제 1 장 굴에 있는 Transglutaminase (TGase)의 정제와 특성
제 1 절 TGase의 정제와 특성
개조개, 굴, 꼬막, 바지락, 백합, 비단가리비, 소라, 홍합 및 돌담치 수용성 추출물의 transglutaminase 비활성을 측정한 결과, 소라가 가장 높은 0.82±0.00 Units/mg-protein/min인 반면 돌담치는 비활성을 보이지 않았다. 굴은 0.27±0.00 Units/mg-protein/min의 활성을 보였다. 20-80% 포화 ammonium sulfate 분획물을 0.1 M NaCl을 포함하는 TEND 완충액에 투석하여 -20℃에서 동결 저장하면서 단백질 함량과 transglutaminase 활성을 측정한 결과, 저장기간의 증가와 더불어 단백질 함량은 감소하였으나, 효소 활성은 저장 33일까지 거의 변화가 없었다. 2회의 DEAE-Sepharose 이온교환 크로마토그래피와 젤 크로마토그래피(Superdex 200 prep grade)를 통해 굴에서 최종적으로 정제한 transglutaminase의 정제도는 1895배였고, 수율은 1.9%였다. 정제한 transglutaminase의 최적 pH와 온도는 각각 9.0과 45℃였으며, 10 mM CaCl2 농도에서 가장 높은 활성을 보였다. 55℃에서 30분 가열 시 활성의 약 50%를 소실하였으며, NaCl 농도는 transglutaminase의 활성에 거의 영향을 미치지 않았다.
제 2 절 패각 단백질의 단백분해효소 저해효과
굴 패각 분말의 조단백질 함량은 0.8%였으며, 굴패각에서 얻은 calcium acetate는 C먜가 전체의 97.15%를 차지하였다. 5% acetic acid 가용성 획분 중 분자량 10 kDa 이하의 획분이 10 kDa 이상의 획분에 비하여 높은 단백질 분해효소 저해효과를 보였으며, 특히 bromelain의 저해에 효과적이었다. 단백질 분해효소 저해제와 calcium acetate에서 세균에 대한 항균 활성과 간세포 독성은 관측되지 않았다. 굴 패각에서 부분정제한 5% acetic acid 추출물은 단배분해효소 저해제로서의 잠재적인 가능성을 가지는 것으로 보인다.
제 2 장 굴 부분 가수분해물의 제조와 기능성 확인
제 1 절 산 가수분해물의 제조와 특성
최대 수율의 산가수분해물을 제조하기 위한 최적 조건은 10 N HCl, 96, 가수분해 22시간이었다. 가수분해에 가장 큰 영향을 미치는 인자는 가수분해 시간, 염산의 농도, 가수분해 온도의 순이었다. 산 가수분해물과 탈염한 산가수분해물의 단백질 함량은 6.5%와 14.0%였고, 염의 함량은 각각 16.2%와 0%였다. 산 가수분해물에 분포하는 peptide의 분자량 400-2500 dalton 사이였다. 산 가수분해물의 아미노산 함량은 Lys>Leu>Tyr≒His≒Gly>Val의 순으로 높았고, Thr, Ser 및 Pro은 검출되지 않았다. 산 가수분해물의 세포 독성은 200 mg/mL까지 유의적인 독성은 보이지 않았다. 산가수분해물은 0.5 mM ascorbic acid와 비슷한 DPPH 라디칼 소거활성을 보였으며, 탈염한 산가수분해물의 DPPH EC50은 0.28 mg/mL였다. Linoleic acid에 대한 분획한 peptide의 농도별 항산화 효과는 0.1-0.01 mg/mL의 농도 구간에서 농도에 따른 유의차는 보이지 않았으나, 분자량이 작을수록 강한 항산화 효과를 가지는 것으로 나타났다. ACE 저해능은 탈염 산가수분해물 분자량 500-1000 dalton 획분이 가장 높았다. 산 가수분해물은 기능성 ingredient로서 잠재성인 사용 가능성은 있으나, 안전성에 관한 면밀한 검토가 아울러 이루어져야 할 것이다.
