■ 복합살균을 활용한 신선편이 농수산물의 식중독 저감화 기술 개발
Ⅳ.연구개발결과
1.대상 식품별 살균소독수의 살균 유효성 평가
○ 12종의 세균,4종의 곰팡이와 4종의 효모를 대상으로 살균소독수의 살균 유효성을 분석한 결과,30 ppm의 미산성 차아염소산수(SlAEW ;slightly acidic electrolyzed water,slightly acidic hypochlorite water)는 다양한 세균,곰팡이 및 효모에 대해 5-6log CFU 수준의 살균 유효성을 나타내었다.10ppm 이상의 이산화염
■ 복합살균을 활용한 신선편이 농수산물의 식중독 저감화 기술 개발
Ⅳ.연구개발결과
1.대상 식품별 살균소독수의 살균 유효성 평가
○ 12종의 세균,4종의 곰팡이와 4종의 효모를 대상으로 살균소독수의 살균 유효성을 분석한 결과,30 ppm의 미산성 차아염소산수(SlAEW ;slightly acidic electrolyzed water,slightly acidic hypochlorite water)는 다양한 세균,곰팡이 및 효모에 대해 5-6log CFU 수준의 살균 유효성을 나타내었다.10ppm 이상의 이산화염소수(ACD ;aqueous chlorine dioxide)도 6 log CFU 이상의 살균 유효성이 있었으며,5 log CFU의 저감의 위해서는 5ppm 이상이 요구되었다.강산성 차아염소산수 (SAEW ; strong acidicelectrolyzedwater,strong acidichypochloritewater)및 약 알칼리성차아염소산수(LAEW ; akaline electrolyzed water, strong acidic hypochlorite water))는 50ppm 이상의 염소농도에서 6-7log CFU 수준의 저감효과가 있는 것으로 나타났다.
○ 유통중인 양상추 등 4종의 채소류와 2종의 과일을 대상시료로 하여 단순 침지살균을 통해 저감효과를 분석한 결과,30ppm의 미산성 차아염소산수는 2log CFU/g 수준이었으며 소비되는 유효염소는 약 10% 수준이었다.2차 및 3차의 다단 침지를 통해 저감효과를 분석한 결과 1log CFU/g 이상의 저감효과는 나타나지 않아 다단 침지방식은 저감화에 효과적이 않다고 판단되었다.또한 3차례의 침지 살균을 통한 잔류 미생물의 오염도는 2-3logCFU/g이었다.
○ 혼합샐러드와 무순 등의 새싹채소,2종의 과일을 대상시료로 하여 침지살균을 통한 이산화염소수의 살균효과를 분석한 결과,30 ppm의 이산화염소수는 약 2-3 log CFU/g의 감균효과를 나타내었으나 무순과 브로콜리순은 엽채류(leafy vegetable)에 비해 낮았으며 이는 내재(integrated)된 미생물이나 줄기와 잔모 등의 구조적 문제에 대한 것으로 판단되었다.
○ 이산화염소수에서 유효염소인 이산화염소는 10ppm에서는 30분후 초기농도의 10%, 20ppm과 30ppm에서는 30-40%가 감소하였으며 표면적인 작은 과일에서는 감소량이 상대적으로 낮았다.
○ 강산성 차아염소산수의 저감효과는 양상추,혼합 샐러드,무순,브로콜리순,방울토마토 및 사과 등 6종의 시료에 대해 대체적으로 1차 침지 20분 이내에 2-3logCFU/g의 저감효과가 나타났다.무순과 같은 새싹채소는 1차보다는 2차나 3차의 추가적인 침지가 미생물 저감에 효과적이었으며 3차 침지살균 이후 약 3log의 저감효과가 있는 것으로 나타나 물리적 손상을 고려하지 않는다면 침지살균 시간을 연장하는 것이 미생물 저감에 보다 효과적이라고 판단되었다.
○ 약알칼리성 차아염소산수도 저감효과는 양상추,혼합 샐러드,무순,브로콜리순,방울토마토 및 사과 등 6종의 시료에 대해 강산성 차아염소산수와 유사한 수준의 저감효과를 나타내었으며 새싹채소의 경우에는 침지살균 시간과 횟수를 늘이는 것이 미생물 저감에 보다 효과적이었다.그러나 전반적으로 강산성 차아염소산수에 비해 약알칼리성 차아염소산의 저감효과는 낮은 것으로 나타났다.
○ 살균효과를 높이기 위해서는 과도한 양의 유기물이 존재하지 않는다면 유효염소량이나 처리시간의 증가보다는 접촉효율을 높일 수 있는 물리적 처리방법을 병행하는 것이 보다 효과적인 수단이 될 것으로 생각된다.
○ 병원성 미생물에 대한 살균 효과를 분석하기 위하여 양상추,혼합 샐러드,무순,브로콜리순,방울토마토 및 사과에 107-108 CFU/g으로 병원성 대장균인 E.coliO157:H7 NCTC 12079와 S.enterotidis를 오염시킨 후 살균소독수로 침지하여 저감화율을 분석하였다.병원성 미생물인 E.coliO157:H7NCTC 12079와 S.aureusKCCM 12214 를 시험균주로 하여 양상추와 혼합샐러드에 인위적으로 오염시킨 다음 미산성 차아염소산수,강산성 차아염소산,약알칼리성 차아염소산수,이산화염소수의 살균 유효성을 평가하였다.엽채류는 수도수는 약 1logcycle의 감균 효과를 나타낸 반면 미산성차아염소산수(SlAEW)는 약 4 log cycle,50 ppm의 이산화염소수(ACD)는 5 log cycle, 강산성 전기분해수(SAEW)는 약 4-5 log cycle, 약알칼리성 전기분해수(WAEW)는 약 4.0log cycle의 감소를 보였다.그러나 절단된 혼합샐러드와 새싹 샐러드의 경우에는 살균소독수별로 약 1logcycle의 낮은 저감 효과를 나타내었다.
2.복합 살균을 통한 저감화 효능 평가
○ 양상추,케일,사과,방울토마토,브로콜리 새싹,무순,혼합 샐러드 그리고 수산물인 고등어와 명태를 시료(n=3-6)로 하여 순차적으로 염소계 살균소독수로 복합살균한 결과,양상추,케일에서는 50ppm의 이산화염소 살균소독이 잔류 오염도 2logcycle 로 가장 우수하였으며,사과와 토마토에서는 50 ppm의 이산화염소 살균소독후 68ppm의 알칼리성 차아염소산수나 30ppm의 미산성 차아염소산수 살균소독이 잔류 미오염도 1-2log cycle로 효과적이었다.그러나 새싹채소인 브로콜리순과 무순에서는 복합살균이 개별 살균소독수 단독처리에 비해 유사하거나 1logcycle의 추가로 감균하는 효과만이 확인되었으며 잔류 오염도 3-4log cycle을 나타내었다.수산물인 고등어와 명태의 경우에도 최대 약 1log cycle의 감균효과가 확인되었으나 품목의 특성상 2log cyle이하로의 감균은 어려운 것으로 판단되었다.침지살균후 모든 시료에서 잔류 염소량은 0.01-0.02ppm으로 측정되어 수도수의 잔류 염소량 수준인 것으로 판단되었으며 관능적인 측면이나 안전성 측면에서 유의할 만한 수준은 아니라고 판단되었다.
○ 50ppm의 이산화염소수를 살균소독수로 한정하여 물리적인 방법을 복합처리한 결과 마이크로버블>초음파>malicacid순으로 저감효과가 우수하였으며 약 1log cycle의 추가적인 저감효과가 있을 것으로 판단되었다.
○ 이산화염소 가스(gaseous chlorine dioxide)를 20 ppm(v/v), 30 ppm(v/v), 50ppm(v/v) 및 70 ppm(v/v)으로 살균 처리한 결과,양상추와 같은 엽채류는 20ppm(v/v)에서는 약 1log cycle,30ppm(v/v)에서는 2log cycle,50ppm(v/v)과 70ppm(v/v)에서는 1log cycle이하로 감소하였다.색차를 분석한 결과 50ppm(v/v)을 기준으로 5분까지는 유의적 차이가 크지 않지만 10분에서는 L값과 b값은 증가하고 a값이 다소 감소하여 관능적 품질이 저하하는 것으로 생각되었다.사과,딸기와 방울토마토의 경우에도 엽채류와 유사한 감균 효과를 나타내었으나 딸기의 경우에는 50ppm(v/v),5분 이상에서는 a값의 감소가 확인되었다.그러나 무순은 70ppm(v/v)의 농도에서 10분후에도 2log이상의 감균이 나타나지 않았으며 혼합샐러드의 경우에는 70ppm(v/v)농도에서 5분후 약 4log cycle이 감소하여 1log cycle의 감균효과가 있었다.
○ 이산화염소 가스 처리후 차아염소산수 복합살균 효과를 시험한 결과 시험한 과채류 모두에서 약 0.5-1 log cycle의 추가적인 감균효과가 확인되었다.특히 무순과 혼합샐러드에서의 효과가 다른 엽채류에 비해 다소 높은 것으로 나타났다.
3.새싹 채소의 식중독균 저감화 공정기술 개발
○ 이산화염소수와 가압 처리(pressurization treatment)에 의한 채소종자(vegetable seed)의 살균효과를 분석한 결과,종자:이산화염소수 비율을 1:10(v/v)로 하고 압력을 6 kg/cm2로 한 경우에는 30 ppm의 이산화염소수 농도에서 처리 5분 이후 모두 1 logCFU/g이하로 감소하였다.
○ 살균후 무종자의 발아율은 대조구와 유사한 발아율(germinationrate)을 나타내어 30ppm 수준의 이산화염소나 최대 6kg/cm2의 가압조건은 발아율에 미치는 영향이 없는 것으로 판단되었다.
○ 저농도 살균소독수를 재배수(cultivation water)로 활용하여 무순의 생육도와 미생물오염도를 분석한 결과 재배 7일에는 모든 처리구가 6log CFU/g으로 유사한 수준의 오염도를 나타내어 발아후 1일 이후에는 미생물 생육 억제효과는 없는 것으로 판단되었다.다만 종자살균을 통해 병원성 미생물을 제어하는 것은 유효한 수단이 될 수있을 것으로 생각되었다.
