The present study was derived from the need of pancreatin containing protease, amylase, and lipase with its various compositions in pharmaceutical field. For the solution of this mission, the feasibility of controlling the selective precipitation of pancreatic proteins was investigated using designed organic anti-solvents in a drowning-out process. First, the characteristics of pancreatic enzymes in aqueous solution were checked in terms of their solubility and stability. Both in literature and experimental approaches, they were dramatically influenced by hydrogen concentration. Specially, the degradation of pancreatic enzymes could be restored by controlling pH of the solution to 5.0 for reducing pH-dependent intra-proteolysis. And semi-precipitable pancreatic lipase in aqueous circumstance was successfully transferred to soluble fraction using fatty acid ester non-ionic surfactants like POSL. The precipitation of pancreatin was then carried out with various anti-solvents composed of alcoholic and non-alcoholic species. The precipitation was promoted along with an anti-solvent of a lower polarity, which was also functionally related with solubility parameter of the solution. The amylase, lipase, and protease proteins contained in the pancreatin displayed different precipitation behaviors depending on the individual physiochemical properties of each protein. Thus, it was possible to selectively separate the proteins and control their composition in the precipitated pancreatin.