제 2 절 일단 효소 가수분해물의 제조와 특성
양식굴을 효율적으로 이용할 목적으로 6가지의 상업적 효소(Alcalase, Flavourzyme, Neutrase, Protamex, pepsin, trypsin)를 이용하여 굴 효소 가수분해물을 제조하고 그 특성에 대하여 살펴보았다. 상ㅇ럽적 효소 굴 가수분해물의 항산화능 및 ACE 저해능은 모두 Protamex로 1 시간 동안 가수분해시킨 것이 가장 우수하였고, 이때 이들의 IC50 값은 각각 1.16 mg/mL 및 1.49 mg/mL였다. 하지만 항균성은 모든 효소 가수분해물에서 인정되지 않았다. 그리고 ACE 저해능 및 항산화능은 가수분해 시간에 따른 의존성이 인정되지 않았다. Protamex로 1시간 동안 처리한 굴 가수분해물의 경우 가수분해 처리하지 않은 대조구에 비하여 29-66 kDa 획분과 6.5 kDa 부근의 획분이 모두 감소하여 저분자화 하는 경향을 나타내었다.
제 3 절 이단 효소가수분해물의 제조와 특성
일단 가수분해물과 이단 가수분해물들의 부패취에 관능검사를 실시한 결과 일단 및 이단 가수분해물과 같이 가수분해물의 종류에 관계없이 모든 시료구에서 전 관능요원들이 부패취를 감지못하였다. Alcalase, Neutrase, pepsin 및 trypsin 등과 같은 4종의 효소 이단 가수분해물 간의 가수분해율은 유의적인 차이가 인정되지 않았다. Protamex 가수분해물을 Alcalase, Neutrase, pepsin 및 trypsin 등과 같은 4종의 효소로 이단 가수분해한 가수분해물의 경우 모두 ACE 저해의 개선 효과 (Alcalase 가수분해물: 0.86 mg/mL, pepsin 가수분해물: 1.30 mg/mL 및 trypsin 가수분해물: 1.20 mg/mL)가 있었고, 그 중에서도 특히 Neutrase 가수분해물의 IC50이 0.40 mg/mL로 가장 개선 효과가 컸다. 2단 가수분해물의 항균성은 관측되지 않았다. 항산화능은 일단 가수분해물의 IC50(1.16 mg/mL)에 비하여 Alcalase, pepsin, trypsin 및 Flavourzyme으로 재가수분해한 이단 가수분해물의 IC50(Alcalase 가수분해물:1.19 mg/mL, pepsin 가수분해물: 1.21 mg/mL, trypsin 가수분해물: 1.19 mg/mL 및 flavourzyme 가수분해물: 1.20 mg/mL)의 경우 오히려 낮았고, Neutrase로 재가수분해한 이단 가수분해물의 IC50(0.94 mg/mL)의 경우 높았다. 2단 분해시 fraction 22-38번 사이의 고분자 획분(29-150 kDa)은 거의 분해되어 fraction 60-72번 사이의 저분자 획분 (6.5 kDa 이하)으로 이행되었으며, 아울러 저분자 획분에서도 보다 분해가 진행이 되어 획분의 감소가 두드러지게 나타났다. 가수분해물의 유리아미노산 함량은 2단 가수준해에 의해 증가하였다.
제 4 절 효소 가수분해물의 극성 기능성 물질의 정제와 구조
상업적으로 많이 사용되고 있는 효소를 바탕으로 1,2차 가수분해를 하였고, 효소 반응 시간에 대한 영향은 0 - 120 분 동안 20 분 간격으로 실험하였을 때 60 분에서 가장 높은 활성을 나타났으며, 효소 농도에 대한 실험은 0.5, 1, 1.5, 2, 5%를 첨가하였으며, 최적농도는 1% 였다. 반응온도는 30 - 70℃ 까지 10℃ 간격으로 실험을 수행 하였을 때 최적 온도는 40℃였다. 일반적인 수도수를 가하여 마쇄하여 가수분해한 것 보다 3% NaCl 을 가하여 마쇄하여 가수분해한 것은 염용성 단백질을 추출하여 더 많은 단백질을 얻을 수 있었다. TGase를 처리하여 효소가수분해물과 일반 효소가수분해물을 5 kDa 이상과 이하로 DPPH radical scavenging activity의 EC50를 측정하였다. 5 kDa 이하가 활성이 높았다. 지질 산화를 위한 산화 시간을 결정하기 위하여 시료 대신에 탈이온수를 사용하여 측정한 결과 60℃에서 linoleic acid는 약 72 시간의 유도기를 거쳐 급속히 산화하는 것으로 나타났다. 그러나 1000 Da 이하의 peptide는 168시간까지 85% 이상의 항산화 활성을 것으로 나타나 대체로 저분자의 peptide가 강력한 항산화 효과를 유지하는 것으로 나타났다. ACE 저해능은 TGase를 처리한 효소가수분해물이 80 %의 저해율로 다른 가수분해물에 비해 다소 높은 것으로 나타났다. 가수분해물의 환원력의 효과는 없는 것으로 사료된다.