4.신선편이 농수산식품별 식중독균 저감화 최적 처리 방법 및 공정 설계
○ 살균소독수 단독,또는 복합살균 효과 분석결과를 바탕으로 단일 살균소독수를 활용한 세척,소독 공정에 대한 작업표준(SOP ;standardoperatingprocedure)과 복합 살균소독 처리공정을 작성하였다.주요 공정은 세척을 기본 공정으로 하여 불림,1차세척,소독,헹굼의 4단계로 구성하고 마이크로버블을 1차 세척과 헹굼공정에 복합처리하는 것으로 설계하였다.
○ 또한 bacteriophage를 활용하는 식중독 세균 특이성 bacteriophage처리공정을 작업 순서 및 작업방법,주의 사항,설비,공정관리 기준을 미생물 오염도 약 4logCFU/g을 기준으로 작성하였다.
5.처리공정 기본모델 설정 및 현장평가
○ 참여기업인 한국코스믹라운드와 테크윈에서 신선편이 농산물 및 수산물가공장(FPC)에 염소계 살균소독제를 효과적으로 사용하기 위하여 본 연구결과를 토대로 민간기업에 제안한 제조시설을 기준으로 식중독균을 저감화할 수 있는 실용적인 제조시설제이아웃과 처리 방법,기본 모델을 작성하였다.엽채류와 구근류의 세척시스템을 기본으로 하여 불림,세척,소독,헹굼으로 구성된 단위공정 유닛으로 구성하였다.
6.신선편이 농수산물의 식중독균 저감화 매뉴얼 개발
○ 신선편이농산물 제조관리기술 동향을 조사하여 결구상추,양파,감자,당근 등 주로 많이 소비되는 농산물을 대상으로 원료관리,공정관리,품질관리,유통관리 등의 주요관리 기술을 정리·요약하였다.
○ 신선편이 농수산물의 제조시설/설비 및 기구에 대한 관리 기준(구획화,동선관리,용수관리,공조·환기관리,온습도관리,조명,방충방서,기계·장비관리 등),위생처리를 위한 관리지침 개발(작업장 정리정돈,청소,세척,소독, 개인위생관리,폐수 및 폐기물처리 등)및 제조시설 및 공정관리 통합 프로그램(GAP,GMP,HACCP,GHP등)을 개발하였다.
○ 서류,구근류,엽경채류,과채류,과실류 등 농산물 유형별 공통 제조공정을 단위공정화 및 표준화하여 공정관리 매뉴얼을 개발하여 공장 여건에 따라 단위공정을 선택·적용할 수 있도록 하였다.
○ 법규관리,품질관리,공정관리,위생관리,시설관리 등 신선편이농수산물의 위생안전관리를 위해 기본적으로 관리하여야 할 사항을 분류,정리하여 위생관리지침을 개발하였다.
■ 신선편이 농수산물의 식중독균 저감화 수준 유지 기술 개발
Ⅳ.연구개발결과
제 1절 신선편이 제품의 유통 안정성 조사
1.예측미생물학을 이용한 시판 셀러드 제품의 미생물학적 유통기한 예측
○ 시판 샐러드제품의 구입부터 가정까지의 이동 및 소비직전까지 적정하지 않은 온도관리를 예상하여 단기간의 온도 abuse상황을 설정하고 미생물적 유통기한을 도출하였다.보다 효율적인 유통기한 설정을 위해 예측미생물학의 3단계 모델인 PMP 7.0을 활용하여 그 활용가능성을 조사하였다.부적절한 온도에서의 abuse시간이 증가할수록 미생물은 빠르게 증식하여 샐러드 제품의 유통기한 시점으로 판단되는 log 7CFU/mL에 도달하는 시간이 짧아졌다.온도가 증가할수록 0.020에서 1.083까지 grow rate도 증가했으며,이 모델의 적합도를 나타내는 r2의 값은 전 실험구에서 0.9이상을 나타내었다.PMP 7.0으로 예측된 미생물의 증식 양상은 온도에 따라 0.017∼0.235CFU/mL/hr로 나타났으며,모든 구에서 0.994∼1.000까지 높은 수준의 r2을 나타내었다.또 PMP를 활용하여 도출한 유통기한의 경우도 온도가 증가함에 따라 감소하였다.실측된 값을 바탕으로 한 샐러드 제품의 유통기한은 유통 매장에 도착하기까지 48시간 소요될 것으로 예상할 경우 유통기한은 109.1∼63.0시간까지로 추정되며 PMP로 도출된 유통기한(24.1∼626.5시간)에 비해 짧게 나타났다.이는 온도 abuse에 의한 영향 및 failsafe에 해당하는 결과로 안전성 측면에서는 유리하나 관리적 측면에서 과도한 기준의 설정 등을 통해 관리비용의 증가 등의 단점이 발생할 수 있는 것으로 판단된다.즉,예측미생물학을 활용하여,유통기한 설정 및 품질관리를 위한 초기 미생물 기준 설정 시 특정 식품에 적용하는 것은 효율적인 시도가 될 것이나,이를 전반적인 기준으로 설정하는 것은,통계적,실제적 오류 발생이 가능할 것으로 오히려 관련 효율을 저해할 수 있을 것이다.
2.시판 손질 고등어(Scomberjaponicus)의 미생물 특성 조사
○ 손질하여 판매하는 고등어 제품의 어획 후 수송,운반,손질 등의 중간단계에서 각종 오염 미생물에 노출되거나 오염될 가능성이 있으며 이에 대한 연구는 미비한 상황이다.본 연구에서는 손질하여 포장판매하는 제품 및 즉석에서 손질하여 판매하는 두 가지 제품의 형태로 대형백화점,대형할인점 및 농협유통점,기타 소매점 등 다양한 유통점에서 구입하여 일반세균수,대장균군 및 식중독균인 Escherichia coli,Staphylococcus aureus,Bacillus cereus의 오염여부를 분석하였다.구입 장소별 고등어의 일반세균수는 평균 3.56 log CFU/g로 나타났으며,백화점 내 동일 판매처에서 구입한 손질 후 포장되어 있는 제품과 즉석에서 손질한 제품의 일반세균수는 각각 5.10log CFU/g와 4.90log CFU/g로 나타나 포장을 위한 전처리과정 및 매대 진열 과정 중에 균이 증식했음을 확인할 수 있었다.손질 고등어의 대장균군 오염정도는 평균 1.51log CFU/g이었으며,할인점 중 한곳에서 구입한 손질 고등어에서 최대 3.26 log CFU/g수준이 검출되었다.대표적 독소형 식중독균인 S. aureus의 경우 모든 구입 장소의 시료에서 검출되었으며,그 수준은 평균 2.90log CFU/g 이었다.B.cereusgroup은 총 12군데의 구입처 중 1군데 샘플의 50%에서 검출되었고 E. co.li의 경우 이미 손질되어 포장된 제품을 구입한 총 4곳 중 1곳(25%)에서,즉석에서 손질을 요청,구입한 총 8곳 중 2곳(25%)의 유통점에서 검출되었다.즉,유통판매점별 식중독균의 검출양상이 매우 다양하며 위생관리 수준이 서로 상이하므로,유통 전반적으로 수산물에 대한 관리강화가 필요할 것으로 판단된다.
제 2절 신선편이 농수산물의 식중독균 증식억제 기술 개발과 적정 포장 방법 설정 및 효과 분석
1.식중독균 증식억제 기술 개발
가.Activepackaging용 이산화염소 가스 발생제 개발
○ 이산화염소 발생을 위해 가장 일반적인 방법인 아염소산나트륨 용액과 염산용액을 반응시켜 이산화염소를 발생시킨 후 유지 능력을 측정하고자 하였다.이를 위하여 용질의 종류를 달리하여 적정한 pH 농도 조건을 설정하였다.pH의 영역은 pH 3~4에서 이산화염소 가스발생의 유도가 원활하였으며,이들의 용질은 구연산 buffer,HCl-NaOH buffer,Lacticacid buffer,succinicacidbuffer,Universalbuffer등이 효과적인 것으로 나타났다.이들 buffer에 대한 이산화염소 가스 발생량을 20여일 동안 조사한 결과 Universalbuffer가 HCl buffer에 비하여 지속성이 높은 것으로 나타났다.또한 아염소산나트륨의 함량과 산의 농도비율에 따른 이산화염소 가스 발생량을 조사한 결과 아염소산나트륨 함량이 일정량 이상 (1000ppm 이상)일 때 이산화염소 가스가 상대적으로 크게 증가하였으며,산의 농도는 큰영향을 미치지 못하였다.이산화염소 가스 발생제의 포장을 위하여 겔 포장재로서 다양한 필름을 이용하였다.이산화염소 가스는 PE 필름 계열이 방출 효과가 높은 것으로 나타나, LLDPE>LDPE>HDPE의 순으로 투과성을 나타내었다.일반적으로 이산화염소 가스의 투과도는 HDPE에 비하여 LDPE와 LLDPE가 각각 8배,13배 이상의 투과성이 있는 것으로 알려지고 있다.본 연구에서는 이산화염소 가스 발생 겔의 포장을 위하여 LDPE를 이용하여 포장한 후 방출제로서 사용하는 것이 유통 중 신선편이 제품의 위해미생물 저감화에 효과적으로 활용할 수 있을 것으로 판단되었다.
나.식중독균에 대한 activepackaging적용 효과 분석
○ 이산화염소 가스를 이용한 위해미생물의 증식억제 효과를 분석한 결과,Bacilluscereus의 경우 1.0ppm 이상의 이산화염소가스 처리구에서는 ND 수준으로 bacilluscereus가 검출되지 않았으며,E.Coli의 경우 2.0ppm의 이산화염소 가스 농도에서 대조구에 비하여 약 1/5수준으로 발생 양상이 감소하였다.Salmonella tiphy.균에서도 0.5ppm의 농도에서 1 log CFU 수준으로 감소하였다가 2.0ppm의 농도에서는 ND 수준으로 감소되는 것으로 나타났다.Staphylococcusaureus균에서도 2.0ppm의 이산화염소가스 처리구는 ND 수준으로 감소하였으며 1.0ppm의 농도에서는 대조구에 비하여 약 1/4수준으로 발생 영역이 감소하였다.
