Acanthopanax senticosus (A. senticosus), called the 'Siberian Ginseng' has been well known to be prophylactic for various diseases such as chronic bronchitis, hypertension and ischemia. A. senticosus is also known to be effective for reducing many kinds of stress or fatigue and so call it 'adatogen'
Acanthopanax senticosus (A. senticosus), called the 'Siberian Ginseng' has been well known to be prophylactic for various diseases such as chronic bronchitis, hypertension and ischemia. A. senticosus is also known to be effective for reducing many kinds of stress or fatigue and so call it 'adatogen'. In recent, Davydov et al. was well documented that the herb included various compounds such as acanthosides, eleutherosides, senticoside, triterpenic saponin, flavon, vitamins and minerals, and they are related to its diverse biological activities. Interestingly, many investigators demonstrated two contrary views on the immunomodulatory effect of A. senticosus: the stimulation and the suppression of immune responses. In fact, our data also showed that the crude extract of stem of the herb has the dual activities according to the method of extraction. These results raise a question which constituents related to each activity.
Antitumor and immunomodulatory activities of an aqueous extract (GF-1) of Acanthopanax senticosus was examined. In experimental lung metastasis of colon26-M3.1 carcinoma cells, intravenous (i.v.) administration of GF-1 2 days before tumor inoculation significantly inhibited lung metastasis in a dose-dependant manner. The i.v. administration of GF-1 also exhibited the therapeutic effect on tumor metastasis of colon26-M3.1 cells, when it was injected 1 day after tumor inoculation. In an in vitro cytotoxicity analysis, GF-1 at the concentration up to 1000 mg/ml did not affect the growth of colon26-M3.1 cells. In contrast, GF-1 enhanced the responsiveness to a mitogen, concanavalin A (ConA), of splenocytes in a dose-dependent manner. Peritoneal macrophage stimulated with GF-1 produced various cytokines such as IL-1β, TNF-α, IL-12 and IFN-g in an in vitro experiment. The macrophages obtained from the mice which were injected with GF-1 (500 mg) 3 gays before the assay showed significantly higher tumoricidal activity against tumor cells than that of the untreated macrophages. In addition, the i.v. administration of GF-1 significantly augmented NK cytotoxicity to Yac-1 cells. The depletion of NK cells by injection of rabbit anti-asialo GM1 serum completely abolished the inhibitory effect of GF-1 on lung metastasis of colon26-M3.1 cells. These data suggest that GF-1 has antitumor activity to inhibit tumor metastasis prophylactically as well as therapeutically, and its antitumor effect is associated with activation of macrophages and NK cells. To identify the immunostimulatory active molecuels in A. senticosus, we fractionated the crude proteins (GF-AS) from aqueous extract of A. senticosus and selected the glycoproteins (EN-SP) from GF-AS. To achieve anti-tumor activity, these samples were applied to experimental tumor metastasis model using colon26-M3.1 carcinoma or B16-BL6 melanoma cells in syngeneic mice, and analyzed the involvement of activation of NK cells and macrophages in its anti-metastatic effect against tumors.
Competitive direct ELISAwas carried out to establish quantitative analysis method of purified immuno-activating substance (EN-SP) from Acanthopanax senticosus (A. senticosus). Mouse antisera (anti-EN-SP) against EN-SP was generated by immunization (s.c.) of EN-SP purified from A. senticosus as an immunogen. The titer of anti-EN-SP was about 1:400, and the optimal dilution of EN-SP-HRP conjugate was 1:1000. When the standard curve was constructed by ELISA, its sensitivity was about 0.2 ㎍/㎖. The coefficient variations of intra- and interassay were 6.13~8.81% and 6.73~8.60% respectively. According to the standard curve, the concentration of EN-SP in various senticosus extracts and was only found to be 59.85 ㎍ in 10 mg of extract of the bark of A. senticosus. Similarly the immunostimulating activity to produce TNF-a or IL-12 among the various extracts of senticosus was shown to be correlation with the content of EN-SP. These results demonstrated that competitive ELISA is a convenient, fast, reproducible, and accurate method for the determination of EN-SP as a immunologically active standard substance in extract of A. senticosus.
Administration of boiled fraction of Acanthopanax senticosus (GF-2) prophylactically and therapeutically inhibited the systemic anaphylactic shock induced by compound 48/80 in mouse. GF-2 significantly inhibited the production of histamine and eosinophyl in serum from mouse by injection of compound 48/80 in a dose dependent manners. GF-2 does-dependently inhibited TNF-α production from peritoneal exudative cells activated by LPS. In addition, intraperitoneal injection of GF-1 suppressed the production of IgG1 and IgE antibody in mice immunized with a mixture of ovalbumin (OVA) and aluminium hydroxide (alum). These results indicated that GF-2 may be beneficial in the treatment of nonspecific and specific anaphylactic reaction and could be clinically applied in the treatment of allergic diseases.