제 5 절 효소 가수분해물의 비극성 기능성 펩티드의 구조 결정
TGase와 효소를 이용한 굴 가수분해물을 한회여과장치를 이용하여 1 kDa 이하로 분리한 후 이를 size-exclusion column을 이용하여 분자량 별로 ACE 저해활성을 측정하였으며 500 Da 부근에서 높은 활성을 나타내었다. 이러한 높은 ACE저해활성을 나타내는 물질이 peptide에서 기인할 것으로 판단하여 이를 정제하여 peptide의 아미노산 구조를 밝히기 위해 reverse-phase HPLC column과 ion-exchange HPLC column을 이용하여 정제한 결과 0.1 % TFA를 포함하는 8 % CH3CN에서 단일 peak를 나타내었으며 1/10의 양을 취해 ACE저해활성을 측정한 결과 약 38 % 의 저해활성을 보였다. 이렇게 정제된 peptide의 아미노산 서열을 Edman 분해법으로 분석한 결과 분자량이 500 Da 이하인 Ile, Leu, Val, Ala, Asp, Gly, Pro, Asn과 같은 아미노산 잔기로 이루어져 있는 것으로 나타났으며 ESI-MS를 이용하여 peptide의 분자량을 측정한 결과 이와 일치하는 결과를 얻을 수 있었다.
제 3 장 새로운 펩티드의 생리학적 기능성
제 1 절 새로운 펩티드의 세포 독성과 간 기능 보호 효과
두 종류의 한약제와 효소가수분해물의 최적 혼합물(HepaCae)의 간세포 독성과 간기능보호효과를 조사하였다. O, I 및 효소 가수분해물은 간 세포에 대하여 독성을 보이지 않았다. 효소 가수분해물인 TGPN과 3PN은 간 세포 독성 회복 효과를 보이지 않는 반면, HapaCare는 대조군에 비하여 50 ug/mL 및 200 ug/mL 농도에서 각각 116와 128%의 유의적인 간세포 독성 보호 효과를 보였다. HepaCare를 경구 투여하여 20일이 경과한 후 GOT(AST)와 GPT(ALT) 활성은 정상적인 수치로 회복되었으며, -GT는 대폭 감소하였다. 그러나 LDH의 감소폭은 크지 않은 것으로 나타났다. 이 같은 결과는 HepaCare는 급만성 및 알코올성 간염의 치료에 유의적인 효과가 있음을 제시한다. 그러나 정확한 판단을 위해 다양한 간 및 담도계 기능 이상과 관련한 다양한 biomarker의 확인이 필요하다.
제 2 절 굴 가수분해물과 굴 가수분해 조합물이 SD-렛트 혈청과 간 균질물에 미치는 영향
기본 식이를 제공한 대조군의 단백질 함량은 45.16±2.25 mg/ml serum이었으며, TGPN 굴 가수분해 물과 굴 가수분해 물에 허브를 첨가한 혼합시료인 Herb M- 100 식이 섭취군은 48.34±3.24 ㎎/ml serum으로 7% 증가하였고, Herb M- 200 식이 섭취군의 단백질함량은 49.24±2.55 ㎎/ml serum으로 대조군에 대비하여 9.0%의 유의적인 증가를 보여주었다. 그렇지만 TGPN-200 식이 섭취군과 PN-NaCl 가수분해물 식이 섭취군의 경우는 단백질 함량이 오히려 저하되었다. 총 콜레스테롤의 함량은 모든 굴 가수분해 식이군에서 총 콜레스테롤 함량이 감소하는 경향을 보여 주었다. 특히 TGPN-200과 Herb-M-200 식이군에서 79.04±12.50㎎/㎗과 84.38±16.20 ㎎/㎗로 각각 29.9%와 24.1%의 유의적인 감소 효과가 있었다. 대부분의 굴 가수분해 식이 섭취 그룹에서 HDL- 콜레스테롤의 함량이 저하하는 경향을 보여주었다. TGPN 가수분해물 식이군과 TGPN 가수분해 식이군에 허브를 첨가한 허브 혼합 식이군에서 LDL- 콜레스테롤의 혈중 농도가 감소하였다. 중성지질의 혈중 함량은 모든 굴 가수분해 식이군에서 감소하는 경향을 보여 주었다. 대조군의 경우 수퍼옥시드 라디칼의 함량은 266.23±60.96 nmol/g protein이었으며, TGPN-100, TGPN-200의 경우 246.49±125.20 nmol/g protein 와 173.82± 75.05 nmol/g protein으로 대조군에 비해 7.4%와 34.7%의 감소 효과가 있었다. 히드록실 라디칼 (hydroxyl radical) 의 함량 변화는 대조군의 경우 2.34±0.20 nmol/㎎ protein (100%)으로 TGPN 굴 가수분해 식이군과 Herb M-200 그룹에서는 감소하였으나, PN 굴가수분해물 식이군은 약간 증가하였다. 