○ 산소 농도 조건과 이산화염소 가스 혼합에 대한 식중독균(Bacilluscereus)의 증식억제 효과 분석한 결과,Bacillus균의 특성상 포장지내에 산소가 첨가된 경우 모두 증식이 잘되어 도말된 배지의 전면에 균이 생육한 것을 나타내었다.반면 질소 100% 충진 포장구에서는 일부 생육이 억제되는 것으로 보였으며,이와 함께 이산화염소가스 혼합 포장구에서는 엷은 층의 균이 생육되어 상당한 억제 능력을 나타내었다.전반적으로 이산화염소가스 혼합처리구의 경우 bacillus cereus 균의 성장이 억제되는 것으로 보였으며,산소농도 90%와 100%와의 혼합구에서는 측정가능한 colony 수를 나타내고 있었다.전반적으로 산소농도가 높고 이산화염소가스 혼합처리구에서 균의 생육 억제 효과가 큰 것으로 나타났다.
○ 이산화탄소 농도 조건과 이산화염소 가스 혼합에 대한 식중독균(Bacilluscereus)의 증식억제 효과 분석한 결과,전반적으로 이산화탄소 가스치환포장구는 대조구에 비하여 균의 생육정도가 약한 것을 볼 수 있었으며,이산화염소가스 혼합포장구에서는 그 효과가 더욱 높은 것으로 나타났다.이산화염소가스 혼합포장리구 중 이산화탄소 30%,70% 및 100% 처리구에서는 균의 생육이 완전히 억제되는 것으로 나타났으며,이산화탄소 20%,40%,50%, 80% 및 90% 농도와의 이산화염소 혼합포장구는 colony의 수를 측정할 수 있을 정도로 균의 생육이 억제되었다.
○ MAP 조건에 대한 식중독균(Bacilluscereus)의 증식억제 효과 분석 결과,Bacilluscereus균은 MAP 포장구 중 이산화탄소 10%/산소 5%를 제외하고 대조구와 큰 차이를 나타내지 않았다.그러나 이산화염소 가스 혼합처리구 모두 대조구 및 MAP 단독 포장구와 큰 차이를 나타내었다.이산화염소 가스 혼합처리구 중 이산화탄소/산소 혼합비 10%/5% 및 20%/5%구에서 균수가 1log CFU 수준 까지 감소하였다.이산화염소 혼합 포장구는 모든배지에서 미생물의 증식이 억제되는 효과를 확인할 수 있었으며 특히 산소 농도 5% 혼합구에서 더욱 우수한 효과를 나타내었다.
2.적정 포장 방법 설정
가.이산화염소 가스를 적용한 신선편이 식품의 미생물 증식억제 효과
○ 이산화염소 가스를 적용한 신선편이 식품의 미생물 증식억제 효과를 조사하기 위하여 사과, 감자,무싹 및 혼합잎채소를 절단하여 포장한 후 각각의 이산화염소 가스 농도로 처리하여 유통 조건 10℃에서 5일간 보관한 후 미생물의 변화를 조사하였다.사과의 경우 0.5ppm 이상의 처리조건에서는 ND 수준을 나타내었으며,감자는 1.0ppm의 농도에서 약 1.8 log cycle이 감소하였고 2.0ppm의 농도에서는 ND 수준을 나타내었다.무 새싹채소의 경우 1.0ppm의 농도에서는 약 1logcycle의 감소와 2.0ppm의 농도에서는 약 1.6logcycle이 감소하였다.반면 혼합 잎채소의 경우 2.0ppm의 농도에서 약 1.3log cycle이 감소되는데 그쳤다.
나.이산화염소 가스를 적용한 신선편이 식품의 위해미생물(E.coliO157:H7)증식억제 효과
○ 이산화염소 가스를 적용한 신선편이 식품의 위해미생물(E.coliO157:H7)증식억제 효과를 조사한 결과,cube모양으로 자른 감자의 경우 약 5log CFU/g 수준으로 오염되었고,25 ppm의 이산화염소 가스에 1일 노출시켜도 감자에 오염된 E.coliO157:H7505B 제어가 되지 않았다.이는 감자 절단면 틈으로 균 접종액이 흡수되어 제어에 어려움이 있었던 것으로 사료된다.사과의 경우 오염 수준이 5logCFU/g 이었으나 이산화염소수 처리구에서는 3logCFU/g로 약 2logCFU/g의 감소가 일어났다.무순의 경우 E.coliO157:H7505B의 제어에 있어서 어려움이 있었으며,약 6 log CFU/g 수준으로 오염된 양상추와 청경채의 경우 이산화염소 처리 후 약 llogCFU/g의 감소효과를 나타내었다.
다.이산화염소 가스를 적용한 신선편이 식품의 유통 중 위해미생물 증식억제 효과
○ 이산화염소 가스를 적용한 신선편이 식품의 유통 중 위해미생물 증식억제 효과를 조사하기 위하여,미니파프리카에 4종의 식중독균을 인위적인 오염을 발생시킨 후 이에 대한 제어효과를 조사하였다.S.aureus의 경우 이산화염소 가스 처리 전 5.60log CFU/g 수준이었고,25ppm의 이산화염소가스를 넣고 포장한 경우 7일 후에도 균의 증식이 일어나지 않았으며 6ppm 및 12ppm의 경우 초기에는 균의 증식이 관찰되지 않았으나 시간이 경과함에 따라 균의 증식이 일어나 대조구와 차이를 나타내지 않았다.B.cereus의 경우 25ppm의 이산화염소수가스 처리구에서 초기에는 균의 증식이 저해되었지만 7일 후에는 대조구와 큰 차이를 나타내지 않았고,6ppm 및 12ppm의 이산화염소가스 처리구는 초기부터 대조구와 차이를 나타내지 않아 저농도의 이산화염소 가스에 의한 B.cereus제어에 있어서는 어려움이 있었다.E.coliO157:H7505B와 Sal.enteritidis에 약 3∼4log CFU/g 수준으로 오염된 미니파프리카의 경우 S.aureus와 B.cereus균과는 달리 비교적 저농도의 이산화염소 가스에 의해서도 제어가 잘 되는 것으로 나타났으며,7일 후에도 균의 증식이 관찰되지 않아 이산화염소 가스에 의한 E.coliO157:H7505B와 Sal.enteritidis의 제어에 있어서는 효과적이라고 판단되었다.
3.품목별 포장재 및 기체조성비에 따른 미생물 증식억제 효과 분석
○ 신선편의 사과의 포장방법별 미생물 및 품질유지 효과를 관찰하기 위하여 다양한 포장조건에서 미생물 및 영양성분의 변화를 관찰하였다.조사 결과,포장 조건에 따른 미생물의 생육억제 효과는 가스치환포장구에서 효과적인 증식억제를 보인 것으로 판단할 수 있었으며, 가스농도의 차이는 크지 않은 것으로 나타났으며,영양성분의 함량 변화에서도 이산화탄소농도 5%와 산소 농도 5% 포장구에서 우수한 효과를 나타내었다.
○ 신선편이 사과의 미생물 및 품질유지를 위한 activepackaging 효과를 분석하기 위하여 이산화탄소 5% 및 10%에 대한 산소농도 5% 혼합구를 선정하고 이에 이산화염소가스 1.0ppm 및 2.0ppm을 혼합하여 실험에 사용하였다.초기 신선편이 사과의 총균수는 2.35 log cfu/g이었으나,10℃에서 5일간 유통조건에서 6.58log cfu/g을 나타내었다.반면 MAP 포장에 의해서 약 2-2.5log cycle이 감소 되었으며,이산화염소 혼합 포장에 의해서 3-3.5 log cycle정도 감소되었다.미생물 제어를 위하여 효과적인 수단이 될 수 있을 것으로 판단되었다.
○ 신선편의 감자의 포장방법별 미생물 및 품질유지 효과를 관찰하기 위하여 다양한 포장조건에서 미생물 및 영양성분 변화를 관찰하였다.조사 결과,가스치환포장구의 경우 5.5-6.3 logcfu/g의 범위를 나타내어 대조구에 비하여 2logcycle이상 감소하는 효과를 나타내었다.가스치환포장구에서는 이산화탄소 농도 20%,산소 농도 10% 구에서 5.5logcfu/g으로 서 가장 우수한 효과를 나타내었다.또한,영양성분 분석에서는 이산화탄소 농도 5%,산소 농도 5%에서 가장 높은 값을 나타내었다.
○ 무 새싹채소의 포장방법별 미생물 및 품질유지 효과를 관찰하기 위하여 다양한 포장조건에서 미생물 및 영양성분 변화를 관찰하였다.조사 결과,미생물에 있어서 일반포장구와 가스치환포장구에서 유사한 경향을 나타내었으며 영양성분 분석에서도 LDPE 포장구와 이산화탄소 농도 30%,산소농도 10% 포장구에서 우수한 것으로 나타났다.
○ 신선편이 혼합채소의 포장방법별 미생물 및 품질유지 효과를 관찰하기 위하여 다양한 포장 조건에서 미생물 및 영양성분 변화를 관찰하였다.미생물에 있어서 전반적으로 가스치환포장구가 함기포장에 비하여 낮은 균수를 나타내었으나,그 차이는 미미하였다.영양성분의 변화에서 이산화탄소 농도 10%,산소 농도 5% 포장구에서 우수한 것으로 나타났다.
4.Activepackaging을 통한 적정 포장방법 설정
가.포장 방법 및 activepackaging에 따른 신선편이 식품별 미생물 증식억제 효과 및 품질 분석
○ 시료의 처리를 위하여 신선편이 사과 및 감자는 20mm × 20mm × 5mm의 크기로 잘라 각각의 포장 조건별로 포장을 실시하였으며,쌈추는 약 20mm × 20mm의 크기로 절단하여 사용하였다.무싹은 구입,선별 후 그대로 사용하였다.포장구는 일반포장구(LDPE 50μm), MAP 포장구(CO2:O2: = 10%:5% 및 CO2::O2: = 5%:5%), MAP+이산화염소가스혼합구[(CO2::O2: =10%:5%)+1ppm,(CO2::O2: =10%:5)+2ppm]으로 포장하여 10℃에서 저장하면서 5일 동안 미생물,pH,적정산도,비타민 C 함량 및 총 페놀함량의 변화를 조사하였다.
○ 신선편이 사과의 경우 초기 총균수는 2.35logCFU/g이었으나,10℃에서 5일간 유통조건에서 6.58log cfu/g을 나타낸 반면 MAP 포장에 의해서 약 2-2.5log cycle이 감소되었으며, 이산화염소 혼합 포장에 의해서 3-3.5logcycle정도 감소되었다. 비타민 C 함량의 경우 MAP 단독 처리구(CO2::O2:=5%:5%)가 다소 높게 나타났으며,다른 포장구에서는 동등한 효과를 나타내었다.또한 pH 및 적정산도는 이산화염소 가스 혼합처리구가 초기값과 유사하게 나타내었으나 포장구에 따른 큰 차이는 없었다.
