Sulforaphane(SFN), a glucosinolate-derived isothiocyanate found in cruciferous vegetables, is known as a drug for treatments of various cancers. SFN was known to induce phase Ⅱ detoxification enzymes, and inhibited chemically induced mammary tumors in rodents. Recently, SFN is known to induce cell cycle arrest and apoptosis in human cancer cells, however its molecular mechanisms are poorly understood. In the present study, it was investigated that anti-proliferative effects of SFN on human hepatocellular carcinoma HepG2 and human cervicse adenocarcinoma HeLa cells were using hemocytometer counts, MTT assay, DAPI staining, fluorescence microscopy and flow cytometric analysis. Treatment of SFN to both cell lines resulted in the growth inhibition, morphological change and induction of apoptotic cell death in a dose-dependent manner. This anti-proliferative effect of SFN was accompanied by a marked inhibition of cyclin A, cyclin B1, cyclin D1 and Cdc2 expressions and concomitant induction of cyclin-dependent kinase (Cdk) inhibitor p21WAF1/CIP1, which appears to be transcriptionally upregulated in p53 dependent manner. In addition, SFN treatment resulted in induction of apoptosis as determined by fluorescence microscopy and flow cytometry. These effects were found to correlate with an activations of caspase-3, a shift in Bax/Bcl-XL, and selective down-regulation of IAP family members in both cell lines. SFN treatment also induced th proteolytic degradation of poly(ADP-ribose)polymerase and inhibition of β-catenin protein expression indose-dependently. Taken together, these findings indicate that SFN may be a potential chemoprevetive/chemotherapeutic agent for the treatment of human cancer cells.
