oxidation-reduction potentials (ORP) of these electrolyzed water were pH 8.0 and 705 mV, respectively.
Also 99.999% reduction against S. aureus was showed only in 85.1 ppm of HOCl concentration. pH and oxidation-reduction potentials (ORP) of tthe electrolyzed water were pH 8.60 and 750 mV, respectively.
Despite of numerous advantages, chlorine dioxide have potential sources of risk for human health due to the inorganic byproducts such as chlorite and chlorate. Most of all studies on disinfection with chlorine dioxide were carried out mainly in the field of chlorination of drinking water, and because dosage and contact time required in operating conditions of food processing are different from those of drinking water treatments, it is important to investigate disinfection efficacy and disinfection byproducts formation in practical application of food processing.
In this research we have investigated changes of some chlorine species including chlorite and chlorate in ACD during immersion disinfection of some leafy vegetables such as leafy lettuce, chicon, sesame leaf and kale. Changes in chlorite and chlorate concentration with whole and cut kale and lettuce during immersion cleaning with ACD at different concentration (10, 30, 50 and 100 ppm) were investigated. In case of whole lettuce, 0.2 2 ppm for chlorite and 2 5 ppm for ~ ~ chlorate increased with immersion time. When it comes to cut leafy lettuce, 0.3~6.3 ppm for chlorite and 0.6~9.1 ppm for chlorate increased. However, although there were few changes in chlorite for whole kale, chlorate concentration increased about 0.4~1.5 ppm. In case of cut kale, 0.9~5.1 ppm for chlorite and 0.1~2.9 ppm for chlorate increased. However, although there were few changes in chlorite for whole kale, chlorate concentration increased about 0.4~1.5 ppm. In case of cut kale, 0.9~5.1 ppm for chlorite and 0.1~2.9 ppm for chlorate increased. Chlorite and chlorate content in ACD used as disinfectant increased with increasing dose of chlorine dioxide and immersion time, and cutting leafy vegetables. Contents of ClO2, free Cl, NH2Cl, NHCl2, and Oxi-Cl of ACD were decreased significantly in all treatments by immersion time. Although concentration of ClO2, free Cl, Oxi-Cl decreased, there were few changes in NH2Cl, NHCl2 and chloride. THMs such as chloroform, dichlorobromomethane and dibrochloromethane were detected, which was thought to be derived from tap water used to generate ACD. The formation of THMs was thought to be due to the natural organic matters derived from fruits/vegetables, soil and contaminated matters.
HAAs concentration in ACD used as disinfectant increased partly with increasing dose of chlorine dioxide and immersion time of leafy lettuce and kale partly.
3. Development of pre-treatment application technology using aqueous chlorine dioxide
Changes of some chlorine species including in ACD added with acetic acid, malic acid or citric acid were investigated. ClO2 concentration of ACD was decreased most rapidly in the case of addtion of citraic acid. Contents of Cl2, and total Cl were influenced by addition of organic acid. Acidified ACD with 0.5% and 1.0% of acetic acid, citric acid or malic acid showed higher disinfection efficacy (more than 2 log cycle) than ACD by itself in immersion disinfection of kale. ClO2 concentration of ACD decreased in proportion to added salt concentration. Especially ClO2 concentration was decreased to 60% and 70% of initial concentration respectively by 20% and 30% of salt addition.
When it comes to immersion cleaning with ACD, it also decreased at 3~4 log level.
In case of fruit, there was similar disinfection efficacy (3~5 log level).
Aqueous chlorine dioxide is a strong oxidizing and disinfecting agent which may have a practical application for non-thermal disinfection of foods especially fresh fruit and vegetables, and food contact surfaces such as food processing facilities. Despite of numerous advantages, ACD represents a potential sources of risk for human health due to the introduction of byproducts such as chlorite (ClO2-) and chlorate (ClO3-).
In this research we have investigated changes of some chlorine species including chlorite and chlorate in 10~30 ppm of ACD during immersion disinfection of some leafy vegetables such as leafy lettuce, chicon, sesame leaf and kale. Contents of ClO2, free Cl, NH2Cl, NHCl2, and Oxi-Cl of ACD were decreased significantly in all treatments by immersion time. After 30 min of immersion, increased amounts of chlorite were 0.0~0.24 ppm in 10 ppm of ACD, and 0.22~0.65 ppm in 30 ppm of ACD. Also those of chlorate were 0.27~0.39 ppm in 10 ppm of ACD, and 0.38~1.24 ppm in 30 ppm of ACD. In case of cut leafy vegetables, 0.39~0.57 ppm and 0.38~2.38 ppm for chlorite in 10 ppm and 30 ppm of ACD increased respectively. Also 0.5 0~1.06 ppm and 0.51~5.95 ppm for chlorate in 10 ppm and 30 ppm of ACD increased. Chlorite and chlorate content in ACD used as disinfectant increased with increasing dose of chlorine dioxide and immersion time, and cutting leafy vegetables.