○ 신선편이 감자의 경우 초기 총균수는 3.30logcfu/g이었으나,10℃에서 5일간 유통조건에서 8.41logcfu/g을 나타낸 반면 MAP 포장에 의해서 약 2logcycle이 감소되었으며,이산화염소 혼합 포장에 의해서 3-3.4log cycle정도 감소되었다.pH 및 적정산도는 이산화염소가스 혼합처리구와 MAP 포장구간의 차이는 없었다.
○ 무 새싹채소의 경우 초기 무 새싹채소의 총균수는 7.05logcfu/g이었으나,10℃에서 5일간 유통조건에서 8.39log cfu/g을 나타내어 포장구간의 차이를 나타내지 않았다.또한,pH 및 적정산도는 이산화염소 가스 혼합처리구와 MAP 포장구간의 차이는 없어 무 새싹채소에 있어서 포장구에서 미생물 제어방법은 절절하지 않는 것으로 나타났다.
○ 신선편이 혼합채소의 경우 초기 총균수는 5.74logcfu/g이었으나,10℃에서 5일간 유통조건에서 8.12log cfu/g을 나타내었고,MAP 포장에 의해서 총균수가 0.6-1.0log cycle정도 감소되는 효과를 나타내었으며,이산화염소 가스 혼합 포장구에서는 약 1.6-2.0log cycle 정도 감소되는 효과를 나타내었다.pH의 경우 이산화염소 가스 혼합포장구의 pH 변화량이 적었으며,적정산도의 경우도 pH와 유사한 경향을 나타내었다.
나.Activepackaging단독 또는 혼합처리를 통한 적정 포장 방법 선정
○ 함기포장,이산화탄소 5% 및 10%,산소농도 5% 조건과 이산화염소 가스 농도 1ppm 및 2 ppm의 농도에 대한 포장효과를 조사하였다.포장 효과는 신선편이 감자,신선편이 사과,무새싹채소 및 혼합채소에 대하여 실시하였다.전반적으로 이산화염소가스 혼합 포장구에서 우수한 효과를 나타내어,신선편이 감자의 경우 MAP 포장구에 비하여 1.52∼2.28 log cfu/g의 감소효과,신선편이 사과의 경우 0.2∼1.59 log cfu/g의 감소효과,혼합채소는 0.5 logcfu/g의 감소효과를 나타내었다.
다.Activepackaging을 위한 이산화염소가스의 최적 기체 조성비 설정
○ 신선편이 감자의 activepackaging을 위하여 이산화염소가스 농도 0.2∼ 2ppm의 조건에서 미생물의 안정성에 대한 조사를 실시하였다.총균수는 초기 3log cfu/g에서 저장중 증가하여 5.33 log cfu/g로 변화하였다.반면,이산화염소 혼입 포장구는 2.9 log cfu/g에서 4.22log cfu/g로 변화하였다.이산화염소 혼입 포장에 의해서 신선편이 감자 처리구의 총균수를 약 2log cycle이상 감소하는 것으로 나타나 activepackaging에 적절한 소재임을 확인할 수 있으며,이 중 1.0ppm의 농도가 혼입될 경우 신선편이 농산물의 미생물적 안정성을 높일 수 있을 것으로 판단되었다.저온성 균주에 대한 시험에서도 1.0ppm의 농도에 서는 약 40%의 감소효과를 나타내었고 2.0ppm의 농도에서는 60% 이상의 미생물 감소효과가 나타났다.
제 3절 식중독균 저감화 공정 기술 적용에 따른 포장방법 검증
1.포장 방법별 신선편이 감자의 유통 품질 효과 검증 및 확립
○ 시료의 처리는 신선편이 감자의 표준적인 세척방법인 수도수 침지,박피,세척 후 20mm × 20mm × 5mm의 크기로 잘라 100ppm의 NaClO2: 용액에 2분간 침지하였고 이 시료를 수도수로 세척하여 1.0%의 구연산 용액에 2분간 침지하여 냉풍건조하여 사용하였다.이들 시료는 저온 조건에서 일반포장구(LDPE 50μm),MAP 포장구(CO2::O2: = 10%:5%),이산화염소 가스포장구(ClOv 가스 2ppm 유지), MAP+이산화염소가스혼합구[(CO2::O2: = 10%:5%)+2ppm]으로 포장하여 5℃에서 저장하면서 포장 내 기체조성,총균수,저온성 균주, 대장균군 및 대장균,총페놀함량,항산화활성,PPO 활성,갈변도,경도,표면색도 및 관능평가를 관찰하였다.그 결과 경도 및 항산화활성의 변화에 있어서는 처리구별 큰 차이가 나타나지 않았으나,미생물,색도,갈변도 및 관능평가에서는 이산화염소 가스 혼합 포장구에서 우수한 효과를 나타내었다.총균수에서는 이산화염소 가스 혼합 포장구가 대조구 및 MAP 단독 포장구에 비하여 약 2.0log cycle이상 감소하는 것으로 나타났으며,대장균군에서는 1log cycle이상 감소하였다.갈변도의 경우 이산화염소 가스 혼합 포장구가 대조구에 비하여 약 15% 감소효과를 나타내었고 표면색도에서 Hunter-L값이 높게 유지되고 PPO 활성이 낮게 나타내었다.이러한 특성은 관능평가에서도 약 4.5일의 상품성 연장효과를 나타내는 것으로 나타나 이산화염소 가스 혼합 포장구가 신선편이 농산물의 미생물적 안정성을 높이는 효과뿐만 아니라 관능적인 상품성 유지기간의 연장 효과를 나타내는 것으로 판단되었다.
2.포장 방법별 신선편이 사과의 유통 품질 효과 검증 및 확립
○ 시료의 처리는 신선편이 사과의 표준적인 세척방법인 수도수 침지,박피,세척 후 20mm × 20mm × 5mm의 크기로 잘라 100ppm의 NaClO2: 용액에 2분간 침지하였고 이 시료를 수도수로 세척하여 1.0%의 구연산 용액에 2분간 침지하여 냉풍건조하여 사용하였다.이들 시료는 저온 조건에서 일반포장구(LDPE 50μm),MAP 포장구(CO2::O2: = 10%:5%),이산화염소 가스포장구(ClO2: 가스 2ppm 유지), MAP+이산화염소가스혼합구[(CO2::O2: = 10%:5%)+2ppm]으로 포장하여 5℃에서 저장하면서 포장 내 기체조성,총균수,저온성 균주, 대장균군 및 대장균,총페놀함량,항산화활성,PPO 활성,갈변도,경도,표면색도 및 관능평가를 관찰하였다.그 결과 경도의 변화에 있어서는 처리구별 큰 차이가 나타나지 않았으나, 미생물,색도,항산화활성 및 관능평가에서는 이산화염소 가스 혼합 포장구에서 우수한 효과를 나타내었다.총균수에서는 이산화염소 가스 혼합 포장구가 대조구 및 MAP 단독 포장구에 비하여 약 1.5log cycle이상 감소하는 것으로 나타났으며,대장균군에서는 1.6log cycle이상 감소하였다.갈변도의 경우 이산화염소 가스 혼합 포장구가 대조구에 비하여 약 10% 감소효과를 나타내었고 표면색도에서 Hunter-L값이 높게 유지되고 항산화활성은 높게 나타내었다.이러한 특성은 관능평가에서도 약 5.1일의 상품성 연장효과를 나타내는 것으로 나타나 이산화염소 가스 혼합 포장구가 신선편이 농산물의 미생물적 안정성을 높이는 효과뿐만 아니라 관능적인 상품성 유지기간의 연장 효과를 나타내는 것으로 판단되었다.
3.청경채의 포장방법에 따른 미생물 저감화 효과
○ 이 연구는 포장방법에 따른 청경채(BrassicaCompestrisL.sspChinensis)의 미생물 제거 효과를 조사하기 위해 실시하였다.청경채는 일정한 크기의 PP tray에 담은 후 이산화탄소, 산소,질소 및 이산화염소 가스를 주입하여 포장하였다.이산화탄소,산소 및 질소의 주입량은 10:5:85로 하였으며 이산화염소 가스는 약 1.2ppm의 농도로 주입하여 10℃에서 11일간 유지하면서 미생물의 변화를 관찰하였다.초기 총균수는 7.3logCFU/g이었으며 10℃ 의 보관 중에 큰 변화는 나타나지 않았다.이산화염소 가스 혼합구(MAP 혼합 처리구 포함)는 3일 보관시 3.9∼ 4.8log CFU/g 수준이 감소하였다.B.cereus는 초기 청경채에서 5.81 log CFU/g에서 이산화염소 가스 혼참 처리에 따라 3일까지 검출되지 않았으며,Sal entritides는보관 11일동안 3.52∼4.04logCFU/g이 대조구에 비하여 감소하였다.이러한 결과는 이산화염소 가스 및 MAP 혼합 처리구가 미생물의 감소를 위해 효과적인 기술이 될수 있다는 것을 시사한다.
