본 연구는 커피원두의 배전공정중 변화되는 주요 화학성분을 HPLC, IC, GC, GC-MS를 이용하여 정성 및 정량하고 전문패널에 의하여 headspace aroma에 대한 관능적 평가를 실시하여 주요 휘발성 향기성분과 headspace aroma의 관능적 지수와의 상관관계를 고찰하였다. 또한 커피원두중의 비휘발성 성분 분석결과를 토대로 model solution을 만든 다음 반응온도 및 시간에 따른 커피향기성분 발현 정도 및 향 전구체에 대한 연구를 하고자 하였다. 연구의 실험모델로 세계 3대 커피원종으로 불리우는 마일드(Mild), 브라질(Brazil), 로브스타(Robusta)를 커피원두의 시료로 선정하였고, 위 커피원두를 로스타(Roaster)를 이용하여 연한 배전(lightroasting), 중간 배전(medium roasting), 강한 배전(dark roasting)으로 하여 배전공정 전, 후의 주요 화학성분의 변화 및 휘발성 향기성분 분석결과와 headspace aroma와의 관능적 상관관계를 고찰하였고, model solution을 통한 커피향기성분 발현 정도를 연구한 결과는 다음과 같다. 비휘발성 성분의 전체적인 경향은 배전정도가 증가할수록 대부분 감소하는 것으로 나타났다. Amino acids, trigonelline, chlorogenic acid 및 monosaccharides 등은 배전공정중 Maillard 반응, Strecker 분해반응, trigonelline 분해반응, chlorogenic acid 분해반응 및 sugars 분해반응에 의해서 화학적으로 분해되어 휘발성 향기성분의 생성에 많은 영향을 주는 것으로 나타났다. 특히 강한 배전공정에서 총 amino acids의 경우 커피원두를 기준으로 약 30% 정도 감소하였고, trigonelline은 50~60% 수준, chlorogenic acid는 90% 이상, 그리고 monosaccharides는 25~34% 정도 감소하였다. 휘발성 향기성분의 전체적인 경향은 배전정도가 증가 할수록 증가하는 것으로 나타났다. 이와 같은 결과는 배전공정중 휘발성 향기성분의 전구체로서 역할을 하는 amino acids, trigonelline, chlorogenic acid 및 monosaccharides등이 여러반응에 의해서 분해되어 휘발성 향기성분들을 보다 많이 생성하기 때문인 것으로 추정된다. 그러나, furfural, diacetyl(2,3-butanedione) 및 2,3-pentanedione등은 감소하는 것으로 확인되었는데, 이는 furfural의 경우 Maillard 반응중 생성속도 보다 분해속도가 크기 때문인 것으로 그리고, α-dicarbonyl의 경우 배전공정이 강해질 수록 환원되기 때문인 것으로 추정된다. 배전정도가 증가할수록 Aroma Index (D+P)/B 즉, (diacetyl +2,3-pentanedione)/2-butanone비율은 연한, 중간, 강한 배전으로 갈수록 낮아지는 경향을 나타냈으며, 커피품종별로는 Arabica가 Robusta에 비해서 월등히 높았으며, Brazil의 경우 Arabica와 거의 비슷한 수준이었다. Aroma Index M/B(2-methylfuran/2-butanone)의 비율은 연한, 중간, 강한배전으로 갈수록 증가하는 경향이 매우 뚜렷하였으며, Robusta 및 Brazil 배전커피가 Arabica에 비해서 상대적으로 높게 나타났다 휘발성 향기성분의 지표와 관능적 특성과의 상관관계를 규명하기 위하여 배전커피의 headspace volatiles에 대한 분석결과를 토대로 도출된 Aroma Index (D+P)/B 및 M/B와 overall headspace aroma의 관능적 지수와는 높은 상관관계가 있는 것으로 나타났다. Aroma Index (D+P)/B와 관능적 지수와의 상관관계를 나타내는 상관계수(correlation coefficient, r)는 Colombia의 경우 0.9885, Tanzania 0.9814, Ethiopia 0.9895, India 0.9991, Thailand 0.9669 그리고 Brazil 배전커피의 경우 0.9937로 매우 높았으며, 또한 Aroma Index M/B와 관능적 지수와의 상관관계를 나타내는 상관계수(correlation coefficient, -r)는 Colombia의 경우 0.9982, Tanzania 0.9834, Ethiopia 0.8691, India 0.9997, Thailand 0.9998 그리고 Brazil의 경우 0.9365로 나타났다. 본 연구에서 확인된 Aroma Index (D+P)/B 및 M/B를 이용하여 배전커피의 headspace aroma의 품질특성을 평가 할 수 있는 Index로서 활용 가능성을 보여주었다. Model solution을 통한 커피 향기성분 발현 및 향 전구체에 대한 연구결과 반응온도 및 시간에 따른 각각의 향 발현 특성은 약한 반응조건(210℃, 480초)에서 발현된 커피 향기성분의 대부분은 aldehyde 화합물로써 acetaldehyde, 2-methyl propanal, 3-methyl butanal 및 2-methyl butanal등이었고, ketone류로는 acetone이, furan 화합물로는 furan, furfural 및 furfuryl acetate등이, 그리고 ethyl alcohol이 발현되었다. 그러나 diacetyl(2,3-butanedione), methyl ethyl ketone, 2,3-pentanedione 그리고 2-methylfuran, pyrazine, 2-methylpyrazine, pyridine, furfuryl alcohol, 2,5-dimethylpyrazine, methyl furfural 및 trimethyl pyrazine등은 극미량 또는 trace로 발현되었다. 중간 반응조건(220℃, 550초) 및 강한 반응조건(230℃, 610초)에서 발현된 대부분의 휘발성 성분들은 약한 반응조건에서 생성된 것에 비해서 상대적으로 분자량이 크고 boiling point가 높으며 그리고 relative volatility가 낮았다. 3가지 반응조건별에 따른 6종류의 model solution의 total aroma 함량은 강한 반응조건 > 중간 반응조건 > 약한 반응조건의 순으로 total aroma 함량이 많았고, 또한 model s olution을 통한 커피원두 품종별에 따른 total aroma 함량 수준은 India > Thailand > Brazil > Ethiopia > Tanzania > Colombia 순으로 나타났는데, 이는 커피원두의 총 amino acids의 함량수준인 India > Thailand > Brazil > Ethiopia > Colombia > Tanzania 순서와 거의 유사한 경향을 나타냈다. 이와 같은 결과는 커피 향 발현에 직접적으로 영향을 미치는 총 amino acids 함량이 많을수록 보다 많은 커피 향이 발현되는 것으로 추정된다.