4. Development of application technology for hygienic pre-treatment of salted foods
Microbial counts of raw radish root was decreased by immersion disinfection with ACD and electrolyzed water as follows. In the case of using ACD, 1.7×105 CFU/g of initial microbial counts was decreased to 9.7×102 CFU/g by 10 ppm of ACD for 30 min of disinfection, decreased to 2.7×102 CFU/g by 20 ppm of ACD for 30 min of disinfection, decreased to 1.6×102 CFU/g by 30 ppm of ACD for 30 min of disinfection. In the case of using strong electrolyzed water (SAEW), 1.7×105 CFU/g of initial microbial counts was decreased to 1.6×103 CFU/g for 30 min of disinfection. In the case of using low alkaline electrolyzed water (LAEW1), 1.7×105 CFU/g of initial microbial counts was decreased to 1.0×103 CFU/g for 30 min of disinfection. And in the case of using low acidic electrolyzed water (LAEW2), 1.7×105 CFU/g of initial microbial counts was decreased to 1.2×103 CFU/g for 30 min of disinfection. Considering of available chlorine, disinfection efficacy against raw radish roots was as follows; ACD>LAEW2>SAEW․LAEW1.
pH change of salted radish was almost stable maintaining between 5.7 and 6.0 after 15 days of salting. Although acidity was relatively lower than that of control group (Treatment TW), there was rapid decrease in acidity in all treated groups after 60 days of salting. Salinity of salted radish was stable between 13% level after increase on 15th day of salting, while salinity of wet process salted radish showed repetitive increase and decrease reaching 17% on the 90th day of salting. Hardness was rapidly decreased on the 30th day of salting showing 205.1~292.2 g/cm2, 50% of the initial value. Reducing sugar was decreased as time passes for dry process salted radish and it was twice higher for dry process salted radish compared to wet processed one after 30 days of salting.
Color of dry processed salted radish showed increase of a and b values in all treatment according to salting time. Total viable counts of dry processed treatment showed 2 log cycle reduction and those of wet processed treatment showed 3 log reduction compared to control treatment (TW). Also coliform count was not detected in all treatment during salting period.
Although there was any significant difference between salted radish root desalted with tap water (TW) and salted radish roots desalted with SAEW, LAEW, ACD or mixed oxidant according to pH, acidity, salinity, and sugar content during desalting, the final microbial counts of SAEW, LAEW, ACD or mixed oxidant was lower than that of TW. Especially desalting using 30 ppm of ACD was more effective than those of SAEW and LAEW in the view of microbial reduction. Also desalting velocity was more rapid in the case of desalting using 30 ppm of ACD or mixed oxidant (30 ppm of LAEW1 + 30 ppm of ACD) than other treatments. There was not any differences in hardness between salted radish root desalted with tap water (treatment TW) and salted radish roots desalted with SAEW, LAEW, ACD or mixed oxidant. Yellow color value b and acdidity of radish root desalted with ACD was lower than those of others. There was 2 log scale 1 log scale reduction in total microbial count and yeast & mould count of radish root desalted with 30 ppm of ACD.
When salted radish was vacuum packed, and stored at 10℃, initial microbial count of danmooji treated with ACD or electrolyzed water showed 1~2 log cycle reduction comparing with TW treatment. After 60 days of storage, microbial counts of damooji packed with seasoning solution using ACD were 102 CFU/g for total microbial count, 101 CFU/g for yeast & mould count, and coliform count respectively. pH, acidity, salinity and hardness was not different comparing with TW. Sensory characteristics such as color and crispness were preferable on container-packaged and vacuum packaged tong-danmooji packed with seasoning solution made from LAEW (low alkaline electrolyzed water) to the others. However ther were any significant differences in acidity, salty, yeast and moldy, et al. between LAEW and others. Considering with microbial and physicochemical quality of danmooji, packing with LAEW and ACD is more effective than those of others.