그렇지만 모든 굴가수분해물 식이군에서 대조군과 비교시 어떤 유의적인 증가 및 감소 효과는 찾지 못했다. 대조군의 카르보닐기 함량은 13.47±1.19 nmol/㎎ protein (100%)이었고, TGPN-200(11.98±2.34 nmol/㎎ protein, 88.9%)과 Herb M-200(12.53±3.10 nmol/㎎ protein, 93.0%) 에서 가장 큰 폭의 카르보닐기 생성 저해 효과를 볼 수 있었다. 혈청의 과산화 지질 함량은 TGPN 굴 가수분해물 식이군과 Herb 혼합식을 섭취한 그룹에서 감소하는 경향을 보여 주었다. 대조군 간균질액중의 SOD 활성은 3.44±0.43 unit/㎎ protein이었으며 PN-100과 PN-200 그룹에서는 그 활성이 농도가 2배로 증가하였을 때 그활성도 증가하였고, 특히 PN-200은 19.0%의 통계적인 유의성도 나타났다. 카탈라제 활성은 대조군 10.26±0.10 nmol/㎎ protein/min (100%)에 비해, TGPN-100과 TGPN-200에서 각각 0.30±0.10 nmol/㎎ protein/min와 0.31±0.11nmol/㎎ protein/min으로 14.5%와 20.0%의 활성증가가 있었다. Herb-100과 200에서도 20%가 넘게 활성이 증가하였으나 유의성은 없었다. 그리고 PN 굴가수분해물은 가장 높은 카탈라아제의 활성증가를 보여 주었다.
제 4 장 굴 부분 가수분해물의 응용
제 1 절 기능성 고형 제품의 제조와 생리적 기능성
굴 가수 분해물을 함유한 기능성 고형 환의 제조를 위해 각 성분의 특성을 측정하였다. Radical 소거능은 인진이 가장 높은 활성을 보였으며, 굴 가수 분해물, 인진추출물은 지금까지 알려진 항산화제보다 높은 활성을 나타내었다. 세 물질을 혼합하여 환을 제조하였을 때, 굴 가수분해물, 인진추출 건조물, 오미자추출 건조물의 함량이 높을수록 radical 소거능도 증가하였으나, 점착력이 증가하여 과량 함유할 경우 성형이 어려웠다. 따라서 기능성 환의 제조 시 밀가루 3.5 g, 인진추출건조물 0.75 g, 오미자추출 건조물 0.75 g, 굴 가수 분해물 2.74 g을 최적 배합비로 선정하였다.
제 2 절 혼합물 실험계획법에 의한 효소가수분해물질과 한약추출물의 최적 혼합비 분석
굴 가수분해물질을 이용하고자 인진 및 오미자 추출물을 혼합하였고, 통계적 모델링과 분석을 통하여 혼합비에 따른 액상의 음료의 최적화를 이루었다. 수학적인 canonical model과 trace plot을 이용하여 액상음료의 중요한 요소로 작용하는 antiradical activity와 관능평가는 각 성분들이 독립적으로 작용하는 선형모델화를 보여주었다. 그 외 색도와 점도는 상호간의 작용을 일으키면서 반응에 기여하는 것을 보여주었다. 굴 가수분해물질 및 한약 추출물들을 이용하여 최적의 액상의 음료 배합비를 결정하기위해, 각 반응의 canonical 계수를 이용하여 수적 최적화를 한 결과, 굴 가수분해물질 3 %, 인진추출물 3.83 %, 오미자추출물 8.17 %로 나타났고 추후 최적화된 제품을 이용하여 항산화성과 같은 기능성 여부를 계속할 예정이다.
제 5 장 유용균주로부터 생리활성물질 대량 생산 및 활용
제 1 절 해양유래의 유용균주의 탐색
해안 뻘에서 protease inhibitor를 생산하는 해양방선균을 분리한 후 저해력이 가장 우수한 균주 C12을 최종 선별하였다. 최종 선별된 균주 C12의 보다 정확한 동정을 위하여 16S rDNA로 동정한 결과 Streptomyces thermocarboxydus C12로 동정하였다. 배양학적, 형태학적, 생리학적 형태로서는 전자현미경에서 긴 원주모양의 형태를 보였으며, 배양과정 중 ISP 9 배지를 제외한 모든 배지에서 성장하였다. 또한, 운동성이 없으며, 그람양성에 포자를 형성, 표면은 smooth type이다. Catalase 양성반응, starch, gelatin 및 casein에 대해서 분해한 것을 확인되었으며, 9.0%의 NaCl 및 50℃까지 성장하였다.
