HBCDs와 TBBPA는 브롬화 난연제로 제품의 연소를 막기 위한 용도로 사용되어 국내에서도 많은 양이 사용되었다. 하지만 HBCDs는 난분해성, 잔류성, 생물 축적성 등의 특성을 가지며 2013년 POPs 물질로 지정되어 사용이 금지되었고, TBBPA에 대한 독성은 정확히 밝혀지지는 않았지만 국내·외적으로 사용량이 가장 많은 브롬화난연제로 관리 및 실태조사가 필요하다. 따라서 본 연구에서는 식품 중 HBCDs 및 TBBPA 동시 분석을 위한 시험법을 확립하고 150건의 식품을 수거하여 오염실태 조사를 수행하였다.
연구
HBCDs와 TBBPA는 브롬화 난연제로 제품의 연소를 막기 위한 용도로 사용되어 국내에서도 많은 양이 사용되었다. 하지만 HBCDs는 난분해성, 잔류성, 생물 축적성 등의 특성을 가지며 2013년 POPs 물질로 지정되어 사용이 금지되었고, TBBPA에 대한 독성은 정확히 밝혀지지는 않았지만 국내·외적으로 사용량이 가장 많은 브롬화난연제로 관리 및 실태조사가 필요하다. 따라서 본 연구에서는 식품 중 HBCDs 및 TBBPA 동시 분석을 위한 시험법을 확립하고 150건의 식품을 수거하여 오염실태 조사를 수행하였다.
연구에 사용한 시료는 육류(돼지고기, 쇠고기, 닭고기), 어류(고등어, 꽁치, 연어), 패류(홍합, 전복), 계란 및 우유를 지역 마트에서 수거하였고 육류, 어류 및 패류는 분쇄 후 냉동보관 하였으며 달걀과 우유는 동결건조 후 냉동보관하여 시료로 사용하였다. ASE를 이용한 추출조건을 최적화하기 위하여 추출 용매, 추출 온도, 추출 횟수를 다르게 하여 추출효율을 비교한 결과 Hexane : Acetone=1:1 v/v으로 90℃에서 1회 추출하였을 때 추출 효율이 가장 우수하여 HBCDs와 TBBPA 분석을 위한 추출조건으로 설정하였다. 정제는 GPC, MLC와 silica column을 이용하여 정제효율을 비교한 결과 GPC의 용출 범위가 30 mL∼110 mL로 넓어 정제의 효율성이 떨어졌고 MLC는 α. β. γ-HBCD와 TBBPA의 회수율이 각각 118.05%, 118.82%, 121.51%, 101.85%로 확인되었고 silica column의 회수율은 109.89%, 100.75%, 101.65%, 96.32%로 유의적인 차이는 나타나지 않았다. 하지만 MLC가 더 높게 회수되는 경향을 보였으나 반복 측정하였을 경우 재현성 면에서 silica column이 우수하여 silica column을 정제 방법으로 선택하였다.
시료에 따른 검출한계는 α, β, γ-HBCD의 LOD는 각각 돼지고기 0.019, 0.006, 0.007 ng/g w.w, 고등어 0.020, 0.013, 0.028 ng/g w.w, 홍합 0.019, 0.008, 0.027 ng/g w.w, 우유 0.020, 0.011, 0.017 ng/g w.w, 계란 0.024, 0.020, 0.014 ng/g w.w으로 검출되었고 TBBPA는 돼지고기 0.042 ng/g w.w, 고등어 0.043 ng/g w.w, 홍합 0.036 ng/g w.w, 우유 0.004 ng/g w.w, 계란 0.047 ng/g w.w으로 검출되었다. 시험법에 대한 밸리데이션을 수행하기 위해 돼지고기, 고등어, 홍합, 우유, 계란에 각각 1, 5, 20 ng/g 농도를 넣은 후 회수율 및 상태표준편차를 측정하였다. 그 결과 α-HBCD 91.0-115.3%, β-HBCD 93.7-121.0%, γ-HBCD 90.7-117.0%, TBBPA 87.8-105.5% 범위의 회수율을 보였고 일내 및 일간 재현성은 상대표준편차가 15%를 넘지 않은 수준으로 확인되어 식품 중 HBCDs 및 TBBPA 분석에 대한 시험법으로 최종 확립하였다. 또한 NIST에서 SRM® 1947를 구매하여 정확성을 확인한 결과 α-HBCD의 회수율이 99.41%로 확인되어 다양한 식품 검체에 본 연구에서 확립한 시험법을 적용하였다.
HBCDs와 TBBPA를 동시에 분석할 수 있는 시험법에 대한 적용성을 검토하기 위하여 150건의 식품에 대한 오염실태를 조사하였다. 그 결과 총 HBCDs의 평균 오염도는 고등어(3.797 ng/g w.w) > 홍합(0.506 ng/g w.w) > 꽁치(0.400 ng/g w.w) > 연어(0.141 ng/g w.w) > 전복(0.042 ng/g w.w) > 닭고기(0.027 ng/g w.w) > 돼지고기(0.019 ng/g w.w) > 계란(0.003 ng/g w.w) 순으로 높았고 쇠고기와 우유는 불검출 되었다. TBBPA는 고등어(0.140 ng/g w.w) > 계란(0.079 ng/g w.w) > 쇠고기(0.032 ng/g w.w) > 연어(0.026 ng/g w.w) > 꽁치(0.016 ng/g w.w) > 우유(0.016 ng/g w.w) > 닭고기(0.012 ng/g w.w) > 돼지고기(0.009 ng/g w.w) > 홍합(0.004 ng/g w.w) > 전복(N.D.) 순으로 검출되었다.
