The objectives of this study was 1) to divide isolated strains of E. coli from beef, pork, and poultry products into pathogenic groups, 2) to improve isolation method of enterohaemorrhagic E. coli (EHEC), and 3) to propose guidelines of microbiological hazard management and basic experimental data for the revision of pathogenic E. coli detection method. The contamination level of E. coli was investigated from beef, pork and poultry products (n=5,000) during the period from March 2004 to October 2006. In the experiment of E. coli classification by polymerase chain reaction, 5 groups were classified on the basis of their virulence traits as follows; the most well-studied members of the diarrheagenic E. coli group include enterotoxigenic E. coli (ETEC), enteropathogenic E. coli (EPEC), enteroinvasive E. coli (EIEC), enteroaggregative E. coli (EAEC), and shiga toxin producing E. coli (STEC), also called verocytotoxin producing or enterohemorhagic E. coli (VTEC). E. coli was isolated from 273 of 5,000 beef, pork, and poultry products and pathogenic strains of E. coli were identified from 39 (14.3%) of 273 strains. A variety of diarrheagenic E. coli was found; 14 strains belong to the virulence type of EHEC and those were mostly isolated from 7 of 14 strains and major serogroups of O111 was found in fresh beef, but E. coli groups such as EAEC and EIEC were not found. Also, this study proposed the optimal enrichment condition and selective medium for detecting non-E. coli O157 such as O26 and O111 in beef, pork, and poultry products. Immunomagnetic separation improved the detection rate of E. coli O26 and O111. In the comparison of enrichment media, mTSB+novobiocin and Buffered peptone water + vancomycin were effective for O26 and O111, respectively, isolating from in beef, pork and poultry products. Using gas modifications of carbon sources and antibiotic agents, the selective medium for O26 developed for the differentiation from O157 and O111. Characterization of the carbohydrate-fermenting ability of the 4 STEC isolates (1 O157, 1 O26, and 2 O111) revealed that either rhamnose or dulcitol could serve as a discriminative marker for the selective isolation of STEC O26.