제 2 절 탐색 균주의 생물활성 및 생리활성 물질 생산 최적 조건 검토
해안 뻘에서 단백질 분해효소 저해력이 우수한 균주 Streptomyces thermocar -boxydus C12를 이용하여 최적 저해제 활성조건을 검토하였다. 그 결과 온도와 초기 pH는 40℃, 8.0에서 높은 것으로 나타났으며, 탄소원의 영향에서는 1.6% (w/v) galactose에서, 유․무기 질소원으로는 0.5% (w/v) proteose peptone에서 저해활성이 가장 높게 나타났다. 염농도로는 1% (w/v) NaCl, 금속이온은 1 mM LiCl을 첨가하였을 때 저해활성이 증가하였다. 선정된 최적배양조건에서 배양한 결과, 84 hr 배양시 저해활성은 균주 최적배양 전의 저해율이 최대로 나타났다.
제 3 절 생리활성 물질의 분리․정제 및 이화학적 특성
정제는 ammonium sulfate, DEAE Sepharose CL-6B, Superdex 200 column으로 specific activity 35,620.6 U mg-1, yield 20.7 %, 119.9 fold로 정제하였다. Native 전기영동에서는 단일 band로 나타났다. SDS-PAGE로 측정한 저해제의 분자량은 33.1 kDa monomer이었고, 등전점은 4.4에 해당하였다. 저해제의 물리․화학적 특성에서는 70℃에서 30 min 전처리한 후에 60%의 저해활성을, pH 6~10범위에서는 80% 이상의 저해활성이 유지되었다. Subitilisin에 대한 저해형태는 비경쟁적 저해로 나타났으며, Km값과 Vmax는 각각 2.67mg. 15.33 unit였다. protease inhibitor의 N- terminal sequence는 DPSALYAPSALVLTVGKGVSAT였다. 이 Sequence는 Streptomyces albogriseolus에서 생산되는 Streptomyces subtilisin inhibitor (SSI)와 일치하는 것으로 조사되었다. Streptomyces thermocarboxydus C12에서 생산된 proteianse inhibitor의 아미노산 조성분석의 경우 Ser, Ala의 함량이 높았으며, Met, Ile, His, Lys의 함량이 낮은 것으로 조사되었다. protease inhibitor를 식품급 저해제로 적용 가능한지 조사하였다. 비교 대조군은 상업적 저해제로 사용되는 pork plasma protein과 egg white type p-39이며, 단백질 농도로 subtilisin에 대해 저해율을 비교하였다. 그 결과 subtilisin에 대하여 pork plasma protein과 egg white type p-39보다 1,648.3, 571.6배의 저해효과가 있었다.
제 4 절 굴 가수분해물을 이용한 생리활성 물질의 대량생산 검토 및 해양 유래 미생물에 대한 적용
Streptomyces thermocarboxydus C12의 최적 회분배양 조건은 굴가수분해물 1.5 %, glactose 1.5 %, 100 rpm, 1 vvm, 접종량 1 %에서 335 unit로 원배지에 비해 약 1/3 정도 낮은 protease 저해활성을 보였다. 반면에 Pseudomonas aeruginosa BYK-2(KCTC 18012P)의 활성은 굴가수분해물을 첨가한 배지가 816 unit로 기존 배지에 비해서 약 2배 정도 높은 결과를 나타내었다. 이는 균주마다 이용하고자 하는 질소원 내의 아미노산 함량 차이에 따른 특이성을 나타내는 것으로 사료된다. 그러므로 균주의 생육조건에 따라 선별적인 질소원을 선택하는 것이 효과적일 것으로 판단된다. 이런 결과를 바탕으로 2007년 9월 6일자로 ‘스트렙토마이세스 서모카르복시두스 씨 12 (Pseudomonas aeruginosa BYK-2(KCTC 18012P)가 생산하는 섭티리신 유사 단백질 분해효소 저해제(Subtlisin like protease inhibitor) 및 그의 제조방법’ (특허출원번호 : 10-2007-0063517호)를 출원하였습니다.