The purposes of this study were : firstly, to investigate the changes of chemical constituents in six different green coffee beans during roasting. Thirdly, to investigate the flavor development using model solution which were prepared based on the analytical data obtained from the chemical analysis of the green coffee beans. Total amino acids were gradually decreased, but cystine, serine, lysine and arginine were markedly decreased during roasting. On the other hand, glutamic acid, glycine, alanine, valine, isoleucine, leucine and phenylalanine were slightly decreased and increased during roasting. Total amino acids, alanine and phenylalanine in Robusta coffee beans were higher than those of Arabica. Caffeine contents in coffee beans did not changed during the roasting process. The content of caffeine in Robusta coffee beans (2.0%) was higher than that of Arabica (1.0%). Trigonelline and chlorogenic acid were rapidly degraded during dark roasting and were lost 50~60% and 90~95%, respectively. The content of trigonelline in Arabica coffee beans (0.87%) was slightly higher than that of Robusta (0.74%), but the content of chlorogenic acid in Robusta coffee beans (4.82%) was higher than that of Arabica (4.38%). The content of total monosaccharides in coffee beans were gradually decreased during the roasting process. Mannose was st able during roasting, but arabinose was labile thermally. A total of 40 volatile compounds on roasted coffee beans were detected using a static headspace method and 27 volatile compounds were identified. Robusta green coffee beans contained higher concentration of acetaldehyde, acetone, furan, 2-methylpropanal, 2,3-butanedione, 3-methylbutanal, 2-methylbutanal, 2,3-pentanedione and furfuryl acetate than those of Arabica, although those compounds were low in concentration. Except furfural, 2,3-butanedione and 2,3-pentanedione, most of volatile compounds were increased throughout the roasting process. Robusta roasted coffee beans had a considerably higher content of aldehydes in Arabica. Twofold higher amounts of diacetyl (2,3-butanedione) and 2,3-pentanedione were formed in Arabica coffee beans compared to the Robusta. The ratio of (diacetyl + 2,3-pentanedione) / 2-butanone as used Aroma Index (D+P)/B, was significantly decreased during roasting for Arabica as well as for the Robusta coffee beans. However, the ratio of 2-methylfuran/2-butanone used Aroma Index M/B , was considerably increased with dark roasting for both Arabica and Robusta coffee beans. The correlation between Aroma Index (D+P)/B and Aroma Index M/B and sensory score of overall headspace aroma of roasted coffees was good in correlation. The correlation coefficient (r) between Aroma Index (D+P)/B and sensory score positively was positively related with a high correlation. Conversely, Aroma Index M/B negatively correlated with sensory scores for headspace aroma.
