본 연구는 phytase 생산 균주의 분리·동정 및 이용성 증진에 관한 것으로 1) phutase 생산성이 높은 균주의 분리 및 생산되는 phytase의 특성, 2) phytase 생산에 미치는 요인 및 저렴한 배지를 이용한 생산 조건, 3) phytate 분해 및 내산성과 내담즙성이 우수한 유산균의 분리 및 발효사료 제조용 균주와 생균제로서의 이용가능성, 4) phytase를 비롯한 복합 효소를 분비하는 Bacillus sp.의 분리 및 대두박을 이용한 대량 생산 조건의 검토 등 4 가지의 실험을 수행하였으며, 그 결과는 아래와 같다. 실험 1. 높은 phytase 활성을 나타내는 P. waksmanii AM301의 배양을 통해 얻은 배양액을 ammonium sulfate에 의한 침전과 CM및 DEAE-Sepharose CL-6B column chromatography를 이용하여 분리·정제한 phytase의 분자량은 SDS-PAGE 및 FPLC system을 이용한 gel filtration을 수행한 결과 각각 24 및 47 kDa이었으며 dimer 구조로 파악되었다. 정제된 phytase의 효소 활성을 나타내는 최적 pH 및 온도는 각각 pH 4.5 및 55℃였으며, 등전점은 약 5.0이며 Na-phytate에 대한 Km 및 Vmax는 각각 0.09 mM 및 37.04 μmol Pi/mg/ml이었고 Fe^(++), Zn^(++) 및 Cu^(++)와 같은 금속이온에 의해 상당부분의 활성이 저해되었다. 실험 2 .P. waksmanii AH1301에 의한 phytase 생산은 배양 온도, P와 Ca 농도 및 배지의 초기 pH에 의해 영향을 받는 것으로 나타났다. 배양 온도에 있어 28℃ 배양에 비해 37℃로 배양시 phytase 생산 속도가 빨랐으며, Ca 농도가 10 mM 이상에서 phytase 생산이 저하되었고, P 농도 증가에 따라 phytase 생산은 감소한 반면, acid phosphatase는 생산은 약간 증가하거나 영향을 받지 않아 결국 P. waksmanii AM301이 생산하는 phytase는 P-repressible phosphatase이며, acid phosphatase는 P-inducible 또는 constitutive phosphatase인 것으로 나타났다. 또한 phytase의 대량생산을 위한 기본 배지로 대두박 및 면실박이 적합하고, 쉽게 이용가능한 질소원의 첨가에 따라 phytase 생산이 크게 향상되는 결과를 얻었으며 최종적으로 P. waksmanii AM301에 의한 phytase의 최적 대량 생산 배지 조성은 대두박이나 면실박 2%(w/v)와 (NH_(4))_(2)SO_(4) 0.5%(w/v)로 나타났다. In vitro 에서 대두박, 밀기울 및 호밀을 기질로하에 P. waksmanii AM301 phytase에 의한 phytate-P 분해도를 측정한 결과 모든 기질에 대해 첨가 효과가 뚜렷 하였으며, 대두박>밀기울>호밀 순으로 분해되었는데 이러한 분해도의 차이는 수용성 phytate 함량 차이로 추정된다. In vitro 에서 산란계 사료에 P. waksmanii AM301 phytase, Bacillus sp. protease 및 cellulase의 첨가에 따른 phytate-P 분해에 미치는 영향을 파악한 결과 각각의 효소 처리에 의해서는 유의적으로(P<0.005) phytate-P 분해가 증가된 반면, 혼합 처리에 의해서는 오히려 감소하는 경향을 나타냈으며, 산란계 사료에 Ca(1.2%) 첨가에 따라 phytase 첨가여부와 관계없이 상등액으로의 P 방출량이 감소하였다. 실험 3. 어린가축의 분변으로부터 phytate 분해력을 나타네는 유산균들을 분리한 후 phytate 함유 조건에서의 균주성장 및 phytate 분해력이 가장 우수한 균주를 선발하여 동정한 결과 L. paracasei subsp paracasei로 밝혀졌으며, 대두박, 밀기울 및 쌀겨에 접종하여 순수 배양한 결과 대두박 및 밀기울에 있어서는 각각 27.07 및 12.18%의 phytate 분해율을 나타낸 반면 쌀겨에 있어서는 phytate 분해가 나타나지 않았다. 또한 L. paracasei subsp paracasei는 내산성 및 내담즙성이 우수한 유산균 으로 처리 전, 산 처리 및 담즙 처리시 각각 9.70, 9.66 및 8.80 log CFU/ml로 나타났으며, 비육돈 사료에 제올라이트를 부형제로 하여 0.4% 수준으로 첨가시 사료효율에 있어 대조구가 3.71인데 반해 3.21로 개선 효과가 나타났다. 실험 4. 자연계로부터 phytase 및 복합 효소(protease, collulase, xylanase 및 amylase) 활성윽 나타내는 균주를 얻어 동정판 결과 B. subtilis로 밝혀졌으며 순수배양한 결과 대두박 및 쌀겨에 있어서는 phytate 분해율이 낮았으나, 밀기울의 경우 80.63%의 phytate 분해율이 나타났다. 또한 대두박을 위주로 한 균주의 대량 생산 조건을 검토한 결과 대두박 1%(w/v)와 당밀 2%(w/v)가 가장 적합하였다.