제 6 장 기능성 음료 제품의 시장 조사
기능성 펩티드의 우리나라 시장 규모는 점차 확대되고 있으며, 직접적인 사용이 가능한 시장 규모는 년간 1176억읜인 것으로 추정되며, 미생물 대량 배양 배지를 위한 pepton시장의 규모는 수입액으로 환산할 때 년간 약 12억원으로 추정된다. Ex분 추출을 위한 굴을 사용할 때 원료 단가는 거의 없으며, 미이용 IGF 굴과 Ex-분 추출 후의 잔사굴을 이용한 산가수분해물과 펩티드 제조원가는 각각 8,018-17,600원과 7,418-17,000원의 범위였다. 일반적인 펩티드 제품에는 식이당 약 0.2-8.0 g이 첨가되며 1 kg을 사용하여 제품을 제조하는 경우 125개의 제품을 생산할 수 있기 때문에 다른 부원료를 포함하여도 충분한 경제성이 있을 것으로 판단하였다. 바이오 관련 제품의 산업화는 연구 보다는 판매가 큰비중을 차지하고 있다. 현재 우리나라에서 개발된 대부분의 기능성 식품들은 방문 판매와 홈쇼핑을 통한 판매를 위주로 하고 있기 때문에 초기 시장 진입에 막대한 금액이 소요된다. 따라서 본 연구결과물의 초기 시장 진입은 기능성 대량 생산 제품에 대한 전국적 네트워크를 구축하고 있는 업체와 연계하는 것이 가장 가능성이 큰 것으로 보인다.

The specific activities of transglutaminase from water extract of 8 species fishshells were compared, and transglutaminase of oyster was purified. Trnasglutaminase was purified by 20-80% saturated ammonium sulfate fractionation, DEAE-Sepharose ion-exchange and Superdex 200 prep grade gel chromatography. The purity and yield of final purified transglutainase were 1895 fold and 1.9%, respectively. The optimum pH and temperature of transglutaminase were 9.0 and 45℃ against N,N'-dimethylated casein as substrate. The half activity was lost after heating at 55℃ for 30 min. The activity of purified transglutaminase depended on Ca2+ ion, and showed the highest in 10 mM CaCl2. The activity was decreased up to 10 mM NaCl ,and not significant difference in the range of 50 -200 mM NaCl.
Section 2 Chemical Composition and Properties of Protease Inhibitor from Oyster Shell
The protease inhibitor was investigated from oyster shell. The crude protein was 0.8%, and calcium oxide composed of 97.15% in calcium acetate from oyster shell. The fraction under 10 kDa had higher protease inhibitor compared with the fraction above 10 kDa in acetic acid soluble fraction. It was more effect against bromelain inhibition. The antimicrobial and hepatic toxicity were not observed in protease inhibitor fraction and calcium acetate. The acetic acid soluble fraction did not protect ultra-violet ray. The results suggested that partial purified acetic acid soluble fraction had a potential application as food ingredient for protease inhibition.
Chapter 2. Preparation and Functional Properties of Acidic and Enzymatic Hydrolysates from Oyster
Section 1 Preparation and Functional Properties of Acidic Hydrolysate from Oyster
The objective of this work was to manufacture acid hydrolysate based on the functionality such as anti-oxidative effect and angiotensin I converting enzyme (ACE) inhibitor. The optimum conditions for maximum yield were 10 N for HCl concentration and 22 hr for hydrolysis time. The main factor for acid hydrolysis was hydrolysis time, HCl concentration and hydrolysis temperature in order. The protein content of acid hydrolysate and desalted acid hydrolysate was 6.5% and 14.0%, respectively. The peptide size in acid hydrolysate was in the range of 400-2500 dalton. The amino acid of acid hydrolysate was high in order of lysine, leucine, tyrosine, histidine, glycine and valine. Threonine, serine and proline was not detected. Acid hydrolysate did not showed toxicity on HepG2 cells up to 200 mg/mL. DPPH radical scavenging activity was showed in acid hydrolysate, and EC50 value was 0.28 mg/mL. The anti-oxidative effect for linoleic acid was not significant difference in the range of 0.1-0.0 mg/mL, and stronger with decreasing peptide size. The fraction of desalted acid hydrolysate with 500-1000 showed high ACE inhbition compared with other fraction. The results suggested that the desalted acid hydrolysate had a potential application as functional ingredient. However, we need more the safety data of acid hydrolysate.

Section 4 Purification and Structure of Polar Substance from Oyster Hydrolysate
The objective of this work was to purify and investigate the functional polar substances from oyster enzymatic hydrolysates. The hydrolysates from oyster were prepared by two step hydrolysis using commercial protease such as Protamex and Neutrase. The optimum conditions for preparing functional substances were 60 min for reaction time, 1% for protease concentration, and 40℃ for temperature. The protein yield was higher in 3% NaCl medium than distilled water. DPPH radical scavenging activities of hydrolysates were higher in small molcular size (< 5 kDa). The linoelic acid had oxidative induced-time of 72 hr at 60℃. however, low molecular peptides extended the induced time up to 168 hr, and had antioxidant effect above 85%. TGPN hydrolysate inhibited the angiotensin-I converting enzyme up to 80%. Most hydrolysates did not have the reducing effect.
