Ⅳ. 연구개발 결과

미더덕(Styela clava) 영양성분의 계절적 변화에 대하여 평가하기 위하여 2005년 1월부터 5월까지 2개월 마다 거제와 통영의 양식장에서 시료를 채취 하였다. Glutamic acid, aspartic acid, arginine, leucine 및 lysine 등이 가장 풍부한 아미노산 성분이었다. 비필수 아미노산에 대한 필수 아미노산의 비율은 다른 어류와 비교하여 우수하였고, 미더덕 육 거제산의 값은 0.55~0.61, 통영산은 0.66~0.67의 범위였다. 미더덕 육에 함유된 고도불포화지방산의 함량이 높아 지방산 공급원으로 적절한 것으로 나타났다. 가장 높은 함량을 보인 지방산은 EPA와 DHA로 육 중 20.0~22.3% 및 16.5~17.9%, 껍질 중20.3~23.2% 및 15.2~18.8%가 각각 함유되어 있었다. 총 무기질 함량은 22.2~27.3%였고, Na, Ca, Mg, K 등이 육과 껍질에 풍부하게 들어있었다.
거제와 통영으로부터 채집한 미더덕 육 및 껍질의 카로테노이드 함량은 높은 값을 보였고, 3월의 시료가 가장 높았으며, 1월 시료가 가장 낮았다. GM(거제 육), GT(거제 껍질), TM(통영 육) 및 TT(통영 껍질) 시료의 평균 카로테노이드 함량은 각각 49.1 mg/100 g, 56.7 mg/ 100 g, 42.0 mg/100 g 및 50.2 mg/100 g으로 조사되었다.
육에 함유된 주성분은 4‘-hydroxyechinenone, canthaxanthin, cynthiaxanthin 및 β -carotene 이었고. 껍질에 함유된 성분은 4‘-hydroxyechinenone, canthaxanthin, cynthiaxanthin 및 halocynthiaxanthin 등 이었다.
미더덕 카로테노이드 화합물의 기능성을 파악하기 위하여 페놀 함량 및 항산화활성을 조사하였다. 모든 시료에서 낮은 함량의 페놀 값을 보였으며, DPPH 라디칼 저해활성도 낮았다. 그러나 모든 시료에서 리놀레인산에 대한 과산화 억제작용은 효과적이었고 껍질 추출물이 더 효과적이었다. 뿐만 아니라 hydroxyl 라디칼 저해활성에 대해서도 강한효과를 나타내었으며, 합성항산화제인 BHA의 IC50 값도 비슷하였다. 따라서 미더덕은 영양성분이 풍부하고 천연 항산화제로서의 가치가 있는 것으로 평가되었다.
미더덕으로부터 유용 물질을 분리하기 위하여 미더덕을 아세톤으로 추출하고 항균성을 조사하여 8개 획분에서 항균성이 있음을 확인하였으며, 그 중구상 ODS 칼럼에서 50% MeOH로 용출된 획분의 항균성 물질을 ODS 칼럼에서 함수 MeOH로 재정제하여 항균 물질 peak no. 2 218.9 mg을 부분 정제하였다. Peak no. 2는 FAB MS 분석 결과, 분자량 272로 확인되었으며, 고분해능 MS에서 C11H21SO4Na의 분자식을 확인 하였다. Proton NMR 과 carbon NMR에서 각각 이들의 C와 H를 assignment하였으며, HMBC에서 최종적으로 확인 결과, 이 물질은 4,8-dimethyl-3-nonene sodium sulfate라는 새로운 물질임을 확인하였다.
4,8-Dimethyl-3-nonene sulfate의 분리 정제는 미더덕 가식부를 3배량의acetone으로 추출하여 여과, 농축 후 hexane으로 저극성 물질 제거하고, BuOH로 추출하여 염기성 alumina에 흡착시켜 1% ammonia-MeOH(1:1)로 용출시키고 silica 칼럼에서는 chloroform-MeOH(7:3) 용매로 용출시켜 구상 ODS 칼럼에서 60% MeOH로 용출하여 ODS 칼럼에서 70% MeOH로 분리정제하는 것이 가능하다. 이 방법으로 분리 정제한 결과 17 kg의 미더덕으로 부터 4,8-dimethyl- 3-nonene sulfate 395.7 mg(수율 0.0023%)을 분리 정제하였다. Bacillus subtilis에 대한 MIC는 200 μg/mL 이었으며, 그 이외의 그람양성, 음성 병원성 균 및 효모, 곰팡이에 대하여 이 보다 약한 활성을 나타내었다.