총 HBCDs의 측정불확도는 95% 신뢰수준에서 돼지 64.2±5.8 ng/g, 고등어 61.1±5.5 ng/g, 홍합 61.5±5.6 ng/g, 계란 60.2±5.1 ng/g, 우유 59.8±5.2 ng/g이며 TBBPA의 측정불확도는 돼지 20.7±2.6 ng/g, 고등어 21.2±2.5 ng/g, 홍합 20.3±2.5 ng/g, 계란 20.9±2.5 ng/g, 우유 21.4±2.5 ng/g로 산출되었다. 따라서 본 시험법을 통해 국내 브롬화 난연제의 실태조사와 위해성 평가를 위한 기초 자료로 제공될 수 있을 것으로 기대된다.

In 2013, HBCDs have been officially listed as persistent organic pollutants (POPs) due to their persistence, bioaccumulation and toxicity to wildlife and human beings. TBBPA hasn’t been reported to toxic in human. However, TBBPA is required to management and regulation because it is consumed the most among the BFRs. Therefore, the objective of study was to determine an analytical method for safety on BFRs and to investigate contamination level by performing monitoring in foods.
The meat (pork, beef, chicken), fish (mackerel, pacific saury, salmon), shellfish (mussel, abalone), egg and milk were purchased from local market. The meat, fish and shellfish were homogenized and stored in a freezer at -20℃ until extraction. Egg and milk were prepared after freeze-dry. The samples were extracted with pressurized liquid extraction (PLE) using a ASE 350 for optimization of extraction condition about solvent, temperature and cycle. A result, extraction condition of HBCDs and TBBPA was optimized at 90 ℃ for 1 cycle using hexane : acetone (1:1 v/v).
The clean-up was compared with GPC, MLC and ASC for simultaneous analysis of HBCDs and TBBPA. Recovery of MLC and ASC clean-up methods were analyzed on α. β. γ-HBCD and TBBPA respectively (MLC; 118.05%, 118.82%, 121.51% and 101.85%, ASC; 109.89%, 100.75%, 101.65% and 96.32%). Between MLC and ASC have no statistically significant differences. Because ASC has a good reproducibility than MLC, it was selected to clean-up method. Otherwise, GPC was considered to have a less purify efficient due to have broad range of elution. The validation of HBCDs and TBBPA was performed in pork, mackerel, mussel, milk and egg. The limit of detections (LOD) for α-. β-. γ-HBCD were confirmed pork 0.019, 0.006 and 0.007 ng/g w.w, mackerel 0.020, 0.013 and 0.028 ng/g w.w, mussel 0.019, 0.008 and 0.027 ng/g w.w, milk 0.020, 0.011 and 0.017 ng/g w.w and egg 0.024, 0.020 and 0.014 ng/g w.w respectively. Also LODs of TBBPA were pork 0.042 ng/g w.w, mackerel 0.043 ng/g w.w, mussel 0.036 ng/g w.w, milk 0.004 ng/g w.w and egg 0.047 ng/g w.w. The validation of HBCDs and TBBPA were accomplished on precision and repeatability in foods. The recovery of α, β, γ-HBCDs and TBBPA were confirmed α-HBCD 91.0-115.3%, β-HBCD 93.7-121.0%, γ-HBCD 90.7-117.0%, TBBPA 87.8-105.5% and relative standard deviations (RSD) of intra/inter-day validation below 15% to 1, 5 and 20 ng/g, respectively. Moreover SRM® 1947 was analyzed to verify accuracy on developed method. A result, recovery of α-HBCD was confirmed to 99.41%. Finally, this method was developed to simultaneous analysis method for HBCDs and TBBPA.
For applicability of developed simultaneous analysis method to HBCDs and TBBPA, we conducted a monitoring each of the 15 cases (pork, beef, chicken, mackerel, pacific saury, salmon, mussel, abalone, milk and egg). As a result of analysis total HBCDs, the average levels were high mackerel (3.797 ng/g w.w) > mussel (0.506 ng/g w.w) > pacific saury (0.400 ng/g w.w) > salmon 0.141 ng/g w.w) > abalone (0.042 ng/g w.w) > chicken (0.027 ng/g w.w) > pork (0.019 ng/g w.w) and egg (0.003 ng/g w.w) in order, but beef and milk were not detected. Also TBBPA was high mackerel (0.140 ng/g w.w) > egg (0.079 ng/g w.w) > beef (0.032 ng/g w.w) > salmon (0.026 ng/g w.w) > pacific saury (0.016 ng/g w.w) > milk (0.016 ng/g w.w) > chicken (0.012 ng/g w.w) > pork (0.009 ng/g w.w) and mussel (0.004 ng/g w.w) in order, abalone was not detected.
The uncertainty value of analytical method for total HBCDs was estimated that pork was 64.2±5.8 ng/g, mackerel was 61.1±5.5 ng/g, mussel 61.5±5.6 ng/g, egg 60.2±5.1 ng/g and milk 59.8±5.2 ng/g in the 95 percentile confidence. And the uncertainty value of analytical method for TBBPA was estimated that pork was 20.7±2.6 ng/g, mackerel was 21.2±2.5 ng/g, mussel 20.3±2.5 ng/g, egg 20.9±2.5 ng/g and milk 21.4±2.5 ng/g in the 95 percentile confidence. Therefore, this study could be used as a resource for the analysis of BFRs and risk assessment in food.
