Flavonoids are polyphenolic compounds with various biological activities in vivo. Many flavovoids show antioxidant activity by scavenging free radicals and chelating some metal ions. In addition, some flavonoids show prooxidant activity which can induce apoptosis of various cancer cells. But, the details about environmental conditions which determine their antioxidant and prooxidant activities, are not yet understood. This study was aimed to characterize the in vitro conditions which determine the antioxidant and prooxidant activities of epigallocatechin gallate (EGCG), the major flavonoid of green tea, by evaluating the survival rate and the generation of free radicals in Chang Liver cells, and the lipid peroxidation of liposomes. Although EGCG did not show toxic effect in low concentrations, at the concentrations higher than 20 μM, it induced the death of cells in concentration dependent manner. In addition, EGCG generated the free radicals in the cells and also induced lipid peroxidation of liposomes. All these toxic effects of EGCG on the cultured cells and liposomes were inhibited by N-acetylcysteine (NAC) and deferoxamine (DFX), a free radical scavenger and an iron ion chelator, respectively. These results suggest that the concentration EGCG and the presence of iron ions may determine if EGCG work as antioxidant or prooxidant, and that NAC and DFX may be useful in inhibiting the EGCG-induced oxidative damage.