Acanthopanax senticosus(A. senticosus) is a useful medicinal plant and have been used to promote robustness and as a remedy for many ailments including anti-blood-sugar duabetes, neuralgia, palsy, sex- and learning-behavior dufficulties, cancer and because of its antioxidant properties, it has been used to treat several stresses. We prepared aqueous extract GF-100 and GF-200 from Acanthopanax senticosus(A. senticosus) and GF-AS purified from GF-100 as a potent protein, which were revealed immune enhancing effects. We examined the hepatoprotective, antioxidative and alcohol detoxifying effect of GF-100, GF-200 and GF-AS in CCl4- or alcohol-induced hepatotoxicity animal model.
Hepatoprotective effect of an aqueous extract(GF-100 and GF-100) of Acanthopanax senticosus was examined. GOT, GPT, ALP and LDH activity were increased by CCl4 i.p. injection but its actitivity was reduced by GF-100 and GF-200. The adminstration of GF-100 and GF-200 also exhibited the hepatoprotective effect against alcohol-induced liver injury by lowering the level of the hepatic injury indices and lipid peroxidation.
Antioxidative activity of GF-100, GF-200 and GF-AS in vitro was evaluated by DPPH radical scavenging ability and by determining lipid peroxide(MDA).
The GF-100, GF-200 and GF-AS significantly increased DPPH scavenging activity and decreased MDA production. EC50 of scavenging DPPH radical was 6.8μg/ml compared with EC 9.8μg/ml. Antioxidative effect of GF-AS was similar to BHT as synthetic antioxidant. These results revealed strong antioxidant on the against scavenging on DPPH free radical and lipid peroxidative activity The antioxidative potency of GF-AS was similar BHA and may be effective against liver disease in which reactive oxygen species play a role as potent causative factors. We examined the inhibitory activities of GF-AS in acute carbon tetrachloride-induced liver injury in ICR mice. 1mg/kg BW of GF-AS adminstration was the most effective to prevent liver injury and ameliorated the increase in plasma GOT, GPT, LDH, bilirubin, glucose and TG, which is represent degree of liver injury. 0.5mg/kg BW of GF-AS and 5mg/kg BW of GF-AS also effective to prevent hepatic injury but 10mg/kg BW of GF-AS promoted liver injury as control. The effective dose of GF-AS revealed U shape curve from 1.0mg/kg BW. According to histochemical changes, severe hepatic lesions induced by CCl4 were reduced by the administration of the GF-AS, which were in good agreement with the results of the biochemical tests.
Catalase, SOD and GSH content were activated in the defense against oxidative cell injury by means of GF-AS being free radical scavengers. In present study, acute sublethal doses of CC4 reduced the activities of the GPx, GR and GSH and increased MDA content, whereas GF-AS reversed these effects. It is conceivable that the effect of GF-AS may be due initially to increases in hepatic antioxidative activities followed by reduction in hepatic peroxidative effects. SOD and catalase activities increased by GF-AS, suggesting that it can restore their enzyme activities or activate enzyme activity in CCl4-damaged liver tissue.
However, Treatment level of GF-AS is very important because GF-AS acted as an adaptogen. In this study, 1mg/kg BW in ICR mice is the most effective dosage to prevent CCl4-induced hepatotoxicity through antioxidative action We examined the inhibitory activities of GF-AS in chronic(SD rats) or acute(ICR mice) alcohol intake-induced liver injury. Because 1mg/kg BW of GF-AS adminstration was the most effective to prevent CCl4-induced liver injury, we treated 0.2, 0.5 and 1.0mg/kg BW in SD rats administered ethanol for 30days and 0.5, 1.0 and 2.5mg/kg BW in ICR mice adminsteded acute dosage of ethanol. In the chronic model, the level of GOT, GPT, LDH, bilirubin, r-GT, glucose, TG and albumin was significantly increased by adminstering ethanol.
There were significantly reduced in GOT and GPT by 0.5mg of GF-AS and in LDH by 0.2, 0.5 and 1.0mg of GF-AS. In the acute model, liver injury indices(GOT, GPT, Albumin, TG, Chol) were significantly decreased by GF-AS administration as a manner of dose-dependent, which was the most effective in 2.5mg GF-AS. Severe hepatic lesions induced by alcohol were reduced by the administration of the GF-AS. The level of GPx, GR and catalase was significantly reduced by adminstering ethanol and ite condition was not reversed by GF-AS supplementation. SOD and catalase activity was significantly increased in GF-AS supplemented group compared with control group. The co-administration of GF-AS with alcohol may release the toxicity of ethanol through antioxidative action but antioxidative effect of GF-AS was compensated by following long-term etahnol feeding. The activity of ADH, catalase, ALDH and MEOS did not change significantly GF-AS in chronic alcohol intake reflecting adaptation or injured liver tissue. In acute alcohol intake, plasma ethanol elearance was significantly increased by GF-AS i.p. injection as a manner of dose dependant and the most effective in 2.5mg GF-AS. ADH, ALDH and MEOS was increased by GF-AS supplementation but not consistant trends;
The effective dose of GF-AS was followed; ADH activity by 0.5mg GF-AS, ALDH activity by 2.5mg GF-AS, MEOS activity by 1.0mg GF-AS.
In conclusion, GF-AS would appear to contribute to alleviating the adverse effect of CCl4 and ethanol by enhancing antioxidant defense system and hepatoprotective and ethanol-detoxifying effect.