HPLC에 의한 정량 분석은 ODS 칼럼에서 0.01 M 인산칼륨 완충액(pH 5.0)-MeOH(25:75)을 용매로 0.8 mL/min 유속으로 흘리며 215 nm에서 모니터링 하여 정량 분석이 가능하며(검출 한계 : 10 ng), 전처리는 80% MeOH로 추출하여 sep-pak 칼럼에서 70% MeOH로 용출하여 분석할 경우 미더덕가식부에 29 μg/g 함유되어 있었다. 항균 활성은 24시간 이후부터 감소하여4일 후에는 활성이 완전히 소실되어 살균보다 정균 효과밖에 없었으며, 100℃에서는 6시간까지, 121℃에서 15분 가열하여도 안정하였다. pH 3~11 범위에서는 24시간 두어도 안정하였으며, 마우스에 대한 치사 독성은 LD100이
1.0 g/kg 이었다.
미더덕 껍질에서 분리한 GAGs의 세포독성을 시험한 결과 미더덕 1은 7.3mg/mL, 미더덕 2는 9.7 mg/mL, 미더덕 crude는 7.0 mg/mL의 IC50 값을 보였다. 이를 바탕으로 평균적인 IC50 값의 1/10 농도를 기준으로 각각 0.75mg/mL, 1.5 mg/mL, 2.0 mg/mL의 농도를 사용하여 NF-κB 형광면역활성을 검색하였다. 그 결과 2.0 mg/mL의 농도에서 NF-κB의 활성이 억제되는 것으로 나타났다. 세포핵에서의 NF-κB의 발현을 관찰한 결과 NF-κB의 핵 내로의 전이를 억제하는 것으로 나타났으며, 염증관련 인자인 iNOS, ICAM-1 및 VCAM-1 등의 변화를 조사한 결과 미더덕 1과 멍게 3 시료에서 단백질 발현을 억제하는 것으로 나타났다. 염증반응을 조절하는 p38, JNK, ERK 대사과 정에서의 단백질 변화를 조사한 결과 p38, p-p38, ERK, p-ERK, p-JNK의 발현에 대해서는 작용이 없었으나 JNK의 발현에는 미더덕 1과 멍게 3 시료에서 효과를 나타내었다.
만성염증은 암과 밀접한 관계가 있는 것으로 보고되고 있다. 특히 최근의 연구결과에 의하면 VCAM과 COX-2와 같은 염증인자들은 암의 유발과 전이에서 매우 중요한 작용을 하는 것으로 보고되고 있다. 따라서 이러한 염증인자들의 발현억제는 염증 및 암 예방에 있어서 중요한 작용을 할 것으로 사료된다. 본 실험의 결과를 종합하여 보면 멍게 껍질로부터 추출한 GAGs는 p38과 Akt의 pathway를 조절함으로써 NF-κB의 활성화를 차단하여 TPA에 의한 VCAM과 COX-2의 발현을 억제하였다. 따라서 멍게로부터 추출한 GAGs를 천연적인 항염증 치료제로 개발할 수 있을 것으로 사료된다.

Changes in the nutritional composition of Mideodeok (Styela clava) harvested during the months of January, March and May 2005 were evaluated. Mideodeok was found to be a good source of essential amino acids, particularly for glutamic acid, aspartic acid, arginine, leucine and lysine. Essential amino acid ratio to non-essential amino acids were comparable to many fish species, with values ranging at 0.55-0.61 and 0.66-0.67 for Geoje muscle (GM) and Tongyeong muscle (TM), respectively.
Mideodeok seemed to be a very suitable source of important fatty acids as it contained high levels of polyunsaturated fatty acids. EPA (20:5n-3) and DHA (22:6n-3) were the most dominant fatty acids, with values ranging at 20.0-22.3% and 16.5-17.9% for muscle, and 20.3-23.2% and 15.2-18.8% for tunic, respectively. Mideodeok also contained rich supplies of essential minerals ranging at 22.2-27.3% of the dry matter, mainly composed of Na,Ca, Mg, and K.
2. Antioxidant activities of Mideodeok carotenoids
Carotenoids were found in high levels in both muscle and tunic samples, with the highest values observed in March and lowestin January.
Average values in GM, GT, TM, and TT samples were 49.1mg/100g, 56.7 mg/100g, 42.0 mg/100g, and 50.2 mg/100g, respectively. The most dominant carotenoids in muscle were 4’-hydroxy-echinone, canthaxanthin, cynthiaxanthin, and β-carotene. On the other hand, tuniccarotenoids contained high levels of 4’-hydroxy-echinone, cynthiaxanthin, canthaxanthin, and halocynthiaxanthin. Crude mideodeok carotenoids were tested for phenolic content and antioxidant activity. All samples have low phenolic levels and exhibited weak DPPH radical scavenging activity.
