본 연구의 목표는 노화에 따른 미토콘드리아 기능이상과 이로 인해 초래되는 여러 가지 병리현상이 노인성 당뇨병 및 노화관련 대사질환의 공통적인 발병기전으로 작용할 것이라는 새로운 가설 하에, 노인성 당뇨병과 노화합병증의 병태생리 기전을 규명하고 이를 극복하기 위한 표적물질 및 생리활성물질을 발굴함으로써 신약개발의 기반을 마련함에 있음. 1세부 과제에서는 베타세포의 노화와 기능조절 생체분자 규명 및 제어기술 개발을 목표로 clusterin 및 ASC-2활성을 통한 베타세포 기능개선 효과를 규명 함 또한 ASC-2의 promoter 분석
본 연구의 목표는 노화에 따른 미토콘드리아 기능이상과 이로 인해 초래되는 여러 가지 병리현상이 노인성 당뇨병 및 노화관련 대사질환의 공통적인 발병기전으로 작용할 것이라는 새로운 가설 하에, 노인성 당뇨병과 노화합병증의 병태생리 기전을 규명하고 이를 극복하기 위한 표적물질 및 생리활성물질을 발굴함으로써 신약개발의 기반을 마련함에 있음. 1세부 과제에서는 베타세포의 노화와 기능조절 생체분자 규명 및 제어기술 개발을 목표로 clusterin 및 ASC-2활성을 통한 베타세포 기능개선 효과를 규명 함 또한 ASC-2의 promoter 분석을 통해 전사조절인자 연구를 통해 ASC-2 조절물질을 발굴하여 활성조절을 통한 치료기술 개발을 가능케 함. 2세부 과제에서는 노인 당뇨병의 대사 이상과 미토콘드리아 기능이상의 관련성을 분석하고, 근감소증이 노인 당뇨병에 미치는 영향을 확인함. 노인 코호트 추적관찰을 통해 혈당조절과 근감소의 연관성을 규명하였으며 노화 및 미토콘드리아 기능관련 후보 유전자를 발굴함. 또한 미토콘드리아 기능개선 후보물질을 발굴하여 이들 물질의 in vivo 효과를 확인함. 3세부 과제에서는 당뇨병 노화합병증의 제어기술 개발을 위해 노인성 당뇨병의 혈관합병증, 신장합병증, 지방간 질환 및 비만과 관련된 지방조직 기능이상을 극복하고자 미토콘드리아-핵수용체-ER 스트레스 network을 확립하여 신약개발의 원천기술을 확보함. 본 연구를 통해 당뇨병의 노화관련 합병증의 발생기전에 대한 이해를 한 단계 높이고 이를 기반으로 한 새로운 치료법 및 예방법을 개발할 수 있는 초석을 마련하였음.

2. Projects #2: "Discovery of markers for diabetes in elderly and development of therapeutic method to overcome diabetes by improving mitochondrial function"
(1) Clinical characteristics and metabolic parameters of Diabetes in elderly
- As the aging progresses, muscle mass decreases in their quantity and quality and the body fat mass decreases.
- In subjects with elderly diabetes, expression of Nampt, clusterin, phospholipase A2(PLA2G7), GPx3 increased significantly, and PLGA2G7 was higher compared to elderly non-diabetics.
(2) Change in skeletal muscle and mitochondrial function in elderly diabetes group
- Decrease in quantity and quality of mitochondrial function was observed in patients with diabetes and more significant in elderly patients.
(3) Discovery of target gene and protein for aging
- Proteomic analysis in muscle tissues of diabetics and controls resulted in isolation of 2,179 proteins, and therefore expressed genes and underlying cellular mechanism on aging and diabetes were discovered. Decrease in fatty acid beta-oxidation and proteins involved in lipid metabolism and glycolysis were observed.
(4) Establishment of system for studying marker proteins of mitochondrial dysfunction
- We screend candidate proteins associated with the translocation of nampt into mitochondria and found that HSPA5 interacts with nampt. HSPA5 is a molecular chaperone and we are investigating if nampt interaction with HSPA5 has any association with ER stress.
(5) The effect of exercise on mitochondrial function and insulin resistance
- In aging model of Fisher Rat, reversal of increased muscular cytokine, decreased catalase/Gpx, increased ROS was demonstrated by exercise.
- In elder group, baseline IL-6 in liver, muscle, plasma was higher than young group. TNF alpha and IL-6 expression was decreased after long endurance exercise.