제 4절 생산 및 유통조건에 따른 식중독 미생물 증식억제 기술 효과 검증
1.생산 및 유통조건에 따른 비타민채의 미생물 증식억제 기술 효과
○ 본 연구는 세척과 포장 처리에 따른 미생물 제어효과를 관찰하기 위해 수행하였다.우선 포장 처리에 있어 ClO2: gas농도를 설정하기 위해 농도별 미생물 제어효과를 관찰하였다.4 ppm으로 처리한 후 B.cereus균수는 3.90log CFU/g 감소하였으며 E.coli와 S.aureus또한 3 log CFU/g 이상 감소하였다.4개의 위해균 중 Sal.spp 의 제어률이 가장 낮아 4 ppm 처리 후 2.68 log CFU/g 감소되었다.포장 방법별 균감소률 결과 S.aureus및 Sal. spp의 경우 모두 염소수 세척 후에 각각 0.47및 0.83log CFU/g 수준 감소하였으며 B. cereus와 E.coli의 경우 1log CFU/g 이상 감소 효과를 나타내었다.ClO2: 가스 발생제를 첨가한 MAP 의 경우 포장방법에 관계없이 1log CFU/g 이상 감소되었으며 특히 E.coli 의 경우 3.34log CFU/g 수준까지 감소한 것으로 나타났다.세척방법에 따른 위해 미생물 제어 결과는 수도수 세척(W-T)후 0~0.61logCFU/g수준염소수 0.47~1.47logCFU/g에 비해 낮은 수준이었다.염소수 세척시에도 MAP+ClO2: 가스 발생제 혼합 포장구에서 다른 포장구에 비해 감소효과가 높은 것으로 나타났다.저장동안 미생물 변화 결과는 전반적으로 증가하는 경향을 나타내었으며 무세척구의 경우세척방법에 관계없이 B.cereus 0.74~1.44,E.coli0.73~0.80,S.aureus0.53~0.63및 Sal.spp0.61~0.91log CFU/g 수준 증가하는 경향을 나타내었다.반면에 MAP+ClO2: 가스 발생체 혼합 포장구의 경우 저장기간동안 B.cereus 0.09~0.45 E.coli0.37~0.82,S.aureus 0.17~0.26 및 Sal.spp 0.28~0.93 log CFU/g 수준 증가하는 경향을 보였으며 특히 B.cereus및 S.aureus은 저장기간동안 거의 증가하지 않아 ClO2: 가스발생제 혼합 포장으로 인한 미생물 증가 억제 효과가 있는 것으로 나타났다.특히 염소수 처리 후 MAP+ClO2: 가스발생제 혼합 포장 시 무세척 대조구에 비해 1.15~3.70logCFU/g수준 감소 효과가 있는 것으로 나타났다.이와 같은 연구 결과,염소수에 세척에 의한 초기균 제어와 저장시 ClO2: 가스발생제 혼합 포장을 통한 비타민채의 저장 시 미생물 제어를 통해 품질을 유지할 수 있는 효과적인 방법으로 사료된다.
2.생산 및 유통조건에 따른 신선편이 양배추의 미생물 증식억제 기술 효과
○ 본 연구는 세척과 포장 처리에 따른 미생물 제어효과를 관찰하기 위해 수행하였다.우선 포장 처리에 있어 ClO2: gas농도를 설정하기 위해 농도별 미생물 제어효과를 관찰하였다.4 ppm으로 처리한 후 B.cereus균수는 3.78log CFU/g 감소하였으며 E.coli와 S.aureus또한 3 log CFU/g 이상 감소하였다.4개의 위해균 중 Sal.spp 의 제어률이 가장 낮아 4 ppm 처리 후 2.68logCFU/g 감소되었다.포장 방법별 균감소률 결과 B.cereus,E.coli, S.aureus및 Sal.spp 의 경우 모두 대조구는 약 1 log CFU/g 수준 감소하였다.하지만 ClO2: 가스발생제를 첨가한 MAP 의 경우 2.39~3.03 log CFU/g 수준 감소하여 2 log CFU/g 이상의 감소효과를 나타내었다.세척방법에 따른 위해 미생물 제어 결과는 수도수세척(W-T)후 0~1.49logCFU/g수준 감소하였으며 염소수 세척 후 0.82~3.03logCFU/g 수준 감소하여 다른 처리구에 비해 높은 효과를 나타내었다.저장동안 미생물 변화 결과는 전반적으로 증가하는 경향을 나타내었으며 무세척구의 경우세척방법에 관계없이 B.cereus 4.90~7.98,E.coli4.90~7.50,S.aureus6.35~8.38및 Sal.spp5.68~7.30log CFU/g 수준을 나타내었다.반면에 MAP+ClO2: 가스발생제 혼합 포장구는 저장기간동안 위해균에 관계없이3.98~7.31log CFU/g 수준을 보이며 약 1log scale이상 차이를 나타내었다.특히 염소수 처리 후 MAP+ClO2: 가스발생제 혼합 포장구의 저장 시 무세척 대조구에 비해 2.0~3.07log CFU/g수준 감소 효과가 있는 것으로 나타났다.이와 같은 연구 결과,염소수에 세척에 의한 초기균 제어 및 저장시 ClO2: 가스발생제 혼합 포장 처리는 신선편의 양배추의 저장 시 미생물 제어를 통해 품질을 유지할 수 있는 효과적인 방법으로 사료된다.
■ Bacteriophage를 이용한 신선편이 농수산물의 식중독균 저감화 기술 개발
Ⅳ.연구개발결과
1.Bacteriophage의 분리 및 특성 분석
○ 가축분변으로부터 E.coliO157:H7과 E.coli의 bacteriophage 28종을 분리하여 E. coli와 Salmonellaspp.등을 대상으로 숙주저해범위를 분석하였고,분리된 파지의 형태학적 특성과 분자생물학적 특성을 분석하기 위해 plaqueassay와 투과전자현미경 (TEM)을 사용하여 검경함
○ 분리된 박테리오파지는 모두 Siphoviridaefamily에 속하는 것으로 확인되었으며 구조 단백질과 제한효소를 처리하여 절단 패턴을 분석하여 각각 다른 파지들로 cocktail을 제조하여 사용함
○ 분리된 파지의 안정성을 확인하기 위해 고온,낮은 산도,알코올 처리에서의 박테리오파지 개체수를 확인하였으며,선별된 bacteriophage들은 극한의 pH와 유기산,차아 염소산에서 안정성을 보임
2.Bacteriophage를 이용한 E.coliO157:H7의 생육 억제
○ 분리된 bacteriophage를 사용하여 E.coliO157:H7의 생육억제를 확인한 결과 MOI에 비례한 E.coli의 생육억제 효과를 보임.또한 바이오필름을 형성하여 이를 제거하기 위한 방법을 모색하여 유기산,차아염소산,박테리오파지를 이용하여 단독처리,복합처리에 따른 살균력을 비교하였으며 저 농도 살균소독제의 복합처리에 의한 synergy 효과를 확인함
3.화학적 처리제와 bacteriophage의 복합처리를 통한 E.coli의 저감화
○ 이산화염소 가스를 이용한 E.coli의 저감화를 확인한 결과 10ppm,5분에서 충분히 저해됨을 확인할 수 있었으나,식품에서는 조건에 따라 저해효과가 없었음.이산화염소 가스에서 bacteriophage가 불안정하여 복합처리 시 동시 처리보다 순차적 처리를 해야 할 것으로 사료됨
○ E.coli의 저감화를 위해 bacteriophage와 이산화염소 가스와 sanitizersolution을 복합처리한 결과 ClO2 gas10ppm에서 3분 처리 시 모두 사멸하는 효과를 보임.또한 식품에서 파지와 이산화염소 가스 복합 처리 시 단독처리보다 약 log 2CFU/g정도 저해효과가 있는 것으로 확인됨
4.Murinenorovirus를 이용하여 위생처리를 통한 norovirus의 저감화 효과
○ 여러 화학적 sanitizer를 이용하여 뮤린노로바이러스의 저감화를 확인한 결과 lactic acid와 aceticacid,ethanol에서는 저감화가 되지 않았으며,차아염소산나트륨의 경우 1000ppm 이상의 농도에서 30분간 처리하였을 때 저감화를 확인함. Trisodium phosphate는 2% 이상의 농도에서 저감화를 확인함
5.Bacteriophagesanitizer의 invivosafety확인
○ Bacteriophage를 sanitizer로 활용하기 위해 invivotest를 실시함.실험용 랫드 20마리를 대조군, 실험군 각각 10마리씩 구성하여 진행하였으며, 실험군에는 bacteriophagecocktail을 log 8 PFU/mL의 농도로 매일 1mL씩 4주 동안 경구투여함.실험기간동안 체중과 사료섭취량,증체량,사료효율을 측정한 결과 대조군과 비교하였을 때 유의적인 차이는 나타나지 않았음

○ As a result of sterility validity of disinfectant against 12 bacteria, 4 fungi and 4 yeasts, 30 ppm of slightly acidic electrolyzed water(SIAEW or slightly acidic hypochlorite water) was showed on sterility validity with the level 5-6 log CFU against bacteria, fungi and yeast. Aqueous chlorine dioxide (ACD) with above 10 ppm had sterility validity with above 6 log CFU and the level of 5 ppm ACD was required for the reduction of 5 log CFU. Strong acidic electrolyzed water (SAEW or strong acidic hypochlorite water) and akaline electrolyzed water (AEW or WAEW; weak acidic hypochlorite water) were reduced within the level of 6-7 log CFU at above 50ppm of chlorine concentration.
○ As a result of sterility validity by the simply dipping disinfectant on 4 vegetables such as leafy lettuce and 2 fruits in distribution, SIAEW reduced the level of 2 log CFU/g foodborne pathogen and the consumption of available residual chlorine in the solution was the 10% of level. Additional second and third dipping of disinfectants reduced just 1 logCFU/g of the level on sterility validity and then it is considered that multistep dipping treatments was not effectively. Furthermore, third dipping treatments of disinfectants reduced 2-3 log CFU/g of residual pathogen.
○ Sprout vegetables such as mixed salad and radish sprout, and 2 fruits were tested by dipping in disinfectants of ACD and analyzed sterile effect. As the result, 30ppm of ACD reduced about 2-3log CFU/g of pathogen. However, the pathogen reduction in the radish and brocoli sprout was lower than in leafy vegetables in that differences were caused by theintegrated microorganism and the structure such as stem and dawn.
○ Chlorine dioxide as available residual chlorine in ACD declined into 10% in 10 ppm of the initial concentration after 30 minutes and into 30-40% in the 20ppm and 30 ppm. Lower chlorine dioxide declined in fruits with small surface.
○ The SAEW in 6 samples of leafy lettuce, mixed salad, radish sprout, brocoli, cheery tomato and apple generally reduced 2-3log CFU/g of the pathogen within 20 minutes by first dipping. Microorganism in sprout vegetables such as radish sprout were reduced more effectively at additional steps of second or third dipping than at first dipping. After third dipping sterilization affected the reduction of 3 log and then extension of dipping time is considered to effectively reduce microorganism regardless of physical damage of foods.
○ Reduction effect of LAEW was similar to SAEW in 6 samples of leafy lettuce, mixed salad, radish sprout, brocoli, cheery tomato and apple. Microorganism in sprout vegetables was more effectively reduced in case of the extended time and number of disinfectant dipping. However LAEW affected sterility validity less weakly than SAEW.