However, they were found to have strong inhibitory effects against linoleic acid peroxidation, especially the tunic extracts. They also exhibited strong hydroxyl radical scavenging activities, with IC50comparable with those of BHA. Results of this study indicate that mideodeok, aside from being rich in nutrients, could be a potential source of natural antioxidants.
3. Mideodeok flavor and reaction flavors
Flavor components of Mideodeok extracts were evaluated using GC-MS and Maillard reaction of the extracts. Major flavor components of extracts include 17 alcohols, 6 acids, 5 aldehydes and 6 hydrocarbons by the simultaneous distillation and extraction (SDE). The affluent components was aldehydes and alcohols. The solid phase micro-extraction (SPME) method was adopted to extraction of reaction Maillard flavor in extracts-sugar- amino acids system. The major components include 9 alcohols, 1 acid, 1 aldehyde, 1 hydrocarbon, 1 ester, 1 amine and 2 ketones and aldehydes was key components in this model systems.
4. Effect on the growth and heamotological factors on fishes of extracts
Effect of muscle fluid concentrates from Styela clava on fish diets were investigated in rainbow trout (Oncorhynchus mykiss) and rockfish (Sebastes schlegeli). Fishes were fed on one of the isonitrogenous (48%) and isolipic (20%) feed containing 5 to 20% of muscle fluid concentrates for 8 week.
Hematological parameters such as hemoglobin, hematocrit, albumin, glucose, total bililubin, triglyceride and glutamic pyruvate transaminase of the rainbow trout fed on the diets varied but no specific trend became apparent. However, glutamic oxaloacetic transaminase value was significantly higher than normal fishes. Therefore, concentrated ascidian fluid which is normally discarded can be combine with commercial diets for the normal growth of fishes.
5. Extraction, purification and evaluation of anti-microbial components
Different fractions of the Midedeok extracts were purified and evaluated for anti-microbial components. Anti-microbial activity was observed on peak No. 2 and the structure was elucidated using FAB MS spectrophotometer, 1H NMR spectrum, 13C NMR spectrum, 1H Detected Single Quantum Coherence (HSQC) spectrum and 1H-1H (Corelation spectroscopy (COSY) spectrum. Appropriate extraction solvents and production processes were evaluation for the mass production of the anti-microbial components, which included liquid-liquid partition, column chromatography, and HPLC analysis. The anti-microbial activity of the fraction was quantified; and its MIC and inhibitory activity on various microbes, sustainability, stability on pH and heat treatment and acute toxicity in mice was evaluated.
6. Extraction and purification of glycossaminoglycans (GAGs)
Proteoglycans combined GAGs with protein. These molecules containing a repeating disaccharide unit were included amino sugar such as N-acetylgalactosamine or N-acetylglucosamine and uronic acid (glucuronic or iduronic acid). Optimum extraction condition of GAGs from Mideodeok tunic was first extract method at 105℃ heat in ion water containing 1/60 M sodium phosphate dibasic. Trichloroacetic acid (TCA), hydrochloride and 5-sulfosalicylic acid (SSA) were used for deprotein of the GAGs. Concentration for deprotein of GAGs were 5.0% TCA (w/v) and 10.0% HCl (v/v) treatment. The sulfate and protein contents of deproteinized GAGs contained 35.1% and 22.0% for TCA and 35.4% and 18.5% for HCl, respectively. Characterization of extracted crude GAGs had similar components, amino acids, composition sugars, mineral contents compared with dermatan sulfate from mammalia. Extracted GAGs from Styela clava tunic had similar content rate of carbon, hydrogen, nitrogen and sulfur element compared with dermatan sulfate and chondroitin-6-sulfate. Crude GAGs were fractioned two peaks by ion exchange chromatography on DEAE-sepharode CL-6B.
7. Cytotoxic effects of GAGs in cells and its immunohistochemistry
There has been much effort recently to probe the long-recognized relationship between chronic inflammation and cancer. For example, epidemiological studies have shown that approximately 15% of human deaths from cancer are associated with chronic viral or bacterial infections.
Therefore, it may be a promising strategy to inhibit the inflammatory process in order to reduce the tumor incidence. In this regard, the present study was performed to investigate the potential anti-inflammatory effects of glycosaminoglycans (GAGs) extracted from ascidian tunic andto elucidate the underlying mechanism of anti-tumor promoting effects of GAGs in TPA-induced mouse skin inflammation model. GAGs inhibited TPA-induced NF-κB activation and subsequent COX-2 expression by blocking IKKβ and Akt/PKB signaling in mouse skin in vivo, thus, suggesting that GAGs from ascidian tunic may be developed as an effective natural anti-inflammatory agent in the future. Our study would lead to the development of new and improved preventive as well as therapeutic approaches against cancer in the future.