(6) Discovery of diabetes prediction indicator by mitochondrial function
- IL6, Follistatin-like 1 were increased and FGF21, PRDM16, SFRP5, C1QTNF5 were decreased in elderly non-diabetics compared to young non-diabetics. Elderly diabetics showed increased C1QTNF5, SFRP5, PRDM16, FGF21 and decreased Follistatin-like 1 compared to elderly non-diabetics.
(7) Discovery and validation of mitochondrial targets related with elderly diabetes
- The ERK-cMyc pathway was newly discovered which is critical in insulin resistance and beta cell dysfunction induced by mitochondrial dysfunction.
(8) Development of therapeutics improving mitochondrial function and evaluation the effects on animal models
- In overexpression of SENP2 (SENP2 Transgenic mouse), increased expression of genes for fatty acid oxidation, decreased weight and fat mass, improvements in glycemic indices and insulin resistance were observed.
- Injection of Rg3 increased mitochondiral extent, and increase in associated genes such as PGC1α, NRF-1, and Tfam was obserrved. Rg3 improves both quantity and quality of mitochondria, thereby improving insulin resistance.
- Decrease in IRS1 expression due to insulin resistance, was reversed by RXRα catalyst.
3. Projects #3: "Development of novel therapeutic measures to overcome aging-associated diabetic complications"
(1) Mechanisms of development of diabetic nephropathy
- Loss of clusterin accelerates angiotensin II-mediated renal fibrosis and A TIR expression
- Increase of endogenous PUFAs prevented lipotoxicity-induced nephropathy in Fat-1 Tg mice
- Myriocin, a potent inhibitor of serin-palmitoyl transferase, ameliorates diabetic nephropathy and podocyte injury
- Orphan nuclear receptor SHP attenuates renal fibrosis in unilateral ureteral obstruction(UUO)-induced renal fibrosis
- Upregulation of clusterin during renal injury is a protective response against the development of renal fibrosis
(2) Preventive mechanisms of macrovascular diabetic complications
- PGC-1α regulates oxidative stress and apoptosis in endothelial cells by increasing fatty acid oxidation and enhancing ANT activity
- Cilostazol and SHP attenuates proliferation of vascular smooth muscle cell through inhibition of PAI-1 expression
(3) Treatment and prevention of non-alcoholic fatty liver disease (NAFLD)
- HMG-CoA reductase inhibitor prevented hepatic steatosis and steatohepatitis through increasing endogenous plasmalogen synthesis
- Myriocin prevents MCDD-induced steatohepatitis
- Expression of SMS1 led to caspase-1-dependent pyroptosis and development of NASH

(4) Development of therapeutic strategies to overcome aging- associated metabolic diseases through improvement of adipose tissue function
- Precursor of plasmalogen reduces HFD-induced insulin resistance and adipose tissue inflammation
- Lack of inducible nitric oxide synthase (iNOS) prevents HFD-induced adipose tissue fibrosis in mice
- Increased expression of 11β-HSD1 in hypertrophic adipocytes is responsible for suppression of adiponectin synthesis
- Treatment with myriocin and overexpression of Fat-1 gene prevent whitening of brown adipose tissue through restoring of AMPK activity
- eNOS plays an important role in adiponectin synthesis in adipocytes by increasing mitochondrial biogenesis and enhancing mitochondrial function
(5) In vitro metabolic efficacy of compounds from natural products
- Identification of new molecules from natural products that increase AMPK and SIRT1 activity
- Identification 6 molecules as GCN5 inhibitors using HTS (high throughput screening)
- Development of novel therapeutic materials identified from Evodia rutaecarpa and Glycyrrhiza uralensis for diabetic complications
4. Commissioned project: "Development of bioactive compounds using drug targets in elderly diabetes"
- During a search for SIRT1 activators, three new dammarane triterpenes, as well as two known triterpenes, were isolated from Panax ginseng leaves. Compounds 1 and 3-5 showed potential as SIRT1 activators, as analyzed by in vitro enzyme-based SIRT1-NAD/NADH and SIRT1-p53 luciferase cell-based assays.
- In the course of our program to search for PTP1B inhibitors, five new 5-deoxyflavonoids along with eight known derivatives were isolated from EtOAc layer of the root bark of Erythrina abyssinica.
- Bioassay - guided fractionation of Morinda longissima extract, using 2-NBDG assay in 3T3-L1 adipocyte cells, yielded two new anthraquinones, along with eighteen known analogues. These anthraquinones showed the potential action as insulin mimetic to improve glucose uptake via activation of AMPK.