Section 5 Purification and Structure of Nonpolar Peptides from Oyster Hydrolysate
The objective of this work was to purify and investigate the functional nonpolar peptides from oyster enzymatic hydrolysates. The nonpolar peptides of TGPN were purified by ultrafiltration (1 KDa), size exclusion chromatography, reverse-pahse and ion-exchange HPLC. TGPN hydrolysate had high angiotensin-I converting enzyme inhibition below 500 Da molecular size. The purified peptide showed single peak in 8% CH3CN containing 0.1% TFA, and 38% inhibition effect. The amino acid composition of purified peptide composed of isoleucine, leucine, valine, alanine, aspartic acid, glycine, proline and asparagine by Edman sequence. The molecular weight of some peptides was equal to the results of ESI-MS for peptide sequence.
Chapter 3. Physiological Function of New Peptides from Oyster Hydrolysate
Section 1 Toxicity and Protective Effect of New Peptides on HepG-2 Cell
The objective of this work was to investigate the effect of two herbs and optimum formulation product with hydrolysate(HepaCare) on toxicity and cytoprotection of THA-induced toxicity for HepG-2 cell. The toxicity for HepG-2 cell did not find in O and I extracts, and enymatic hydrolysates. TGPN and 3PN did not show the cytoptoective effect. However, HepaCare revealed survival capacity of 116% for 50 ug/mL and 128% for 200 ug/mL, respectively, compred to control. HepaCare had significantly hepatoprotective effects lowering the activity of GOT(AST), GPT(ALT) and -GT after oral administration. This results suggested that HepaCare has significant hepatoprotective effect for acute chronic, and alcoholic hepatitis. In addition, HepaCare played a protective role against lipid peroxidation by TGPN and two herbs.
Section 2 Effect of Oyster Hydrolysate and Herb Mixture on Serum and Liver Homogenate of SD-Rat
The effect of enzymatic hydrolysate from oyster and its formulation with herb on SD-rat was investigated. The protein content with basic diet was 45.16±2.25 mg/ml for serum, and increased up to 7% for HepaCare-100 and up to 9.0% for Herb-200, significantly. compared with control group. However, The protein content of TGPN-200 and PN diet group was decreased, Total cholesterol was decreased in all diet group compared with control group. Especially, the total cholesterol was significantly decreased up to 29.9% for TGPN-200 and 24.1% for Hapacare-200, respectively. Enzymatic hydrolysates decreased HDL-cholesterol, TGPN and HapaCare group decreased LDL-cholesterol of serum. The neutral lipid in serum was decreased in all enzymatic hydrolysate. Superoxide radicals of TGPN-100 and TGPN-200 groups was decreased up to 7.4% and 34.7%, respectively, compared with control groups. Hydroxy radicals in hydrolysate and HapaCare group were not significant difference compared with control group. TGPN-200 and HepaCare group decreased greatly carbonyl content in serum. The lipid peroxide in serum was decreased in TGPN and HepaCare groups. PN-100 and PN-200 groups increased superoxide dismutase activity. Especially, PN-200 group showed significant difference compared with control gorup, and increased superoxide dismutase activity up to 19.0%. The catalase activity was 10.26±0.10 nmol/㎎ protein/min for control groups, 0.30±0.10 nmol/㎎ protein/min for TGPN-100, and 0.11 nmol/㎎ protein/min for TGPN-200 group. PN group increased the catalase activity significantly.
Chapter 4. Application of oyster hydrolysate
Section 1 Manufacture of the Functional Solid Tablet Using Enzyme Hydrolysate from Oyster and Herb Extracts
The objective of this work was to manufacture the solid tablet based on the functionality with oyster hydrolysate and extracts from plants such as O and I herb. The oyster hydrolysate was obtained by polymerization with transglutaminase, followed by hydrolyzation with Protamex and Neutrase, while O and I herb were extracted with water at 100℃ for 5 hr, and powdered with freeze dryer. Wheat powder was used as basis of the tablet forming. The functional properties such as radical scavenging activity and angiotensin-I converting enzyme(ACE) inhibitory activity were determined before and after formulation of the tablet. I extract showed the highest radical scavenging activity. On the other hand, the oyster hydrolysate and O extract showed higher radicla scavenging activity than other materials such as vitamin C and tocopherol. According to the ACE inhibitory activity and forming condition, the optimum levels of oyster hydrolysate, O extract, I extract and wheat powder were 2.74g, 0.75 g, 0.75 g and 3.5 g, respectively. Each functional property was verified before and after forming the tablet, and optimum mixing ratio for the solid tablet was provided based on the manufacturing process and functionality.
