연구의 목적 및 내용
1) 지구온난화에 따른 기후변화는 보균동물과 매개체의 서식조건의 변화를 일으키며,최근 국내에서 신종 병원체들이 검출되고 있음.
2) 국내에서는 절지동물 매개질병 병원체중 Bartonella, Borrelia, Anaplasma, Ehrlichia,Rickettsia, 진드기뇌염바이러스 등이 앞으로 지구온난화와 더불어 더욱 문제시 될것으로 예측됨.

4) B. grahamii 분리주를 국내 야생설치류로부터 확보하여 전자현미경적, 분자생물학적,혈청학적 특성을 밝히고자 함.
5) 국내 분리주에 대한 병원성 및 특성을 파악하고자 함.
연구결과
1) B. grahamii ITS 유전자를 대상으로 PCR을 진행한 결과, 고라니 비장에서 11(15.8)개의 B. grahamii와 9(12.9%)개의 B. schoenbuchensis가 검출되었다. rpoB 대상PCR에서는 6개의 B. grahamii가 검출됨. 설치류 비장을 대상으로 진행한 실험에서는,ITS 대상 PCR에서 124(62.0%)개의 샘플이 그리고, rpoB 대상 PCR에서는 63(31.5%)개의 샘플들이 Bartonella spp.가 검출되었음.
2) 야생설치류 혈액으로부터 2 종류의 B. grahamii 분리주가 분리되었음.
3) 분리주들로부터 16S rRNA, hemin-binding protein E (hbpE), glutamate dehydrogenase 1 (gdh 1), invasion associated protein B (ialB), cell division protein (ftsZ), citrate synthase (gltA), 60 KDa heat shock protein (groEL),rpoB 유전자 그리고 ITS region 유전자염기서열을 확보하여 계통발생학적으로 B.grahamii 임을 증명하였음.
4) KRBG28, KRBG32 분리주들은 NF-kB활성을 통하여 염증반응물질을 강력하게 유도하여 HUVEC 세포를 강하게 자극함. 또한 분리주들은 ATCC균주인 V2보다 강한 염증반응이 유도되었음.
5) 분리주를 접종한 실험마우스에서 V2를 접종한 마우스보다 조직병리학적소견상 매우 심한 병소를 일으키는 것으로 관찰되었음
6) 비글견을 대상으로 감염실험을 하였으나 개에 있어서는 병원성이 나타나지 않았음.

1) 국내 최초로 B. grahamii 균을 분리 동정하여 한국생명공학연구원 유전자원은행에 기탁

1) Climate change due to global warming cause changes in habitat conditions of reservoirs and vectors, recently new pathogens have been detected in Korea.
2) It is predicted that the arthropod-borne pathogens, Bartonella, Borrelia,Anaplasma, Ehrlichia, Rickettsia, tick-borne encephalitis virus, etc. will become a growing problem in the future due to global warming in Korea.
3) Bartonella grahamii infection causes endocarditis and neuroretinitis and as a zoonosis which fleas and ticks serve as the vectors and it has been uncovered in Korea that wild rodents are the reservoirs.
4) Ensure isolation of B. grahmaii from wild rodents in Korea and electron microscopic, molecular biological, and serological characterization.
5) To understand pathogenicity and characteristic of B. grahmaii Korean isolate.
Result
1) Using ITS-based nested PCR, B. grahamii and B. schoenbuchensis were detected in 11 (15.8%) and 9 (12.9%) of 70 Korean water deer (KWD) spleens,respectively. By rpoB-based nested PCR, B. grahamii was detected in 6 (8.6%)of 70 Korean water deer spleen samples. The prevalence of Bartonella species,in rodent spleen samples, was 62.0% (124/200) by ITS region detecting PCR and 31.5% (63/200) by rpoB targeting analysis.
2) Two B. grahamii strains were isolated from the blood of rodents in Korea.
3) Comparison of 16S rRNA, hemin-binding protein E (hbpE), glutamate dehydrogenase 1 (gdh 1), invasion associated protein B (ialB), cell division protein (ftsZ), citrate synthase (gltA), 60 KDa heat shock protein (groEL), and rpoB gene and ITS region sequences from the isolates indicated that they were identified as B. grahamii.
4) KRBG28 and KRBG32 strongly stimulated HUVEC, as determined by nuclear factor-κB (NF-κB) activation and enhanced inflammatory adhesion molecules.KRBG28 and KRBG32 were much more active in inducing inflammatory response of HUVEC than B. grahamii V2.
5) Mice inoculated with KRBG28 and KRBG 32 showed severe histopathological changes compared to B. grahamii V2.
6) Beagle dogs inoculated with Bartonella grahamii did not show any pathogenic symptoms and abnormal clinical pathology.
Expected Contribution
1) It will be the first isolation of B. grahamii in Korea, and deposited to Korea Cell Line Bank (KCLB) for research and development
2) Research and develop using the pathogens in different discipline studies.