○ If there are not excessive organism, parallelt reatment among physical actions to increase contact efficiency is considered to become effective way in order to raise sterility validity
○ In order to analyze sterility validity on pathogenic microorganism, 6 samples of leafy lettuce, mixed salad, radish sprout, brocoli, cheery tomato and apple were contaminated by 107-108 CFU/g of E.coli O157:H7 NCTC 12079 and Salmonella. enterotidis, and then dipped in disinfectants and investigated the reduction rate. After contamination by pathogenic microorganism of E.coli O157:H7 NCTC 12079 와 S.aureus KCCM 12214, leafy lettuce and mixed salad were investigated in sterility validity of SIAEW, SAEW, WAEW and ACD. Tap water reduced about 1 log cycle of pathogen in leafy vegetables, while SIAEW, ACD, SAEW and WAEW reduced about 4,5, 4-5 and 4 log cycle of pathogen, respectively. However pathogen in mixed salad and sprout reduced about 1 log cycle, in that low reduction effect was showed.
2. Assessment of reduction effect by complex sterilization
○ Complex sterilizations with chlorine disinfectants in series were treated on samples of vegetables (leafy lettuce, kale, brocoli sprout, radish sprout and mixed salad) fruits(apple and cheery tomato) and fish (mackerel and pollack). As a result, leafy lettuce and kale included 2 log cycle of residual pathogen after chlorine dioxide in that sterilization effect was the most excellent. As well as apple and cherry tomato included 1-2 log cycle of residual pathogen after sterilization with the 30ppm of SIAEW. However complex sterilization on brocoli and radish sprouts showed the similar effect of treatment with each chlorine disinfectants or the effect with the3-4 log cycle of residual pathogen level with additional 1 log cycle of reduction. Fishes of mackerel and pollack after treatment included maximum about 1 log cycle of reduced pathogen. Ii is considered that to reduce below 2 log cycle is difficult owing to fishery characteristics. After dipping sterilization, the residual chlorine contents in all sample were 0.01-0.02 ppm that were similar level of tap water. Therefore the level is considered to be not significant in sensory evaluation or safety.
○ Physical action after disinfectants treatments as 50 ppm of ACD were used for complex sterilization. As a result, reduction showed excellent effects in order of microbubble, sonic wave and malic acid with additional 1 log cycle of reduction effect.
○ Gaseous chloine dioxide of 20,30,50 and 70 ppm (v/v) were treated and then pathogen in leafy vegetables such as leafy lettuce were reduced about 1, 2, 1 and 1 log cycle, respectively. Colormetric analysis showed that treatment with 50 ppm (v/v) changed insignificantly after 5 minutes. However after 10 minutes, L-value and b-value were increased and a-value was a little decreased, in that sensory quality is considered to be declined. In apple, strawberry and cherry tomato, pathogen reduction was similar effect to leafy vegetables, while a-value were decreased in strawberry with 50 ppm(v/v) treatment for above 5 minutes. However pathogen in radish sprout reduced below 2 log cycle after treatment of 10 minutes. Furthermore pathogen in mixed salad reduced about 4 log cycle at 70 ppm of treatment concentration after 5 minutes with additional 1 log cycle of reduction
○ After treatment of gaseous chlorine dioxide, complex sterilization with hypochlorous acid water reduced about 0.5-1 log cycle of pathogen reduction additionally. Especially, treatment on radish sprout and mixed salad were more effectively than on the other leafy vegetables.
3. Development of reduction technology and process against foodborne pathogen in sprout vegetables
○ As a result from sterile effect of ACD and pressurization treatments on vegetable seeds, the 30 ppm concentration of ACD reduced the 1 log CFU/g of pathogen in the ratio of 1:10 of seed and ACD, and the 6kg/cm2 of pressure
○ The germination rate of radish seed after sterilization was similar to control group and then it is not considered that the condition of the 30 ppm of ACD concentration and the maximum 6 kg/cm2 of pressurization did not affect the germination rate.
○ As a result of analysis on the growth rate and the microbial contamination of radish sprout used the disinfectants with low concentration as cultivation water, the contamination in all treatment group showed similar level of 6 log CFU/g. Moreover, it is considered that disinfectant treatment at 1 day after gemination did not inhibit microbial growth. However the control of pathogen through sterilization of seed is considered as effective way.
4. Design of optimum reduction treatment and processa gainst foodborne pathogen for agricultural and fisheries fresh produces
○ Based on results from sterility validity on single and complex sterilization of disinfectants, SOP (Standard operating procedure) and treatment process of complex sterilization for washing and disinfection processes using single disinfectant were drawn up.The main process was consisted of soaking, washing, disinfection and rinsing and microbubble was designed to be treated in the processes of first washing and rinsing.
○ The process of bacteriophage treatment specified in foodborne pathogen was designed to controlled within 4 log CFU/g of microbial contamination of the level on working step and procedure, cautions, facility, and process control
5. Establishment and field assessment on standard model of treatment process
○ In order to utilize effectively chlorine disinfectants in factories of agricultural and fisheries fresh produces for interested parties such as Cosmic Round Korea Co. and Techwin Co.,we drew up practical facility layout, treatment method and standard model that can reduce foodborne pathogen for manufacturing facilities of private enterprises based on the results of this report. Founded in washing system of leafy and bulbous vegetables, we organized unit processes consisted of soaking, washing, disinfection and rinsing.
6. Development of reduction manual against foodborne pathogen of agricultural and fisheries fresh produces
○ As a result of investigating trends of technologies for manufacturing and management, main management technologies were organized and summarized on the agricultural produces mainly consumed in distribution such as head lettuce, onion, potato, carrot, etc.
○ Management standards on manufacturing facility/utility control, instrument of agricultural and fisheries fresh produces (compartmentation, flow line, industrial water, air conditioner/ventilation, temperature/humidity control, lighting, insect pest control, machinery and equipment control, etc), hygiene management guide (workplace set-up and arrangement, cleaning, washing, disinfection, personal hygiene control, waste water and industrial waste disposal, etc) and integration program of manufacturing facility and process management (GAP,GMP,HACCP, GHP,etc) were developed.
○ According to the types of agricultural produces of potatoes, bulbs and tubers, leaf and stem vegetables, fruit vegetables, fruits, etc, general manufacturing processes were converted and standardized to unit-process and process control manual were developed. Then, unit-process is suggested that can be selected and applied depending on the conditions of manufacturing system.
○ Hygiene control guide was developed by categorized and arranging basic control points to manage hygiene and safety of agricultural and fisheries fresh produces.

Section1. The study on distribution reliability of Fresh-cut product
1. Application of Predictive Microbiology for Microbiological Shelf Life Estimation of Fresh-cut Salad with Short-term Temperature Abuse
○ The aim of this study was to investigate the growth of aerobic bacteria in fresh-cut salad during short-term temperature abuse (4∼30℃ temperature for1, 2, and 3 h) for 72 h and to develop predictive models for the growth of total viable cells (TVC) based on Predictive food microbiology (PFM). The tool that was used, Pathogen Modeling program (PMP 7.0), predicts the growth of Aeromonas hydrophila (broth Culture, aerobic) at pH 5.6, NaCl 2.5%, and sodium nitrite 150 ppm for 72 h. Linear models through linear regression analysis; DMFit program were created based on the results obtained at 5, 10, 20, and 30℃ for 72 h (r2 >0.9). Secondary models for the growth rate and lag time, as a function of storage temperature, were developed using the polynomial model. The initial contamination level of fresh-cut salad was 5.6 log CFU/mL of TVC during 72h storage, and the growth rate of TVC was shown to be 0.020∼1.083 CFU/mL/h (r2 >0.9). Also, the growth tendency of TVC was similar to that of PMP (grow rate:0.017∼0.235 CFU/mL/h;r2=0.994∼1.000).The predicted shelf life with PMP was 24.1∼626.5h, and the estimated shelf life of the fresh-cut salads with short-term temperature abuse was15.6∼31.1h. The predicted shelf life was more than two times the observed one.This result indicates a ‘fail safe’ model. It can be taken to a ludicrous extreme by adopting a model that always predicts that a pathogenic microorganism will grow even under conditions so strict as to be actually impossible.
2. Microbial Analysis of Marketplace Fresh-cut Mackerel
○ The purpose of this study was to determine the contamination status of marketplace fresh-cut mackerel. The samples of fresh-cut mackerel were collected from department stores, wholesale markets and small scale of fish-markets within the Seongnam and subsequently analyzed for food-borne pathogens. Total aerobic counts ranged from 2.50 to 5.10logCFU/g. Higher levels of contamination was detected in ready made mackerel of same department store. Coliform counts of 1.51 log CFU/g were detected in fresh-cut mackerel averagely. The maximum coliform counts of 3.26 log CFU/g was detected in fresh-cut mackerel purchased at a large scale of discount store. The prevalence of pathogens, namely Staphylococcus aureus and Bacillus cereus,in the fresh-cut mackerel were 100% and 8.3% respectively. The prevalence of E.co.liwas 25% in ready made samples and in custom made samples respectively. Therefore, these results suggest that the tightening supervision systems on marketplace are needed in terms of microbial safety as well as quality and shelf-life of fresh-cut mackerels.
Section 2. Development of food poisoning bacteria reduction level maintaining technology for fresh cut agricultural products, and Setup and effect analysis of appropriate package method
1. Development of food poisoning bacteria reduction technology
1) Development of chlorine dioxide generator to maintain a reduced level through active packaging
○ We aimed to measure maintaining capability by reacting sodium hypochlorite solution with hydrochloric acid solution to generate chlorine dioxide, which is the most widely used method to generate chlorine dioxide. To do this, an appropriate pH concentration condition was set as a type of solutes was used differently. The gas of chlorine dioxide was generated well with a range of pH 3 to pH 4 and the following solute types were effective: citrate buffer, HCl-NaOH buffer, Lactic acid buffer, succinic acid buffer, and Universal buffer. The gas generation of chlorine dioxide with the above buffers was studied for about 20 day sand the study result showed that Universal buffer had higher persistence generation rate than HCI buffer. In addition, gas generation of chlorine dioxide according to a ratio of a sodium chlorite amount and acid concentration was studied.The study result showed that chlorine dioxide gas was generated significantly when a sodium chlorite amount was contained above a certain amount (1,000ppm and over) whereas acid concentration had no significant effect. To pack the chlorine dioxide gas generator, various films were used as a gel packaging material. It was found that a PE film type has a high emission effect on chlorine dioxide gas so their permeability was shown in order of LLDPE>LDPE>HDPE. In general, it has been known that permeability of chlorine dioxide gas was 8 and 13 times higher in LDPE and LLDPE than HDPE respectively.This study found that it was effective to use LDPE as an emission material to pack the chlorine dioxide gas generation gelto reduce harmful microbes in fresh agricultural products during the distribution.