Section 2 Manufacture of the Functional Drink with Enzyme Hydrolysate from Oyster and Herb Extract by Using the Mixture Design
The objective of this work was to manufacture the functional drink with oyster hydrolysate and extracts from plants such as O and I herb by using mixture design and to provide the optimum ratio for the drink using the response surface methodology (RSM). The oyster hydrolysate was obtained by polymerization with transglutaminase, followed by hydrolyzation with Protamex and Neutrase, while O and I herb were extracted with water at 100℃ for 5 hr. Interaction effects of these mixtures were investigated by modified distance based design and analyzed by regression canonical model, and trace plot. The optimization of mixture ratio was made by statistical modeling. Antiradical activity and sensory properties which are the important target constraints in drink showed linerar canonical form, while color and viscosity of drink showed nonlinear canonical form indicating the highly interaction among mixtures. The reponse trace plot revealed that antiradical activity, sensory properties, color and viscosity were quite sensitive to the drink blending. According to the RSM, the optimum formulation of drink was 3% of oyster hydrolysate, 3.83% of I extract, and 8.17% O extract. This work shows the interaction effects of each mixture and provides the optimum ratio of the functional drinks for antiradical activity, ACE inhibition, color and viscosity.
Chapter 5. Mass Production and Application of Bioactive Materials from Marine Bacterium
A marine bacteria which produced the subtilisin-like proteinase inhibitor was screened from the sediment in the southern sea of Korea. The cultureal, morphological and physiological characteristics of an isolated strain were investigated for identification. Cultural characteristics based on ISP(International Streptomyces Project) were as follows: white and gray aerial mycelium and good growth on various medium. Also, the strain did not produce the soluble pigment. Also, this strain could be grown up to 9% salt concentration. Morphological and physiological characteristics showed cylindrical spore chain and smooth spore surface by SEM(Scanning Electron Microscope). A phylogenetic analysis of the 16S rDNA provided a clue that the isolated strain was actually a member of the genus Streptomyces, because the determined sequence exhibited a higher homology with Streptomyces thermocarboxydus.
A subtilisin-like proteinase inhibitor has effectively inhibited the activity of subtilisin and proteinase K by complexing with the enzyme in the ratio of 1 : 1. The inhibitor has no effect on the trypsin and collagenase.

Natural organic nitrogen sources such as poly peptone and proteose peptone were good for the production of inhibitor, however beef extract, urea, casamino acid and inorganic nitrogen sources such as (NH4)2SO4 and NH4Cl were poor. Optimal culture conditions for inhibitor production were composed of 1.6% galactose, 0.5% proteose peptone, 1% NaCl and 1 mM Li+, respectively. The optimal temperature and initial pH for the subtilisin-like proteinase inhibitor activity were 40℃ and pH8.0, respectively. The subtilisin-like protease inhibitor from Streptomyces thermocarboxydus C12 showed a maximum inhibitor activity after the cultivation for 60 h under the optimized medium.
A subtilisin-like proteinase inhibitor produced by Streptomyces thermocarboxydus C12 was purified by ultrafiltration, ammonium sulfate precipitation, DEAE Sepharose CL-6B and Superdex 200 column chromatography.
The final inhibitor was purified 119.9 fold, with a yield of 20.7%. The inhibitor has a monomer with a molecular weight of 33.1 kDa estimated by SDS-PAGE. Also, the inhibitor showed the one band in native-gel PAGE. The isoelectric point was 4.4. The inhibitor activity was stable in the range of pH 6.0~10.0 and temperature 30~60℃, respectively. The purified inhibitor was slowly activated by Mg2+ and K+. The N-terminal sequence of subtilisin-like proteinase inhibitor was determined as DAPSALYAPSALVLTVGKGVSAT. The sequence (residues from 1 to 23) was 100% identified with the Streptomyces subtilisin inhibitor(SSI) produced in the Streptomyces albogriseolus. The Km and Vmax of the inhibitor were calculated to be 2.67 mg and 15.3 unit, respectively. The inhibitor showed a noncompetitive inhibitor mode for subtilisin. As a result of a amino acid compositon analysis, the purified inhibitor had a high content of serine and alanine. The degree of inhibition of subtilisin-like proteinase inhibitor was much greater than that of food inhibitor such as bovine plasma, pork plasma and egg white protein.