Four experiments were conducted for the purposes; 1) isolation of P. woksmonii AM301, being good ability of phytase production, and purification and characterization of phytase originated from P. waksmanii AM301, 2) determination of factors affecting phytase production from P. waksmanii AM301 and conditions for mass enzyme production, 3) isolation of L. paracasei subsp paracasei, having ability to degrade phytate and investigation of possibility used as a dirert-fed-microbials or a fermented feed source, 4) isolation and determination of conditions for mass production of B. subtilis SY4-3, being capable of secreting phytase, frotease, cellulase, xylanase and amylase. The results were as follows; Experiment 1. Extracellular phytase from culture filtrate of P. waksmanii AM301 was purified by precipitation with ammonium sulfate followed by CM and DEAE-Sepharose CL-6B column chromatographies. The molecular weight of the purified enzyme was estimated be 47 kDa on gel filtration and 24 kDa on SDS-polyacrylamide gel electrophoresis, suggesting that the native enzyme is a dimeric protein. The outimum pH and temperature of the purified phytase were pH 4.5 and 55℃, respectively. The enzyme had the isoelectric point of 5.0, the Km value of 0.9 mM, and the Vmax value of 37.04 ㎚μmol Pi/mg/min for sodium phytate. Its activity was moderately inhibited by metal lons such a.s Fe^(++), Zn^(++), and Cu^(++). Experiment 2. Factors affecting phytase production from P. waksmanii AM301 were identified as temperature, P and Ca concentrations, and initial pH in medium. Phytase production was observed earlier at 37℃ than at 28℃ and inhibited by addition of Ca concentrations at more than 10 mM. With increasing concentrations of P, phytase production decreased while acid phosphatase production increased in small extent or did not change. Thus, two types of phosphatases produced by P. waksmanii AM301 were identified as P-repressible phytase and P-inducible or constitutive acid phosphatase. Basal media for mass prpduction of the enzyme were determined as soybean meal and cottonseed meal and phytase production increased distinctively with addition of easily available N source. Optimal levels of basal media and N source for mass production were 2% (w/v) for soybean meal and cottonseed meal, and 0.5% (w/v) for (NH_(4))_(2)SO_(4), respectively. Dephosphorylation effects of phytase on various substrates were noted distinctively in vitro and extent of dephosphorylation ranked soybean meal>what bran>rye, probably due to differences in soluble phytate concentrations. Experiment 3. Lactobacillus sp. isolated from young animal feces and selected a bacterium having high phytate degradation ability, being identified as L. paracasei subsp. paracasei. When inoculated into soybean meal, wheat bran and rice bran, estimated phytate-P degradability by the bacterium was 27.07 for soybean meal and 12.18% for wheat bran.