2)Analysis on the effect of the active packaging application against food poisoning bacteria
○ The analysis result of the growth inhibition effect against harmful microbes using chlorine dioxide gas showed that bacillus cereus was not detected as low as ND level in the chlorine dioxide gas treatment group above 1.0ppm while E.Coli was also reduced to about 1/5 of control group in chlorine dioxide gas concentration of 2.0ppm. Salmonella typhi was reduced to 1 log CFU in 6ppm concentration while it was reduced to a N.D. level under 2.0 ppm concentration.Staphylococcus aureus was also reduced to a N.D. level in chlorine dioxide treatment group with 2.0ppm while it was reduced to about 1/4 level under 1.0ppm concentration compared to control group.
○ The growth inhibition effect against food poisoning bacteria (Bacillus cereus) with respect to oxygen concentration condition and chlorine dioxide gas mixture was analyzed and the result showed that Bacillus bacteria was proliferated well when oxygen was added to the inside of the package due to the characteristic of Bacillus bacteria thereby showing the growth of bacteria on the smeared agar plate. In contrast, some of bacteria growth was suppressed in nitrogen 100% filling packaging group. In addition, chlorine dioxide gas mixture packaging group showed a thin layer of bacteria growth, indicating a considerable inhibition performance. Overall, chlorine dioxide gas treatment group showed inhibition of bacillus cereus bacteria growth. A mixture group of 90% and 100% oxygen concentration showed measurable colony counts. In summary, with high oxygen concentration, chlorine dioxide gas mixture treatment group showed a high inhibition effect against bacteria growth.
○ The growth inhibition effect against food poisoning bacteria (Bacillus cereus) with respect to carbon dioxide concentration condition and chlorine dioxide gas mixture was analyzed and the result showed that carbon dioxide modified atmosphere packaging (MAP) group had less growth of bacteria than control group overall while chlorine dioxide gas mixture packaging group had the higher effect than other groups. In chlorine dioxide gas mixture packaging group, 30%,70%, and 100% carbon dioxide treatment group showed complete inhibition of bacteria growth while chlorine dioxide mixture packaging group with 20%,40%,50%,80%, and 90% concentration of carbon dioxide showed some inhibition of bacteria growth up to measureable colony counts.
○ The growth inhibition effect against food poisoning bacteria (Bacillus cereus) with respect to MAP condition was analyzed and the result showed that MAP packaging group had no significant difference in Bacillus cereus compared to that of control group except for carbon dioxide 10%/oxygen 5% mixture. However, all of chlorine dioxide gas mixture treatment group showed a significant difference with control group and MAP-only packaging group. In chlorine dioxide gas mixture treatment group,10%/5% and 20%/5% groups of a mixture ratio between carbon dioxide and oxygen reduced the bacteria counts up to 1 log CFU level. Chlorine dioxide mixture packaging group had a inhibition effect against microbial growth in all agars. In particular, better inhibition effect was found with 5% of oxygen concentration.
2. Setup and effect analysis of appropriate package method
1)The study of inhibition effect against microbial growth in fresh cut agricultural products by applying chlorine dioxide gas
○ To study the inhibition effect against microbial growth in fresh cut agricultural products by applying chlorine dioxide gas, apple, potato, radish sprout, and mixed leaves vegetable were cut into packaging followed by being treated with chlorine dioxide gas concentration and stored under 10℃ of distribution condition for five days.Then changes in microbes were studied. The result showed that apple indicated a ND level under the treatment condition of 0.5ppm and over while potato indicated a reduction by about 1.8 log cycle with 1.0 ppm concentration and ND level with 2.0 ppm concentration. Radish sprout revealed a reduction by about 1 log cycle with 1.0ppm concentration and about 1.6 log cycle with 2.0 ppm concentration. In contrast, mixed leaves vegetable showed a reduction only by about 1.3 log cycle with 2.0 ppm concentration.
2) A growth inhibition effect against harmful microbe (E.coliO157:H7) of fresh cut agricultural products by applying chlorine dioxide gas
○ A growth inhibition effect against harmful microbe (E.coliO157:H7) of fresh cut agricultural products after chlorine dioxide gas was applied was studied. The result showed that cube-shape cut potato was contaminated with a level of about 5 log CFU/g and E.coli O157:H7 505B contaminated inside potato cannot be controlled even if it was exposed to chlorine dioxide gas of 2.0ppm for one day. This result was obtained because bacterial inoculation solution was absorbed into a cutgap of potato, which made the control difficult. The contamination level of apple was about 5 log CFU/g whereas apple in chlorine dioxide treatment group had 3log CFU/g, showing a reduction of about 2 log CFU/g. Radish sprout had difficulty in controlling E.coli O157:H7 505B while lettuce and bok choi, which were contaminated containing about 6 log CFU/g, had a reduction effect by about l log CFU/g after chlorine dioxide treatment.
3) The study of growth inhibition effect against harmful microbes during food distribution of fresh cut agricultural products by applying chlorine dioxide gas
○ To study a growth inhibition effect against harmful microbes during food distribution of fresh agricultural products that were treated with chlorine dioxide gas, artificial contamination of four different types of food poisoning bacteria was generated in mini paprika and then control effect was studied. S. aureus had a level of 5.60 log CFU/g before chlorine dioxide gas was treated. Once chlorine dioxide gas of 2.0ppm was added to packaging, no bacterial growth was generated even after seven days. With 6ppm and 1.0ppm, no bacterial growth was observed initially but since then some bacterial growth was found over time thereby showing no difference with control group. The growth of B. cereus was suppressed initially in chlorine dioxide gas treatment group of 2.0ppm. However, after seven days, no difference was found with control group. Chlorine dioxide gas treatment group of 0.5ppm and 1.0ppm did not show any difference with control group from the beginning, which indicated a difficulty in controlling B. cereus via chlorine dioxide gas of low concentration. Mini paprika contaminated with E. coli O157:H7 505B와 Sal. enteritidis containing about 3∼4 log CFU/g showed a good control of the bacteria even with chlorine dioxide gas of relatively low concentration in contrast with S. aureus and B.cereus.The bacterial growth was not observed even after seven days so it was found that chlorine dioxide gas was effective in controlling E.coli O157:H7 505B and Sal. enteritidis.
3. Microbial growth inhibition effect on fresh agricultural products and quality analysis according to packaging method
○ To observe the effect of microbial growth inhibition and quality maintaining in fresh agricultural apple, changes in microbial growth and nutrition facts were observed with various packaging conditions. The study result showed that microbial growth inhibition effect due to packaging condition revealed effective growth inhibition in MAP group and no significant difference was found in gas concentration while a superior effect on changes in nutritional contents was revealed in a group of carbon dioxide concentration 5% and oxygen concentration 5%.
○ To analyze the effect of active packaging that aims to microbes and quality maintaining in fresh agricultural apple, a mixture of 5% and 10% of carbon dioxide with 5% of oxygen concentration was selected. Then, 1.0ppm and 2.0ppm of chlorine dioxide gas were mixed to be used in the experiment. The total count of fresh agricultural apple was 2.35 log cfu/g initially but it was increased to 6.58 log cfu/g after five days under 10℃ of distribution condition. On the contrary, MAP packaging reduced about 2-2.5 log cycle while chlorine dioxide mixture packaging reduced 3-3.5 log cycle.Therefore,it was found that active packaging was an effective means for microbial control.
○ To observe the effect of microbial growth inhibition and quality maintaining in fresh agricultural potato, changes in microbial growth and nutrition facts were observed with various packaging conditions. The study result showed that MAP showed a range of 5.5-6.3 log cfu/g, which showed a reduction effect by 2 log cycle compared to control group. In MAP group, the best effect was shown with a group of 20% of carbon dioxide concentration and 10% of oxygen concentration, showing 5.5 log cfu/g.In addition, nutrition fact analysis showed that the best result was obtained with 5% of carbon dioxide concentration and 5% of oxygen concentration.
○ To observe the effect of microbial growth inhibition and quality maintaining in fresh radish sprout vegetable, changes in microbial growth and nutrition facts were observed with various packaging conditions. The study result showed that general packaging group and MAP group had similar trend with each other with respect to microbial growth while nutrition fact analysis showed a good resultin LDPE packaging group and 30% of carbon dioxide concentration and 10% of oxygen concentration packaging group.
○ To observe the effect of microbial growth inhibition and quality maintaining in fresh agricultural mixed vegetable, changes in microbial growth and nutrition facts were observed with various packaging conditions. Overall, MAP group showed fewer bacterial counts than aerobic packaging group but the difference was minimal. Regarding changes in the nutrition fact, a packaging group of 10% carbon dioxide concentration and 5% oxygen concentration showed a good result.
4. Setup of active packaging treatment condition
1) Analysis of microbial growth inhibition and quality by different packaging method and active packaging
○ To treat a sample, fresh cut potato and apple were cut into a size of 20mm × 20mm × 5mm, and were packaged under a low temperature condition with general packaging group (LDPE 50μm), MAP packaging group (CO2:O2 = 10%:5% orCO2:O2 = 5%:5%), MAP + chlorine dioxide gas mixture packaging group [(CO2:O2 = 10%:5%)+2ppm] and stored under 10℃. Total bacterial counts, pH, titratable acidity, vatamin C conent and total phenolic content were observed.
○ The observation result of fresh cut apples showed that initial total microbial count was 2.31 log CFU/g. The control treatment increased 4.27 log CFU/g at 5 th day, but total microbial count of MAP and active packaging treatment decreased by 2∼3.5 log CFU/g at 5 th day. Vitamin C content showed slightly higher in MAP treatment. Also, pH value and titartable acidity were similarly showed in the all packaging methods. Therefore, it was found that active packaging was an effective means for microbial control.
○ The observation result of fresh cut potatos showed that initial total microbial count was 3.03 log CFU/g. The control treatment increased 5.11 log CFU/g at 5 th day, but total microbial count of MAP and active packaging treatment decreased by 2∼3.4 logCFU/g at 5 th day. Also, pH value and titartable acidity were similarly showed in the all packaging methods. Therefore, it was found that active packaging was an effective means for microbial control.
○ The observation result of radish sprouts showed that initial total microbial count was 7.05 log CFU/g. The control treatment increased 1.34 log CFU/g at 5 th day, and all packaging method didn't show reduction of the microorganisms. Also, pH value and titartable acidity were similarly showed in the all packaging methods.
○ The observation result of mixed salads showed that initial total microbial count was 5.74 log CFU/g. The control treatemnt increased 2.38 log CFU/g at 5th day, but total microbial count of MAP and active packaging treatment decreased by 0.6∼2.0 log CFU/g at 5th day. pH value and titartable acidity were similarly showed in the all packaging methods. It was found that active packaging was an effective means for microbial control.
2) Selection of appropriate packaging method through single or mixed treatment
○ A packaging effect was studied with respect to aerobic packaging, condition of 5% of carbon dioxide and 10% of oxygen concentration, and 1ppm and 2ppm of chlorine dioxide gas concentration. The experiment of packaging effect was conducted with fresh agricultural potato, apple, radish sprout, and mixed vegetable. Overall, chlorine dioxide gas mixture packaging group showed a superior result. For example, fresh agricultural potato had better reduction effect by 1.52∼2.28 log cfu/g, fresh agricultural apple by 0.2 ∼1.59 log cfu/g, and mixed vegetable by 0.5 log cfu/g compared to those of MAP packaging group.
3) Setup of optimum gas composition ratio of chlorine dioxide gas for active packaging
○ Microbial stability under 0.2 - 2 ppm concentration condition of chlorine dioxide gas was studied for active packaging of fresh agricultural potato. The total bacterial count was initially 3 log cfu/g and then increased into 5.33 log cfu/g in the middle of storage. In contrast, the total bacterial countin chlorine dioxide mixture packaging group was changed from 2.9 log cfu/g to 4.22 log cfu/g. That is, the total bacterial count in fresh agricultural potato treatment group was reduced by about 2 log cycle thanks to the chlorine dioxide mixture packaging, which indicated it was an appropriate material for active packaging. If 1.0ppm concentration is mixed, it would be expected to increase microbial stability of fresh agricultural products. A test of low temperature-type strain also showed that about 40% of reduction effect was revealed with 1.0ppm concentration and 60% of microbial reduction effect was found with 2.0ppm concentration.
Section 3. Verification and establishment of distribution quality effect per packaging method
1. Verification of distribution quality effect for fresh cut agricultural potato
○ To treat a sample, a standardized cleaning method for potato, which followed tap water immersion, peeling, and cleansing, was conducted. Then, potato was cut into a size of 20mm × 20mm × 5mm and immersed in NaClO2 solution of 100ppm for two min. The sample then was cleansed with tap water followed by being immersed in citric acid solution of1.0% for two min. and dried by cool air. Then, the samples were packaged under a low temperature condition with general packaging group (LDPE 50μm), MAP packaging group (CO2:O2 = 10%:5%), chlorine dioxide gas packaging group (ClO2 gas 2ppm maintained), MAP + chlorine dioxide gas mixture packaging group [(CO2:O2 = 10%:5%)+2ppm] and stored under 5℃. Gas composition in packages, total bacterial counts, low temperature-type strain, coliforms and E.coli, total phenolic content, antioxidant activity, PPO activity, browning, hardness, surface color and sensory evaluation were observed. The observation result showed that no significant difference in changes in hardness and antioxidant activity was found between treatment groups whereas chlorine dioxide gas mixture packaging group showed a superior effect on total bacterial count, surface color, browning, and sensory evaluation. Chlorine dioxide gas mixture packaging group reduced 2.0 log cycle in the total bacterial count while it reduced 1 log cycle in coliforms more than control and MAP only packaging groups. Chlorine dioxide gas mixture packaging group showed about 15% reduction effect in browning while maintaining ah igh Hunter-L value in surface color and showing low PPO activity. The above characteristic resulted in longer merchantability of about 4.5 days in sensory evaluation as well, revealing that chlorine dioxide gas mixture packaging group is effective in not only increasing microbial stability of fresh agricultural products but also extending merchantability in terms of sensory evaluation.

The above characteristic resulted in longer merchantability of about 5.1 days in sensory evaluation as well, revealing that chlorine dioxide gas mixture packaging group is effective in not only increasing microbial stability of fresh agricultural products but also extending merchantability in terms of sensory evaluation.
3. Effect of Microbial Removal on Bock choy (Brassuca Compestris L ssp Chinensis) with Different Packaging Methods
○ This study was carried out to investigate the microbial removal effect of bok choy (Brassica Compestris L. ssp Chinensis) by different packing methods. The bok choy pul into PP tray and injected partial pressure of CO2, O2, N, and ClO2 gas. The ratio of CO2, O2, N, of MAP condition was 10 :5 :85 and control was 0 :20 :80. The concentratiou of ClO2 gas was 1.2 mg/L and storage temperature was 10℃. Initial total bacterial count of control was 7.3 log CFU/g did not chang eremarkably during storage at 10℃. The microbial count of groups added ClO2 (ClO2 and MAP+ClO2) decreased 3.9 ∼4.8 log CFU/g level at 3rd day. B.cereus count in initial bok choy was 5.81 log CFU/g while ClO2 gas groups were not detected at 3rd day. Also the result of Sal. entritidis counts of ClO2 gas group was reduced by 3.52∼4.04 log CFU/g compared to control at11th day. These results suggest that the treatments of ClO2 gas and MAP can be an effective technology for microbial reduction.
Section 4. Technological improvement on maintaining the reduction effect via field application experiments per item
1.Removal Effect of Food Hazard Microorganism in Tah Tasai chinese cabbage (Brassica campestris var. narinasa) by Different Packaging Method
○ This study was investigated for removal effect of food hazard microorganism in Tah Tasai were treated in packing film contained different concentration of ClO2 gas (0-4 ppm). The result of microbial reduction of Tah Tasai treated with 4 ppm ClO2 gas concentration, B. spp decreased by 3.90 log CFU/g and E. coli and S. aureus was reduced above 3 log CFU/g. Initial B. spp and Sal. spp counts were 6.53 and 6.50 log CFU/g and those of chlorine washing treatment decreased by 4.90 and 5.68 log CFU/g respectively. And B.spp and Sal.spp count of ClO2 beads in MA packing treatment (MA+ClO2 beads) decreased by > 2 log CFU/g at 1 days. The result of B.cereus and S.aureus removal with different washing method (no washing, water washing, chlorine water washing), water washing treatment (W-T) and chlorine water washing treatment (CA-T) decreased up to 1.15∼ 3.70 log CFU/g compared with control (no washing). Regardless of packaging and washing, MAP+ClO2 beads treatment was most effective method for removal food hazard microorganism. Results suggest gaseous ClO2 is effective in inactivating different microorganisms present on the surface of Tah Tasai chinese cabbage.

○ This study was investigated for removal effect of food hazard microorganism in cabbage were treated in packing film contained differentcon centration of ClO2 gas(0-4 ppm). The result of microbial reduction of cabbage treated with 4 ppm ClO2 gas concentration, B. spp decreased by 3.78 log CFU/g and E. coli and S. aureus was reduced above 3 log CFU/g. Initial B. spp and Sal. Typimurium counts were 6.00 and 6.45 log CFU/g and those of no washing treatment decreased by 5.30 and 6.40 log CFU/g respectively. But B. spp and Sal. Typimurium count of ClO2 beads in MA packing treatment (MA+ClO2 beads) decreased by >2 log CFU/g. The result of Sal. Typhimurium removal with different washing method (no washing, water washing, chlorine water washing), water washing treatment (W-T) and chlorine water washing treatment (CA-T) decreased up to 1.00 log CFU/g compared with control (no washing). Regardless of packaging and washing, MAP+ClO2 beads treatment was most effective method for removal food hazard microorganism. Results suggest gaseous ClO2 is effective in inactivating different microorganisms present on the surface of cabbage.
■ Development of reduction technologies for food-borne pathogens in agricultural and fishery fresh produces using bacteriophage
Ⅳ. Results
1. Isolation and characterization of bacteriophages
28 bacteriophage of E. coli O157:H7 and non-pathogenic E. coliwere isolated from cattle feces. And they were analyzed for host spectrum, morphological and biological characteristics by plaque assay, transmission electron microscope(TEM). All phages isolated belonged to the Siphoviridae family. Five phages were variously selected as bacteriophage cocktail by restriction enzyme patterns and protein structure. To determine the stability of isolated phages, the factors such as high temperature, various pH, and ethanol were applied to the phages. Five bacteriophages as cocktail were stable in low pH, organic acids, and sodium hypochlorite.
2. Growth inhibition of E.coli O157:H7 using bacteriophage
And they showed effect of growth inhibition of E. coli O157:H7 In proportion to MOI. The ways to remove biofilm on food and stainless steel surface, there were compared and analyzed sterilizing by single treatment and complex treatment using organic acid, sodium hypochlorite, and bacteriophage. Synergy effect were confirmed under a low concentration of sanitizer on condition of phage combined treatment with sanitizer.
3. Reduction of E. coli by complex treatment
E. coli was enough inhibited by 10ppm of ClO2 gas for 5 min, but it depends on the situation in foods. It is necessary to sequentially processing than the simultaneous processing because bacteriophage is unstable in ClO2 gas. The results of complex treatment with bacteriophage, sanitizer solution, and ClO2 gas were an effect of all inhibition on condition of 10 ppm of ClO2 gas for 3 min. And complex treatment with bacteriophage and ClO2 gas reduced to approximately log 2 CFU/g more than single treatment in cabbage.
4. Reduction effect of murine norovirus as a surrogate for human norovirus using chemical sanitizer
In case norovirus and murine norovirus as a surrogate for norovirus, they showed no reduction effect using lactic acid, acetic acid, and ethano, but reduction effect confirmed only sodium hypochlorite at a concentration of 100ppm and upper 2% trisodium phosphate.
5.In vivo safety of bacteriophage as a sanitizer
To investigate the safety of bacteriophage as sanitizer, assessment of the immune response has been performed in rats. Control group and experiment group contained each 10 rats and rats ofexperiment group were orally fed with log 9 PFU/mL of bacteriophage cocktail once a day for 4 weeks. For 4 weeks, it examined the body weight, the feed intake, the cytokine profiling, and the histopathological changes of liver, kidney, and spleen in rats. There were confirmed no significant change by bacteriophage in body weight, feed intake, cytokine level. After experiments, the result of autopsy showed significantly difference of male's liver, female's ovary (left) compared with control group. But according to see with the naked eye, all organs observed on significant change both control and experiment groups, and also histopathological analysis of liver, kidney, and spleen observed no significant change.
